Modulation of leukotriene generation by pertussis toxin.

The purpose of our study was to characterize the properties of the interaction of pertussis toxin with human polymorphonuclear leukocytes for the modulation of leukotriene generation and metabolism. The cells were stimulated with either the Ca ionophore A23187, opsonized zymosan, or the bacterial peptide formyl-methionyl-leucyl phenylalanine. Incubation of the cells with pertussis toxin led to a rapid inhibition of LTB4 generation when formyl-methionyl-leucyl phenylalanine was used as the stimulus, whereas there was no effect with the Ca ionophore and just a low effect with opsonized zymosan. The inhibition of leukotriene generation was dependent on the incubation time, temperature, and pertussis toxin concentration. The effect was not dependent on the presence of calcium. Incubation of the cells with guanosine 5'-O-(3-thiotriphosphate) the stable analog of GTP, led to a time-dependent increase in leukotriene generation induced by formyl-methionyl-leucyl phenylalanine which was abolished by the simultaneous addition of pertussis toxin. Our data suggest that the formyl-methionyl-leucyl phenylalanine-induced generation of leukotrienes is dependent on a GTP-binding protein. The participation of the various G proteins has yet to be elucidated.

Arachidonic acid metabolites from polymorphonuclear leukocytes of healthy donors, severely burned patients and children with cystic fibrosis--routine monitoring by high-performance liquid chromatography.

A rapid and sensitive high-performance liquid chromatographic method for the determination of arachidonic acid metabolites was developed. This method provides a clear and simple separation of the omega-oxidation products as well as of leukotriene B4 and 5S,12S-dihydroxy-6-cis-8-trans-10-trans-14-cis-eicosatetraenoic acid. Furthermore, a solvent switch enables the detection of the monohydroxyeicosatetraenoic acids together with the cysteinyl-leukotrienes as well as leukotriene B4 and the dihydroxyeicosatetraenoic acids within 70 min (including column equilibration for 10 min). The efficiency of the method for the routine monitoring of leukotrienes was performed with human polymorphonuclear leukocytes of healthy donors, severely burned patients and of children with cystic fibrosis. The advantages of this method include high sensitivity and easy handling, which are important for routine analyses of the arachidonic acid metabolites.

Mass spectrometer artifact: simultaneous detection of two volatile anesthetics.

In several cases, a mass spectrometer indicated that both enflurane and isoflurane were being detected while only isoflurane was in use. The concentration of expired enflurane reported was always higher than the inspired concentration, but never exceeded 0.66% regardless of the concentration of isoflurane. Such mass spectrometer readings may be evidence of vaporizer contamination or malfunction of the vaporizer; however, in this case, the mass spectrometer was found to be out of calibration. Because mass spectrometers are recent additions to clinical equipment, procedures for routine calibration and maintenance must be carefully scrutinized and evaluated.
