[Local immunotherapy with streptococcal preparation, OK-432 in superficial bladder tumors, and common antigens between OK-432 and the tumor].

In 38 patients with superficial bladder, local immunotherapy with streptococcal preparation OK-432 has been performed. We investigated whether OK-432 was an effective biological response modifier (BRM) against bladder tumors or not, and the relationship between the common antigens which OK-432 shared with the tumors, and antitumor effects of OK-432. In six out of 28 patients treated by intravesical instillation, and three out of 10 cases treated by intratumor injection, tumors were eliminated endoscopically. In the other patients, the tumors did not change. The PAP study using an anti-OK-432 antibody, showed a positive reaction in 66.7% of the instillation cases and in 40.0% in the injection cases. In 66.7% of the patients with the common antigens treated by the instillation and in 75.0% of patients with the antigens treated by the injection, tumors were eliminated. However, the PAP study showed a positive reaction in 9.1% of the no-change cases treated by the instillation and in 14.3% out of the no-change cases treated by the injection. We concluded that OK-432 was a favorable BRM for topical immunotherapy against bladder tumor and the presence of the common antigens between OK-432 and tumor may enhance the immune response of patients and promote tumor regression.

Scanning electron microscopic studies on the embryonic development of the surface structures of the paraventricular organ in the Brown Leghorn chicken.

The development of the surface specialization of the paraventricular organ (PVO) was studied in the domestic chicken from the 10th embryonic day to the day of hatching by scanning electron microscopy. On the 10th embryonic day, the ventricular surface of the PVO was found to be covered with many oval-shaped processes. On the 12th embryonic day, an additional kind of elongated processes appeared in the dorsal area of the ventricular surface of the organ. From the 16th to 18th embryonic day, such elongated processes were present on the entire ventricular surface of the PVO. At the same stage, the elongated processes in the dorsal portion of the PVO began to form small, meshed networks over the surface of the ependyma. Both the oval-shaped processes and the elongated processes are thought to be dendritic terminals of the PVO neurons.

Supraependymal cells and fibers in the third ventricle of the domestic chicken. A scanning electron microscopic study.

Supraependymal cells, fibers and what are presumed to be neuronal bulb-like projections were found in the third ventricle of the domestic chicken with a scanning electron microscope. At least two types of supraependymal cells were found: neuron-like cells and phagocyte-like cells. The former were predominantly seen in the area of the paraventricular organ and infundibular recess. The latter were abundant on the ventricular surface of the median eminence and subfornical organ. Bulb or club-like projections thought to be the dendritic terminals of CSF-contacting neurons were observed in the area of the paraventricular organ and infundibular recess. Similar structures were observed at the preoptic recess as well. The supraependymal neuronal components found in the domestic chicken differed from those of mammals in several respects: 1. the wall of the third ventricle was devoid of supraependymal fibrous plexus except for that of the paraventricular organ; 2. bulb-like projections were abundant in the area of the paraventricular organ; 3. supraependymal neuron-like cells were unipolar or bipolar in appearance. These data underline the dissimilarity of the CSF-contacting neuronal system of birds and mammals.

[Serum uptake of doxorubicin intravesically administered soon after transurethral resection of bladder carcinoma].

To obtain the maximum prophylactic effect of intravesical chemotherapy on the bladder tumor recurrence treatment should be started early. Since 1983, we have been initiating prophylactic instillation of doxorubicin hydrochloride (DXR) on the first post-transurethral resection (TUR) day. This study was conducted to define serum uptake of DXR and systemic toxicity in the early post-TUR period. Fifteen TURs were carried out on 14 patients with superficial bladder carcinoma. DXR (30 mg) in normal saline (30 ml) was intravesically administered 1, 3, 5, 7 and 14 days after TUR, and every 4 weeks thereafter. DXR solution was kept in the bladder for 2 hours. The serum DXR concentration was measured 30 minutes and 2 hours after the instillation through 1 to 5 days after TUR. Intolerable vesical irritability was seen in 4 of 60 instillations. No systemic side effects, however, were observed. Three of 24 samples contained a detectable level (10 ng/ml) of DXR on the post-TUR day 1, 6 of 22 samples on the post-TUR day 3, and 6 of 14 samples on the post-TUR day 5. Overall, 15 of 60 samples contained more than 10 ng/ml DXR. The highest serum DXR level was 47 ng/ml at the post-TUR day 1. Frequency of detection and average levels of serum DXR in 30-minute and 2-hour samples were not significantly different. Average concentrations in patients with multiple or diffuse tumor and solitary tumor were also not significantly different. These results indicate that intravesical instillation starting within 24 hours after TUR does not produce significant serum uptake of DXR, and systemic toxicity can thereby be avoided.

Reconstitution of partially purified opioid receptors with a GTP-binding protein.

