Levels of selected metals in ambient air PM10 in an urban site of Zaragoza (Spain).

An assessment of the air quality of Zaragoza (Spain) was performed by determining the trace element content in airborne PM10 in a sampling campaign from July 2001 to July 2002. Samples were collected in a heavy traffic area with a high volume air sampler provided with a PM10 cutoff inlet. The levels of 16 elements (Al, Ba, Ca, Co, Cr, Cu, Fe, K, Mg, Mn, Na, Ni, Pb, Sr, V, and Zn) were quantified after collecting the PM10 on Teflon-coated glass fiber filters (GFF). Regarding the PM10, 32% exceedance of the proposed PM10 daily limit was obtained, some of them corresponding to summer and autumn periods. The limit values of toxic trace elements from US-EPA, WHO, and EC were not exceeded, considering Zaragoza as a moderately polluted city under the current air quality guidelines. The contribution of anthropogenic sources to atmospheric elemental levels was reflected by the high values of enrichment factors for Zn, Pb, and Cu compared to the average crustal composition. Statistical analyses also determined the contribution of different sources to the PM10, finding that vehicle traffic and anthropogenic emissions related to combustion and industrial processes were the main pollutant sources as well as natural sources associated with transport of dust from Africa for specific dates. Regarding the influence of meteorological conditions on PM10 and trace elements concentrations, it was found that calm weather conditions with low wind speed favor the PM10 collection and the pollution for trace elements, suggesting the influence of local sources.

Application of polymerase chain reaction to detect adulteration of sheep's milk with goats' milk.

The polymerase chain reaction has been applied for the specific detection of goats' milk in sheep's milk using primers targeting the mitochondrial 12S ribosomal RNA gene. The use of goat-specific primers yielded a 122-bp fragment from goats' milk DNA, whereas no amplification signal was obtained in sheep's, cows', and water buffaloes' milk DNA. Polymerase chain reaction analysis of raw and heat-treated milk binary mixtures of sheep/goat enabled the specific detection of goats' milk with a sensitivity threshold of 0.1%. This study demonstrates the usefulness of the proposed polymerase chain reaction assay for authentication of milk products in routine analysis.

Longitudinal assessment of nutritional status in children treated for acute lymphoblastic leukaemia in Cuba.

Malnutrition has a deleterious effect on the results of therapy for malignant diseases in childhood. The impact of radiotherapy on growth is well known but the impact of cytotoxic drugs on nutritional status is more controversial. The purpose of this study was to determine the nutritional status of a cohort of children treated for acute lymphoblastic leukaemia (ALL) in Cuba. The study involved 49 children admitted to a single center and treated with a Berlin-Frankfurt-Munster-based protocol. Nutritional assessment included measurements of height, weight, body mass index and skin-fold thickness, made at diagnosis, after the intensive phase of treatment and at the end of therapy. Z-scores were used for height and comparison of percentiles for the rest of the variables. All the patients were above the third percentile in all the measurements. There were no statistically significant differences between the results at diagnosis, after intensive therapy and at the end of treatment. Although the sample was small, there was no demonstrable effect of chemotherapy on nutritional status in this Cuban paediatric population, in contrast to that reported in children with ALL in other developing countries.

Rapid detection of cows' milk in sheeps' and goats' milk by a species-specific polymerase chain reaction technique.

A polymerase chain reaction (PCR) assay was developed for the specific identification of cows' milk in sheep's and goats' milk by using primers targeting the mitochondrial 12S rRNA gene. The use of a forward primer complementary to a conserved DNA sequence, along with a reverse primer specific for cow, yielded a 223-bp fragment from cows' milk DNA, whereas no amplification signal was obtained in sheep's and goats' milk DNA. The technique was applied to raw, pasteurized, and sterilized milk binary mixtures of cow-sheep and cow-goat, enabling the specific detection of cows' milk with a good sensitivity threshold (0.1%). The proposed PCR assay represents a rapid and straightforward method applicable to the authentication of milk and other dairy products in routine analysis.

Three-ring PAH removal from waste hot gas by sorbents: influence of the sorbent characteristics.

