Isolation and characterization of DNA polymerases from mature oocytes of the starfish, Asterina pectinifera.

Four distinctive DNA polymerases have been isolated from mature oocytes of the starfish, Asterina pectinifera, and characterized. The N-ethylmaleimide-resistant DNA polymerase activity was separated from the other three DNA polymerases by hydroxyapatite column chromatography. The enzyme, whose approximate sedimentation coefficient was 3.3S, effectively utilized poly(dA)-oligo(dT) as a template-primer and was completely inhibited by 10 microM 2', 3'-dideoxythymidine-5'-triphosphate but was not inhibited by aphidicolin. Therefore, this enzyme was classified as a eukaryotic DNA polymerase-beta. Three species of the N-ethylmaleimide-sensitive DNA polymerase were separated with a phosphocellulose column. Two (5.5S and 6.5S) of the enzymes eluted from the column preferred activated DNA as a template-primer. Both of the DNA polymerases were inhibited by aphidicolin but not by 2',3'-dideoxythymidine-5'-triphosphate. Judging from the above results, these two DNA polymerases were classified as eukaryotic DNA polymerase-alpha. The third N-ethylmaleimide-sensitive enzyme (approximately 7S) eluted from the phosphocellulose column utilized poly(rA)-oligo(dT) as well as poly(dA)-oligo(dT) as template-primers. The enzyme activity was resistant to aphidicolin but sensitive to 2',3'-dideoxythymidine-5'-triphosphate. This enzyme resembles eukaryotic DNA polymerase-gamma. Thus, it is concluded that the four species of DNA polymerase found in starfish oocytes resemble eukaryotic DNA polymerase-alpha, -beta, and -gamma.

Pre- and postoperative arrhythmias in congenital heart disease: from the results of surgery using surface-induced deep hypothermia.

The incidence of arrhythmias in 3614 preoperative patients with congenital heart disease was 1.6 per cent. Pre- and postoperative serial ECGs in 186 patients with an isolated VSD and in 58 patients with TOF were reviewed. The incidence of CRBBB and CRBBB-LAH following VSD repair was 33% and 6.6%, respectively, and for TOF was 55% and 5.2%, respectively. Complete A-V block (CAVB) was seen in 2 patients following VSD repair, and none of TOF patients developed CAVB. Late cardiac or dysrrhythmic death was not recognized in our limited follow up period. The indications for surgery in membranous VSD should be determined more carefully, because the risk of the surgical damage to the conduction system is still higher. Moreover, we must keep in mind that damage to the A-V conduction system may occur naturally even in a patient with small VSD. Finally, it can never be overemphasized that physicians must pay attention to the occurrence and course of postoperative arrhythmias.

Relationship between the degree of injury at operation and the change in antimyosin antibody titer in the postpericardiotomy syndrome.

Successive measurements of cardiac myosin light chain I (MLC), creatine kinase isoenzyme MB (CKMB), and the titer of antimyosin antibody (AMA) were performed prospectively in 19 patients following open heart surgery. Seven of these patients showed the postpericardiotomy syndrome (PPS). No differences in serum concentrations of MLC or CKMB were observed between the patients with and without PPS, and all patients in both groups had abnormal MLC values after surgery. However, only patients with PPS had significantly elevated AMA titers. The maximum AMA titer was significantly correlated with the severity of the effusion. These data suggest that PPS is unrelated to the severity of myocardial injury during operation. Furthermore, the AMA titer may be useful as one of the indicators for determining the patient's clinical condition.

[Latentiation of therapeutic drugs. VI. Preparation of l-amino-4-naphtylazoic ambonates of schistosomicides].

With the purpose of obtaining latent forms, with prolonged action and less toxic, of four schistosomicidal 1-amino-4-naphthylazoderivatives synthesized early, embonates of these compounds were prepared by reacting disodium and tetrasodium embonates with hydrochlorides of the four naphthylazoderivatives. The eight embonates thus obtained were sent for biological assays.
