Subcellular localization of insulin-like growth factor I (IGF-I) in Purkinje cells of the adult rat: an immunocytochemical study.

Insulin-like growth factor I (IFG-I) was localized by light and electron microscopic immunocytochemistry in the adult rat cerebellum. Neurons showing the same cytological characteristics and pattern of synaptic input as Purkinje cells were IGF-I immunoreactive. In these neurons, intense electron-dense reaction product was consistently located in the rough endoplasmic reticulum and, occasionally, in multivesicular bodies and associated with the nuclear envelope. Thus, the results of this study suggest synthesis IGF-I in Purkinje cells and furthermore provide anatomical support for IGF-I participation in paracrine or autocrine regulatory systems in the brain.

Maturation of the coherence of EEG activity in normal and learning-disabled children.

The age effect on coherence has been studied in control (98) and learning-disabled (LD, 54) school-aged children (from 6.0 to 16.8 years old). The EEG recordings were made at rest in 15 leads, and 105 pairwise combinations for coherence were calculated (each lead was compared with all the rest) for delta, theta, alpha, beta and total frequency bands. A significant increase of coherence with age was found in both groups, with a different pattern of maturation. In the control group, a significant increase with age was found in the coherences between posterior regions and vertex (Cz). A significant decrease with age in the coherence between frontal areas was observed, especially in the theta band. The LD group showed a different pattern: no significant relation with age was found in the coherence between any lead and vertex. A high effect of age on coherence between temporal regions was observed with a predominance of the left side in comparison with the contralateral and the ipsilateral. No decrease in frontal coherence was found: in the same region where the control group showed negative values with age, the LD groups had no age effect. The results obtained are discussed as differences in brain organization, in myelogenesis and synaptogenesis and an explanation of the etiology of LD is proposed.

Assessment and management of opioid withdrawal symptoms in buprenorphine-dependent subjects.

The spontaneous physical dependence of buprenorphine was assessed in opioid addicts who switched from heroin to sublingual or intravenous buprenorphine. Twenty-two patients were randomly assigned to double-blind administration of methadone (n = 11) or placebo (n = 11) for 13 days after abrupt withdrawal of buprenorphine. Methadone was administered according to four pre-established dosing schedules depending on the previous amount of daily consumed buprenorphine. No methadone-treated patient required modification of the therapeutic regimen, whereas eight of eleven placebo-treated patients needed treatment with methadone. Buprenorphine withdrawal syndrome was of opioid type, began somewhat more slowly, and showed a peak until day 5. The occurrence, time-course and characteristics of buprenorphine withdrawal syndrome make it necessary to reconsider the abuse potential of this analgesic.

Immunocytochemical localization of insulin-like growth factor I in the hypothalamo-hypophyseal system of the adult rat.

Insulin-like growth factor I (IGF-I) is shown to be involved in the regulation of pituitary hormones. High IGF-I concentrations were detected in hypothalamus and pituitary during adulthood. This study was undertaken to analyze the cellular distribution of IGF-I in the hypothalamo-hypophyseal system of the adult rat using immunocytochemical procedures. IGF-I was found to be widely distributed throughout the hypothalamus; it was present in the magnocellular neurons of the supraoptic, paraventricular, and accessory nuclei. Moreover, nerve fibres and puncta containing immunoreactive IGF-I were localized in the median eminence and the posterior lobe of the pituitary. These results support possible IGF-I neuromodulatory or neurohormonal action in the hypothalamus on pituitary hormone regulation.

Effect of sex, psychosocial disadvantages and biological risk factors on EEG maturation.

Broad-band frequency analysis was performed on the EEGs of school-age children in order to study the effects of sex, socioeconomic status (SES) and biological risk factors on EEG maturation. Absolute power and relative power in delta, theta, alpha and beta frequency bands were computed in monopolar recordings from F3, F4, C3, C4, P3, P4, O1, O2, F7, F8, T3, T4, T5 and T6. Total absolute power and power in the delta and theta bands decreased with age, following a quadratic polynomial expression. Relative power followed a linear regression with age. Delta and theta decreased while alpha and beta increased. Sex differences in relative power were observed, possibly related to a pubertal spurt: slopes of regression equations were steeper in girls than in boys. Children from low SES (very low income and/or illiterate mother) had higher values of absolute power and a higher percentage of delta and lower of alpha than children with good SES, suggesting a maturational lag. A group of children with personal antecedents of risk factors associated with brain damage were compared with children without antecedents. The former group had higher values of absolute power. Depending on the severity of the risk, children with more severe antecedents had greater differences from children with no antecedents than children in whom antecedents were considered as slight. Risk antecedents had no effect on relative power.