Three-ring polycyclic aromatic hydrocarbons (PAH) are one of the most abundant PAH groups emitted during coal combustion. Four of them-acenaphthene (Ac), phenanthrene (Phe), fluorene (Fu), and anthracene (An)-have been listed by the U.S. EPA as priority pollutants. The aim in this paper is to study the abatement of this particular group of three-ring PAH from hot gas emissions during energy generation in coal combustion. The three-ring PAH adsorption capacities are related to the morphological and chemical properties of the 16 sorbents used in this work. Single and multiple linear regressions-principal component regression (PCR)-were applied in this study. The main conclusions reached are, first, that the micropore volume is the most determinant parameter for removal of these PAHs and, second, that the adsorption of three-ring PAH by sorbents from waste hot gas emissions is inversely proportional to their volatility: the lower the PAH volatility, the higher the adsorbent adsorption capacity. The adsorption isotherms show that Phe and An, both examples of PAH with three aromatic rings, behave similarly. However, their behavior differs from that of Ac and Fu, compounds in which only two of their three rings exhibit an aromatic nature.

Antidisialosyl antibodies in chronic idiopathic ataxic neuropathy.

Antidisialosyl antibodies were found in two out of 13 patients with chronic idiopathic ataxic neuropathy (CIAN) and not in 32 patients with different sensory neuropathies of known cause. This finding confirms the association of antidisialosyl antibodies and CIAN regardless of the absence of the M band. These antibodies may have pathogenic relevance; however, larger series are needed to establish their clinical significance.

Benzo(a)pyrene, benzo(a)anthracene, and dibenzo(a,h)anthracene emissions from coal and waste tire energy generation at atmospheric fluidized bed combustion (AFBC).

The main aim of this work was the analyzing of the release to the atmosphere of benzo[a]pyrene (BaP), dibenzo(a,h)anthracene (D(a,h)A), and benzo[a]anthracene (BaA), three of the most carcinogenic PAHs listed by US-EPA as priority pollutants, emitted from combustion at the last generation reactors used nowadays in power generation, fluidized bed reactors, trying to establish their incidence when waste materials are used as "new fuels". BaP, D(a,h)A, and BaA emissions, once collected in the sampling system, have been analyzed by fluorescence spectroscopy in the synchronous mode (FS) after extraction by ultrasonic bath with dimethylformamide (DMF) as solvent. Concerning to the combustion variables influence, the conclusion reached was that, in coal combustion, the lowest emissions of BaP are generated at percentages of excess oxygen of 20%, at flows corresponding to good fluidization conditions, 860 L/h (double of the minimum fluidization velocity) and temperatures out of 850 degrees C; the lowest emission of D(a,h)A are emitted at 20% excess oxygen, 900 L/h and out of 750 degrees C, and the lowest emission of BaA are generated at 20% excess oxygen, 860 L/h and out of the range 750-850 degrees C. Regarding tire as nonfossil fuel, higher emissions of BaP, BaA, and D(a,h)A are detected in comparison to coal combustion. In coal-tire blend, the BaP, BaA, and D(a,h)A emissions are more similar to the values obtained in coal combustion and not intermediate values as it could be expected by the blend composition.

Removal of naphthalene, phenanthrene, and pyrene by sorbents from hot gas.

It is the first time that the removal of polycyclic aromatic hydrocarbons (PAH) containing different aromatic rings number [naphthalene (Np), phenanthrene (Phe), and pyrene (Py)] from combustion hot gas has been carried out. The aim was to relate the sorbents textural characteristics with the adsorption capacity of these 2-4-ring PAH at the conditions emitted at energy generation. The sorbents textural parameters [total micropore volume (VN2), narrow micropore volume (VCO2), mesopore volume (VBJH), and the free active sites] were analyzed trying to correlate them with their Np, Phe, and Py adsorption capacities. To get this aim, single and multiple linear regressions (MLR) were applied to the three PAH. A principal component analysis was performed to generate new and uncorrelated variables. It enabled us to show that the relations between the textural parameters were analyzed using a principal components regression (PCR). The PCR analysis had a good statistical quality, but neither did it allow differentiating free active site types nor did VN2 and VCO2. The correlations were thus set up applying a MLR to the original variables. The regression statistical quality was similar to the PCR analysis, and it could give an easier explanation of the parameters that affected the adsorption. In Np adsorption, the 87% data variance was explained, and the adsorption was positively correlated to VCO2 and the micropore mean diameter (I.). In the Phe regression there was 98% variance explained, and its adsorption was positively correlated to the VN2 and the micropore distribution, n. Finally, in the Py adsorption, the 96% data variance was explained, and this adsorption was positively correlated to VN2 and VBJH. These dependencies were according to the molecular parameters of these compounds (molecular diameter and volatility) because the higher the number of aromatic rings of the PAH, the more favored the adsorbate-adsorbate interactions. Besides, the higher the mean diameter micropores, the lower the diffusional problems showed by Np, Phe, and Py.

