Catalytic oxidation of benzene using DBD corona discharges.

Plasma oxidation of benzene (C(6)H(6)) in oxygen and nitrogen was investigated using a dielectric barrier discharge (DBD) reactor with or without MnO2 or TiO2 at atmospheric pressure and without external heating except plasma heating. An alternative current power supply was used to generate corona discharges for the plasma oxidation. The energy density was controlled under 200 J/L to keep an increase in gas temperature less than 167 K. C(6)H(6) was oxidized to carbon monoxide (CO) and dioxide (CO(2)). Typically, the energy efficiency at an energy density of 92J/L was about 0.052, 0.039, and 0.024 mol/kWh with MnO2, TiO2, and without MnO2 and TiO2, respectively. Benzene oxidation mechanism was mentioned. A comparison on energy efficiency as a function of initial concentration of hydrocarbons, inorganic sulphur compounds, and chloro (fluoro and bromo) carbons was given.

Temperature-programmed desorption study of the selective oxidation of alcohols on silica-supported vanadium oxide.

The partial oxidation of methanol and ethanol on silica-supported vanadium oxide catalysts was studied using temperature-programmed desorption (TPD), Raman spectroscopy, and diffuse reflectance infrared spectroscopy (DRIFTS). Methanol TPD results for V2O5/SiO2 samples as a function of vanadia loading in conjunction with X-ray diffraction data and Raman spectra indicated that dispersed vanadia on silica agglomerates into vanadia crystallites during a CH3OH TPD experiment. For ethanol-dosed samples, agglomeration of the dispersed vanadia was less severe, and it was possible to measure the activation energy for the dehydrogenation of adsorbed ethoxides to produce CH3CHO. Assuming a preexponential factor of 10(13) s(-1), the activation energy for this reaction was estimated to be 132 kJ/mol. The results of this study further demonstrate that there is a relatively weak interaction between vanadia and silica and suggest that adsorbed methoxide species help facilitate agglomeration of dispersed vanadia.

Needle sharing and sex-related risk behaviours among drug users in Shenzhen, a city in Guangdong, southern China.

Intravenous drug users (IDUs) are at high risk of contracting HIV, but there is a geographic bias in such research efforts in China. The aim was to study the pattern of needle sharing and sex-related risk behaviours among drug users in southern China. About 260 drug users in a detoxification centre completed an anonymous questionnaire (response rate=87.3%). The results show that needle sharing among IDUs was prevalent (60.6% and 45.3% for male and female IDUs, respectively); the sharers were often friends, spouses, and acquaintances. Few IDUs sterilized the used needles properly. Commercial sex behaviours (males: 44%; females: 60%) as well as multiple partnership (males: 60%; females: 53%) were also prevalent among all male and female drug users studied. Inconsistent condom use with commercial sex partners, spouses, and regular sex partners was also very common. Yet, few of all drug users studied perceived the susceptibility of contracting HIV (11%) or sexually transmitted diseases (24%) in the future. A better HIV-related knowledge level was associated with lower levels of risk behaviours. It is concluded that harm reduction programmes are urgently warranted in China.

Fluorescence-based functional assay for Wnt/beta-catenin signaling activity.

Aberrant activation of beta-catenin signaling has been implicated in the development of human cancers. As a Wnt signal transducer, beta-catenin forms a complex with the lymphocyte enhancer-binding factor/T cell factor transcription factor and activates downstream targets that promote cell proliferation. Here we developed a Wnt-dependent beta-catenin-mediated heterologous transactivation system, which consisted of a chimeric transcription factor constructed by fusing the GAL4 DNA-binding domain with the full-length beta-catenin, and a GAL4-responsive reporter expressing GFP. The chimeric transcription factor was highly unstable and exerted no detectable transactivating effect on the GAL4-responsive reporter. However, lithium and Wnt1 significantly stabilized this chimeric transactivator, indicating that this transactivation system is regulated by beta-catenin in a Wnt-responsive fashion. Thus, this transactivation system could be used as a functional reporter to identify potential upstream factors that deregulate beta-catenin signaling during tumorigenesis, as well as to screen for potential anti-cancer agents that specifically inhibit beta-catenin signaling in human tumors.

Tetracycline-regulated gene expression mediated by a novel chimeric repressor that recruits histone deacetylases in mammalian cells.