Partially purified opioid receptors, obtained from rat brains using an affinity resin, AF-Amino Toyopearl with [D-Ala2, Leu5]enkephalin, were reconstituted with an inhibitory GTP-binding protein (Gi). In the reconstituted system, the displacement curve for the binding of a delta-agonist, [D-Ala2, D-Leu5]enkephalin, showed two states, high and low affinity binding ones, with different affinities for the agonist. The high affinity binding was eliminated by the addition of a guanine nucleotide analog to the system. These results directly showed that opioid receptors, at least the delta-type, could interact with Gi.

Specialized ependyma in the posterior mesencephalon of the chicken: the fine structure of the subtrochlear organ.

A formation of specialized ependymal cells in the posterior mesencephalon of the domestic fowl, designated as the subtrochlear organ, was examined with light-, scanning- and transmission electron microscopy. This organ possessing the form of the letter "V" is located in the ventricular wall of the posterior mesencephalon. Its apex marks the median sulcus, while the arms of the V are directed rostrolaterally. Ependymal cells lining the subtrochlear organ usually project an extremely elongated process into the subependymal region and are classified into three types according to their surface features: (1) cells with a bulb-shaped protrusion that projects into the ventricle, (2) single cilium-bearing cells, and (3) cells with a tuft of cilia. The first type of cell is restricted to the median portion of the subtrochlear organ; its bulb-shaped protrusion contains numerous ribosomes. The second type of cell predominates in the arm (rostrolateral) area; in its apical cytoplasm such ciliary structures as basal body are rarely seen. The third type of cell is usually assembled into several small islands on the arm area; it has many basal bodies and other ciliary structures in the apical cytoplasm.

Lectin and ultrastructural cytochemistry of glycoconjugates in the cecal epithelium of the chicken.

In the cecal epithelium of the chicken, glycoconjugates were studied by light microscopy with lectin-peroxidase and correlated procedures, and by electron microscopy with high iron diamine-thiocarbohydrazide-silver proteinate (HID-TCH-SP) and periodic acid (PA)-TCH-SP methods. Mucous granules of goblet cells and striated border of columnar cells at the basal cecum contained acidic and neutral glycoconjugates with sulfate ester, vicinal diol groupings and alpha-D-mannose, alpha-D-glucose, beta-D-galactose, N-acetyl-D-glucosamine, and sialic acid-galactose dimers. In addition, terminal galactose-(1-3)N-acetylgalactosamine disaccharides were detected. In the apical cecum, similar glycoconjugates involved in the mucous granules of goblet cells and the striated border of columnar cells were found to be smaller in amount. Terminal galactose-(1-3)N-acetylgalactosamine disaccharides together with glycogen particles were also visualized in columnar cells of the basal and apical cecum. The histophysiological significance of glycoconjugates involved in the chicken cecal epithelium was discussed.

The distribution of acetylcholinesterase(AChE)-positive nerves in chicken pancreas demonstrated by light and electron microscopy.

The distribution of acetylcholinesterase (AChE)-positive nerve fibers in the chicken pancreas was investigated with histochemical methods at the light and electron microscopic level. AChE-positive nerve bundles were found to run along the pancreaticoduodenal artery and their branches proceed into interlobular connective tissue, form a plexus around the interlobular secretory ducts and small arteries, and penetrate the exocrine parenchyma. Intrapancreatic ganglia showing a strong AChE activity were detected within the interlobular connective tissue or between acini. The exocrine pancreas was richly innervated with AChE-positive terminals which contained a few large dense-cored vesicles (about 100 nm in diameter) and many small clear vesicles (about 50 nm in diameter). Such terminals made contact with intercalated ductular cells and the smooth muscles of larger blood vessels. The endocrine pancreas was supplied with fewer nerves than the exocrine pancreas. A different distribution of AChE-positive fibers was noticed between A- and B-islets which were distinguished immunohistochemically. B- and D-cells were richly innervated by AChE-positive nerves, whereas A-cells, only poorly. These observations make clear that the cholinergic system relates to the regulation of both exocrine and endocrine tissues, except A-cells, in the chicken pancreas.

Ciliocytophthoria (ccp) in nasopharyngeal smear from patients with acute tonsillitis.

Ciliocytophthoria (ccp) in nasal or nasopharyngeal secretion from the patients with acute tonsillitis, acute rhinosinusitis and allergic rhinitis was studied. The ratio of the presence of ccp among acute tonsillitis, acute rhinosinusitis and allergic rhinitis was compared. Acute tonsillitis had a much more positive ccp test than other diseases. This might be related to the anatomical feature of palatine tonsil having a complicated hollow-crypt. The presence of Haemophilus influenzae was the most frequent in patients with positive ccp test. Further study will be necessary to determine possible relationship between viral infection and bacteria or endotoxin from bacteria.