Familial atypical progressive supranuclear palsy associated with homozigosity for the delN296 mutation in the tau gene.

Heterozygous missense and splice-site mutations in the tau gene have been previously identified in familial frontotemporal dementia with autosomal dominant inheritance. Here we report a Spanish kindred in which two brothers born from a third-degree consanguineous marriage were both affected with atypical progressive supranuclear palsy. A homozygous deletion at codon 296 (delN296) was identified in one of the affected siblings. Among the heterozygous carriers, two members with probable Parkinson's disease were identified, but none of heterozygotes developed atypical parkinsonism. The delN296 mutation lies in the sequence corresponding to the second tubulin-binding repeat of tau protein and affects one asparagine residue absolutely conserved in other species. This finding indicates that homozygous mutations in the tau gene may also cause hereditary tauopathies.

PCR-SSCP: a simple method for the authentication of grouper (Epinephelus guaza), wreck fish (Polyprion americanus), and Nile perch (Lates niloticus) fillets.

A method of DNA analysis has been developed to verify the authenticity of grouper (Epinephelus guaza), wreck fish (Polyprion americanus), and Nile perch (Lates niloticus) fillets. A short fragment (208 bp) of the mitochondrial 12S rRNA gene was amplified by the polymerase chain reaction and analyzed by single-strand conformation polymorphism to get species-specific patterns of single-stranded DNA (ssDNA). DNA strands were separated by native polyacrylamide gel electrophoresis and visualized by silver staining. Discrimination among the three fish species studied was possible, because each one expressed a specific ssDNA pattern.

Identification of goose (Anser anser) and mule duck (Anasplatyrhynchos x Cairina moschata) foie gras by multiplex polymerase chain reaction amplification of the 5S RDNA gene.

Polymerase chain reaction (PCR) amplification of the nuclear 5S rDNA gene has been used for the identification of goose and mule duck foie gras. Two species-specific reverse primers were designed and used in a multiplex reaction, together with a forward universal primer, to amplify specific fragments of the 5S rDNA in each species. The different sizes of the species-specific amplicons, separated by agarose gel electrophoresis, allowed clear identification of goose and mule duck foie gras samples. This genetic marker can be useful for detecting fraudulent substitution of the duck liver for the more expensive goose liver.

Kinetic modelling of the synthesis of 2-hydroxy-5-hexenyl 2-chlorobutyrate ester by an immobilised lipase.

An esterification process was developed for the direct synthesis of 2-hydroxy-5-hexenyl 2-chlorobutyrate ester from 2-chlorobutyric acid by using the epoxide 1,2-epoxy-5-hexene and Mucor miehei immobilised lipase as the biocatalyst in a batch reactor. The effect of temperature, catalyst concentration, and substrate and product concentration has been studied. An ordered Bi Uni enzymatic mechanism with competitive inhibition by the epoxide and acid has been proposed. The corresponding kinetic parameters were calculated by non-linear regression. Activation energy shows a value of 8.04kcal/mol. The thermodynamic parameters of the process, enthalpy and entropy, were 15.4kcal/mol and 45cal/mol, respectively.

A study on the microbiota from olive-mill wastewater (OMW) disposal lagoons, with emphasis on filamentous fungi and their biodegradative potential.

The microbial composition of olive mill wastewater (OMW) from four disposal ponds has been studied. Such OMW samples contained a variable (but high) number of bacteria, yeasts and molds. Among the latest, members of twelve different genera (Acremonium, Alternaria, Aspergillus, Chalara, Fusarium, Lecytophora, Paecilomyces, Penicillium, Phoma, Phycomyces, Rhinocladiella and Scopulariopsis) were found. Members of five genera (Chalara, Fusarium, Paecilomyces, Penicillium and Scopulariopsis) were widely distributed, and they were able to grow efficiently in undiluted OMW as a sole source of nutrients. Strains of Fusarium, Paecilomyces, Penicillium and Scopulariopsis showed a marked capacity for OMW detoxification, depleting its antibacterial activity almost completely.

Identification of the clam species Ruditapes decussatus (Grooved carpet shell), Venerupis pullastra (Pullet carpet shell), and Ruditapes philippinarum (Japanese carpet shell)by PCR-RFLP.