Regulated gene expression will provide important platforms from which gene functions can be investigated and safer means of gene therapy may be developed. Histone deacetylases have recently been shown to play an important role in regulating gene expression. Here we investigated whether a more tightly controlled expression could be achieved by using a novel chimeric repressor that recruits histone deacetylases to a tetracycline-responsive promoter. This chimeric repressor was engineered by fusing the tetracycline repressor (TetR) with an mSin3-interacting domain of human Mad1 and was shown to bind the tetO(2) element with high affinity, and its binding was efficiently abrogated by doxycycline. The chimeric repressor was shown to directly interact with mSin3 of the histone deacetylase complex. This inducible system was further simplified by using a single vector that contained both a chimeric repressor expression cassette and a tetracycline-responsive promoter. When transiently introduced into mammalian cells, the chimeric repressor system exhibited a significantly lower basal level of luciferase activity (up to 25-fold) than that of the TetR control. When stably transfected into HEK 293 cells, the chimeric repressor system was shown to exert a tight control of green fluorescent protein expression in a doxycycline dose- and time-dependent fashion. Therefore, this novel chimeric repressor provides an effective means for more tightly regulated gene expression, and the simplified inducible system may be used for a broad range of basic and clinical studies.

Transgenic rice plants expressing the ferredoxin-like protein (AP1) from sweet pepper show enhanced resistance to Xanthomonas oryzae pv. oryzae.

We used particle bombardment to cotransform mature seed-derived rice callus (Oryza sativa L., ssp. japonica, cv. Eyi 105) with plasmids containing the linked marker genes gusA and hpt, and the ap1 gene encoding an amphipathic protein previously shown to delay the hypersensitive response induced in non-host plants by the pathogen Pseudomonas syringae pv. syringae (Pss). Thirty-two independent lines of transgenic rice plants were regenerated, and 27 of these lines carried all three transgenes as shown by molecular analysis. A bacterial blight inoculation test was carried out on ten lines. In each case, plants carrying the ap1 gene showed enhanced resistance to Xanthomonas oryzae pv. oryzae (Xoo) race 6 at various levels. This suggests the ap1 gene could be a useful candidate for genetic engineering strategies in rice to provide bacterial blight resistance.

Adenoviral vector-mediated gene transfer for human gene therapy.

Human gene therapy promises to change the practice of medicine by treating the causes of disease rather than the symptoms. Since the first clinical trial made its debut ten years ago, there are over 400 approved protocols in the United States alone, most of which have failed to show convincing data of clinical efficacy. This setback is largely due to the lack of efficient and adequate gene transfer vehicles. With the recent progress in elucidating the molecular mechanisms of human diseases and the imminent arrival of the post genomic era, there are increasing numbers of therapeutic genes or targets that are available for gene therapy. Therefore, the urgency and need for efficacious gene therapies are greater than ever. Clearly, the current fundamental obstacle is to develop delivery vectors that exhibit high efficacy and specificity of gene transfer. Recombinant adenoviruses have provided a versatile system for gene expression studies and therapeutic applications. Of late, there has been a remarkable increase in adenoviral vector-based clinical trials. Recent endeavors in the development of recombinant adenoviral vectors have focused on modification of virus tropism, accommodation of larger genes, increase in stability and control of transgene expression, and down-modulation of host immune responses. These modifications and continued improvements in adenoviral vectors will provide a great opportunity for human gene therapy to live up to its enormous potential in the second decade.

[Analysis of HIV-1 seropositive individual infected with different subtypes].

OBJECTIVE: To investigate whether there existed any individuals infected with different subtypes of human immunodeficiency virus type 1 (HIV-1) and to analyze their source of infection. METHODS: Membrane protein gene (env) of HIV-1 in uncultured single peripheral blood monocytes (PBMCs) from HIV-1 seropositive individuals found in Shenzhen was amplified by nested polymerase chain reaction (PCR). The C2-V3 region and its adjacent region (360 bp) of them were sequenced and analyzed, and their divergence rate and systematic phylogenetic tree (cladogram) were calculated. RESULTS: There existed sequences of B and C or B and E strains of HIV-1 subtype in three infected individuals in Shenzhen. Gene divergence rates between different subtypes and corresponding international subtype reference strains Bcon, Ccon and Econ were 5.07%-7.26%, 3.23% and 5.38%-5.99%, respectively, and those between different subtypes strains in the same individual were 23.52%, 20.69% and 28.44%, respectively. GPGR sequence of typical Europe and America subtype B was found in strain sz46-1 from prostitute with intravenous drug abuse, based on their tetra-peptide sequence characteristics in the crown of gp 120 V3 loop, strain sz8-2 was GPGH and other strains were GPGQ sequence characterized by Thai B (B') and E subtypes. Furthermore, phylogenetic tree analysis showed that strains sz1-2, sz8-1 and sz46-1 were clustered with international reference sequence Bcon, strain sz1-1 clustered with Econ, and strains sz1-1 and sz8-2 clustered with Ccon. CONCLUSION: These findings suggested that an individual could be simultaneously infected with different HIV-1 subtypes, which could probably be caused by his or her own multiple high-risk behavior.

