Regulation of microM vs microS mRNA expression in an inducible B cell line.

During the course of B lymphocyte differentiation into immunoglobulin secreting cells the abundance of mRNA for the heavy chain of secreted IgM (microS) increases dramatically. In order to understand the regulatory events responsible for the selective increase in micS mRNA we have looked for transcriptional alterations of VDJC mu gene segments as well as changes in the relative stability of microM and microS mRNA in BCL1 lymphoma cells which can be stimulated to increase the expression of microS mRNA. These experiments showed that although the transcriptional level of the mu gene locus is not preferentially augmented after stimulation, an alteration in the sites of polymerase termination is a significant factor contributing to the higher microS to microM ratio. This switch is dependent on new RNA synthesis. In addition, although the half-life of microS mRNA is not selectively increased, stimulation of the cells does result in a specific enhancement of the half-lives of both species of mu mRNA, which accounts for the higher steady state levels of total mu message.

Effects of chronic dietary aluminum on local cerebral glucose utilization in rats.

Beginning at 4 weeks of age normal, male, Sprague-Dawley rats were reared on Purina Laboratory Chow and drinking water containing 100 microM AlCl3. After 2 years, local rates of cerebral glucose utilization were determined with the autoradiographic [14C] deoxyglucose method in the brain as a whole and in 25 brain regions in 6 treated rats and 4 age-matched controls. The results indicate that any effects of chronic aluminum in the diet on rates of cerebral glucose utilization are small. In the brain as a whole, the mean rate of glucose utilization in the aluminum-treated rats was 6% lower than that of the controls (p = 0.09). In 21 of the 25 brain regions examined mean rates of glucose utilization were generally lower in the aluminum-treated rats but in none of the region were the effects statistically significant.

Adenovirus-mediated combination suicide and cytokine gene therapy for bladder cancer.

BACKGROUND: The present study tests the hypothesis that adenovirus-mediated transfer of murine IL-2 (ADV/RSV-mIL-2) alone or in combination with HSV-tk + GCV will improve antitumorigenic response in the murine MBT-2 model. MATERIALS AND METHODS: mIL-2 production and toxicity were determined in vitro using an ELISA and a cell proliferation assay. Tumor-bearing animals were randomly assigned into four treatment groups and directly injected with combinations of ADV/RSV-tk and ADV/RSV-mIL-2. In a separate experiment, the above-mentioned groups were followed by two subsequent treatments with ADV/RSV-mIL-2. RESULTS: Transduced MBT-2 cells were able to express mIL-2 in a time and dose dependent fashion. We could not demonstrate any improvement in antitumorigenic response with mIL-2 gene therapy alone or in combination with HSV-tk-suicide gene therapy over HSV-tk suicide gene therapy alone. CONCLUSIONS: Although ADV/RSV-mIL-2 transduced MBT-2 cells were able to produce large amounts of mIL-2 in vitro, we could not demonstrate significant tumor growth inhibition by adding mIL-2 gene therapy to suicide gene therapy. The growth inhibitory effects of sequential suicide and cytokine gene therapy were transient and not superior to single dose suicide and cytokine gene therapy.

[The surgery of peripheral nerves. New perspectives in treatment by the microsurgical techniques (author's transl)].

The author describes the different techniques of reconstructive surgery of peripheral nerves and the failures that marked these trials until the advent of microsurgical techniques. After stressing the importance of the biology and physiopathology of a traumatized nerve and its stem cells, the author detailes the regeneration stages of the nerve cell when ideal conditions of axonal alignment are established by microtechniques. Considering the different notions of biology and pathology in relation to the time delay between the traumatism and referral to the surgeon, the author insists on the absolute necessity of operating during the period of full regeneration of the nerve cell, i.e. within the first 6 months.

[Indications and operative conditions of aneurysms in old patients (author's transl)].

The age factor is analysed for the operative indication in old patients. For the authors, operative indications are depended of mental and general status, clinical grade (I or II), localization and anatomical conformation of the aneurysm. The operation is performed only if the patient is asymptomatic or three weeks after the onset of the subarachnoid hemorrage. The peroperative hypotension is prohibited.

Non traumatic cerebellar hematomas.

Five cases of spontaneous cerebellar hematomas are reported. In four cases the diagnosis was made by computerized tomography. The clinical diagnosis being difficult, surgical treatment was often applied too late and the diagnosis sometimes never made. Systematic use of computerized tomography should allow earlier treatment of this type of lesion, the clinical evolution determining the prognosis. (Acta neurol. bels., 1980, 80, 73-79).