PCR-RFLP analysis has been applied to the identification of three clam species: Ruditapes decussatus (grooved carpet shell), Venerupis pullastra (pullet carpet shell), and Ruditapes philippinarum (Japanese carpet shell). PCR amplification was carried out using a set of primers designed from the DNA nucleotide sequences reported for alpha-actins from humans and various animals. Restriction endonuclease analysis based on sequence data of the PCR products of each clam species revealed the presence of species-specific polymorphic sites for MaeIII and RsaI endonucleases. Electrophoretic analysis of the amplicons digested with MaeIII and RsaI produced species-specific profiles that allowed the genetic identification of the three clam species.

Successful surgical treatment of haemophilic pseudotumour, filling the defect with hydroxyapatite.

A preliminary report is made on the use of hydroxyapatite for surgical procedure of pseudotumour, a rare but serious complication of the bleeding diathesis in patients with inherited bleeding disorders. Surgical or percutaneous treatment and refilling with fibrin sealant is shown to be a successful therapy in a 19-year-old male with severe haemophilia B. The pseudotumour, in the upper pad of the left leg, was filled with hydroxyapatite after surgery. We suggest that the use of hydroxyapatite is a new and useful option in the surgical treatment of haemophilic pseudotumour.

Development of a combined PCR-culture technique for the rapid detection of Arcobacter spp. in chicken meat.

A combined PCR-culture technique was developed to detect Arcobacter spp. in fresh chicken meat. Following a short selective enrichment of chicken samples, bacterial DNA was extracted and amplified using primers targeted at the genes encoding 16S rRNA of Arcobacter spp. The selected primers amplify a 181-bp fragment from all Arcobacter spp., whereas no PCR product is generated for other bacteria, including the closely related Campylobacter and Helicobacter species. The assay was used to screen 96 retail-purchased chicken samples for the presence of Arcobacter spp. Fifty-three percent of the samples analysed were positive for this micro-organism. The assay is simple and sensitive and reduces the amount of time required to positively detect Arcobacter spp. in poultry meat.

Identification of Nile perch (Lates niloticus), grouper (Epinephelus guaza), and wreck fish (Polyprion americanus) by polymerase chain reaction-restriction fragment length polymorphism of a 12S rRNA gene fragment.

Restriction site analysis of polymerase chain reaction (PCR) products from a conserved region of the 12S rRNA gene has been used for the specific identification of Nile perch (Lates niloticus), grouper (Epinephelus guaza), and wreck fish (Polyprion americanus). Amplification of DNA isolated from muscle samples was carried out using a set of primers flanking a region of 436 bp from the mitochondrial 12S rRNA gene. Digestions of the PCR products with RsaI and Sau96I endonucleases, followed by agarose gel electrophoresis of the digested PCR products, yielded specific profiles that enabled direct identification of each species analyzed.

[Chagas diseases seroepidemiology in schoolchildren of Jujuy].

Chagas disease constitutes the main zoonosis in the province of Jujuy, Argentina, where it is one of the most important issues in public health. The purpose of this paper is to analyze the results of a serologic evaluation carried out for a seven-year period among schoolchildren in the Jujenean capital city. The population under study consisted of all seventh grade students of all schools in San Salvador de Jujuy. They were classified into three socioeconomic levels: High, Medium and Low levels. Indirect hemagglutination and immunofluorescence tests were performed. Percentages of seroprevalence were determined by sex, age group, and socioeconomic level. To analyze and check results, the following tests were applied: ANOVA, Tukey's and chi-square test. General prevalence was 1.95% with inter-annual statistically non-significant variations. Statistically significant variations were found among: 1) sex, where the feminine sex exhibited higher seroprevalence; 2) age groups, in which 12-year-olds showed higher seroprevalence; 3) socioeconomic levels, where seroprevalence increased as socioeconomic level decreased. Chagasic seroprevalence in children populations is an indicator that allows assessing both transmission risks in the community and the efficiency of preventive measures to control the vector. Data resulting from this study would indicate: 1) an adequate control both of non-vectorial transmission as well as of the vector, since no temporal variation was recorded in seroprevalence in the age-group analyzed; 2) higher seroprevalence in children belonging to a lower socioeconomic level, probably due to migrations of already-infected mothers coming from neighboring endemic, less epidemiologically controlled areas.