Reduced-glutathione concentrations in Boleophthalmus pectinirostris tissues exposed to benzo(a)pyrene.

The concentrations of reduced-glutathione (GSH) in liver and ovary of Boleophthalmus pectinirostris are quantified. The concentrations of GSH in the ovary are much higher than that of GSH in the liver(nearly 3 times of the liver). The study also investigates the changes of GSH contents in the two organs while the fishes were exposed to benzo(a)pyrene(BaP) at concentrations of 0, 0.05, 0.2 and 0.5 mg/L respectively for up to a week. The concentrations of GSH in the liver of BaP-exposed fish increased significantly with dose, whereas the concentrations of GSH in the ovary decreased significantly compared to controls. The results suggested that both the liver and the ovary are the primary organ in BaP metabolism, and that the changes of GSH levels may represent an adaptive response or toxic effect to BaP exposure.

[Effect of benzo(a)pyrene on antioxidant enzyme activities in Boleophthalmus pectinirostris liver].

The effect of different BaP concentrations on the activities of superoxide dismutase(SOD), glutathione peroxidase (GPx) and catalase(CAT) in Boleophthalmus pectinirostris liver was studied under experimental condition. The results showed that the activities of antioxidant enzymes did not change too much with BaP exposure in lower concentration, whereas the SOD and GPx activities were increased significantly with BaP exposure in higher concentration. No statistical changes were observed in the effect of BaP exposure in different concentrations on CAT activities throughout the experiment.

Regulation of osteopontin expression in a rat model of urolithiasis.

OBJECTIVE: To investigate the relationship between the expression and regulation of osteopontin (OPN) and urolithiasis. METHODS: Normal and stone model rats were treated with 1,25-dihydroxyvitamin D3(D3), vitamin K, testosterone or estradiol for 7 days, and the expression of osteopontin and its mRNA were detected with immunohistochemistry and Northern blot, respectively. Crystals deposited in rat kidneys were observed with a polarization microscope. The concentrations of crystal components in rat urine were determined. RESULTS: The results showed that vitamin K, testosterone and estradiol up-regulated the expression of OPN mRNA and its protein, thus decreasing the precipitation of calcium oxalate in rat kidneys. D3 increased the concentration of calcium in urine, and accelerated the sedimentation of calcium oxalate in rat kidneys. CONCLUSIONS: These findings indicate that OPN may be an important macromolecule in the normal endogenous inhibition of the formation of urolithiasis. Vitamin K, testosterone and estradiol inhibit the formation of stones via up-regulating the expression of OPN in kidneys, while D3 over dose may accelerate the process.

[Cytomegalovirus infection associated with plasma endothelin and P-selectin in patients with essential hypertension].

BACKGROUND: To investigate the relationship between human cytomegalovirus (HCMV) infection and plasma endothelin and P-selectin in patients with essential hypertension. METHODS: Blood samples of 98 patients with essential hypertension were collected for HCMV IgM and IgG antibodies, plasma endothelin and plasma P-selectin by chemiluminescence technique, radio-immunoassay and ELISA. RESULTS: The rates of positive HCMV IgM antibodies and positive rates for both HCMV IgM and IgG antibodies were 56.12% and 54.08%, respectively, which were higher than that of the control group (P=0.05). The level of plasma endothelin and P-selectin in patients with positive HCMV IgM antibodies was 95.53 +/- 5.06 ng/ml and 42.50 +/- 3.28 ng/ml, respectively, which were higher than that in patients with negative HCMV IgM antibodies (71.60 +/- 4.34 ng/ml and 27.41 +/- 5.28 ng/ml) and that of the control group (48.77 +/- 5.96 ng/ml and 19.80 +/- 4.62 ng/ml). CONCLUSIONS: These results suggest that HCMV active infection may aggravate vascular endothelium lesi on in patients with essential hypertension and contribute to the increase of endothelin and P-selectin.

[Clinical observation on sensorineural hearing loss secondary to secretory otitis media].