Comparative study of cell-immunoenzymatic methods for the estimation of IgG and IgM anti-Brucella antibodies in the diagnosis of human brucellosis.

Two methods which employ whole cells are described and compared for the detection of human IgG and IgM anti-brucella antibodies. Dot ELISA and ELISA were shown to be suitable for a screening diagnosis of human brucellosis. Titres of antibodies obtained by dot ELISA showed 100% coincidence for IgG and 97% for IgM, compared with agglutination and complement fixation tests; when ELISA was used 11% positive sera were not detected. The comparison of these two methods with the conventional serological test kit indicated that both dot ELISA and ELISA were sensitive, reproducible and specific for the quantification of IgG and IgM antibrucella antibodies.

[Surface markers and humoral factors in spontaneous murine leukemia].

This paper presents a model of spontaneous BALB/c lymphoma (LB) useful for the study of the relationship between the tumor and the immune system. Characterization of this tumor was done by FACS and it was determined that the tumor cells are T lymphocytes which express both CD4 and CD8 membrane markers but not the viral protein gp 70. In addition, tumor cells express IL-2 receptors and class I, but not class II, H-2 antigens. Antibodies were not detected in sera or ascitic fluid of syngeneic mice bearing a tumor transplant while both sera and ascites contained a suppressor factor that inhibited in vitro the proliferation of tumor cells and blastogenesis of normal BALB/c splenocytes after Concanavalin A stimulation. The results indicate that this tumor may be used as an experimental model for adult T cell leukemia as well as for the isolation and characterization of cytokines which may be responsible for the immunological unresponsiveness found in certain cancer patients.

Autophosphorylation of yeast hexokinase PII.

Autophosphorylation of hexokinase PII was studied using an enzyme purified from Saccharomyces cerevisiae. Incubation of this enzyme preparation with [gamma-32P]ATP and Mn2+ or Mg2+ gave a phosphoprotein of molecular mass 58,000 which corresponded to hexokinase PII. D-Xylose stimulated autophosphorylation of hexokinase PII. Dilution of hexokinase PII over a 10-fold concentration range did not change the specific activity of hexokinase PII autophosphorylation suggesting that it may occur by an intramolecular mechanism.

Glucose-stimulated phosphorylation of yeast isocitrate lyase in vivo.

Incorporation of 32P into Saccharomyces cerevisiae isocitrate lyase was observed after addition of glucose to a culture incubated with [32P]orthophosphoric acid. A band of 32P-labelled protein was coincident with the enzyme band when immunoprecipitates were subjected to SDS-PAGE and autoradiography. No label was found in the band corresponding to the isocitrate lyase when immunoprecipitation was done with a control pre-immune serum or in the presence of excess pure unlabelled enzyme. The incorporation of phosphate was associated with a decrease in enzyme activity. Phosphorylated isocitrate lyase was not proteolytically degraded when cells were cultured in mineral medium. The loss of protein antigenicity only took place when the yeast was grown in a complex medium containing glucose.

Initiation factor 2 isolated from rat brain contains kinase activities responsible for its phosphorylation.

Initiation factor 2 from adult rat brain was isolated from salt-washed microsomes using a three-step purification process consisting of heparin-Sepharose, phosphocellulose and diethylaminoethyl-cellulose (DEAE cellulose) column chromatographies. The initiation factor 2(eIF-2) was phosphorylated in subunits alpha and beta by the endogenous protein kinase activity present in the pruified preparation. This protein kinase activity proved to be mostly a casein kinase, although the possible presence of a very specific alpha kinase activity cannot be dismissed.