OBJECTIVE: To study whether secretory otitis media causes sensorineural deafness or not, especially in stubborn cases. METHODS: The 0.5 kHz, 1 kHz, 2 kHz, 4 kHz BC stimuli were administered to 164 patients with secretory otitis media, and their BC loss calculated. As a control, BC threshold was also recorded in the contralateral ear of 66 unilaterally deaf patients. RESULTS: BC threshold was raised in 94 out of 164 patients (57.3%). There was no statistically significant difference between bilaterally and unilaterally deaf groups. All patients had similar BC hearing loss in the same frequency, but had different BC hearing loss in different frequencies and the most serious loss was in 4 kHz. CONCLUSION: The rate of sensorineural deafness caused by secretory otitis media is higher than expected. Frequency and intensity of hearing loss between unilaterally and bilaterally deaf patients with secretory otitis media were no different. All ears had different hearing loss in different frequencies, but the most serious hearing loss was in high frequency and it had the tendency to spread to the speech frequency.

Salivary and gastric epidermal growth factor in patients with Zollinger-Ellison syndrome: its protective potential.

OBJECTIVE: Evidence is accumulating that epidermal growth factor (EGF) is a major molecule contributing to the maintenance of the integrity of the upper alimentary tract mucosa before and after injury by acid and pepsin. Patients with Zollinger-Ellison Syndrome (ZES) typically have hypersecretion of acid and pepsin; however, the concentration and rate of secretion of salivary and gastric EGF that could counteract these potentially aggressive factors are unknown. Accordingly, this study was conducted to determine whether EGF affords mucosal protection in ZES patients. METHODS: The concentration and output of salivary (sEGF) and gastric epidermal growth factor (gEGF) were measured in eight patients with ZES and the results compared to those in 17 patients with nonulcer dyspepsia (NUD), serving as a control group. All patients had normal esophageal and gastric mucosa as determined by endoscopy. Total saliva was collected during 1-h parafilm- and 1-h pentagastrin/parafilm-stimulated conditions, as well as basal and pentagastrin-stimulated gastric juice. The concentration and output of EGF were determined by radioimmunoassay. RESULTS: The concentration of EGF in saliva collected from ZES patients after parafilm chewing was significantly higher compared to that in NUD patients (4.61 +/- 0.59 vs 2.75 +/- 0.50 ng/ml, p < 0.05). The concentration of EGF in saliva collected after pentagastrin stimulation in ZES patients was also significantly higher than in NUD patients (4.37 +/- 0.73 vs 2.22 +/- 0.37 ng/ml, p < 0.05). Salivary EGF output during parafilm chewing in ZES and NUD were similar (68 +/- 6.4 vs 109 +/- 25.2 ng/h). Salivary EGF output after administration of pentagastrin in ZES and NUD was also similar (66 +/- 6.1 vs 132 +/- 45.4 ng/h). Basal EGF output in the gastric juice of patients with ZES was 3-fold higher than in patients with NUD (801 +/- 73 vs 271 +/- 32 ng/h, p < 0.01). Pentagastrin-stimulated EGF output was similar in both groups (705 +/- 92 vs 675 +/- 168 ng/h). CONCLUSIONS: Patients with ZES have a significantly higher EGF concentration in saliva and EGF output in basal gastric juice. This elevated content of salivary and gastric EGF in ZES patients may play a protective role in preventing the development of reflux esophagitis and gastric ulcer under the impact of gastric acid and pepsin hypersecretion.

[Molecular epidemiology analysis of HIV-1 subtype E strains found in Shenzhen, China].

OBJECTIVE: To investigate the prevalent status of HIV-1 subtype E epidemic among different populations in Shenzhen, China. METHODS: HIV-1 proviral DNA from peripheral blood mononuclear cells (PBMCs) of 3 HIV-infected individuals found in Shenzhen was amplified by nested-PCR. The C2-V3 and its flanking region (350-450 nucleotides) of HIV- 1 env gene were sequenced directly from the PCR products and analysed. RESULTS: These 3 cases were confirmed to be infected with HIV-1 subtype E strain (sz-E). The innergroups distance of sz-E was 2.6%. In comparison with the consensus sequence of international A-E subgroups and some of subtype B epidemic in China, the genetic distances among sz-E and each of A-D subgroups and some of subtype B were more than 24 %. Furthermore, the phylogenetic tree analysis showed that sz-E clustered closely to Econ. CONCLUSIONS: These findings suggested that the epidemic time for HIV-1 subtype E, which was mainly prevalent among IDUs population and some professional blood donors, had been about 2-3 years in Shenzhen.
