Stimulation of human immunodeficiency virus expression in permanent monocytic cells by Sarcocystis gigantea extract.

As recently reported, a strong stimulation of noninfected CD4+ H9- cells by Sarcocystis gigantea (syn. S. ovifelis) extract (SGE) was observed using the lymphocyte proliferation assay. After SGE prestimulation, human immunodeficiency virus (HIV)-infected H9+ cells showed an exacerbation of viral replication. In the present study we investigated the reactivity of HIV-infected human monocytes using SGE. The highly sensitive p24 core-profile enzyme-linked immunosorbent assay (ELISA) was used to examine directly the amount of HIV produced. Experiments were performed using U937 permanent monocytic cells. Permanent incubation as well as preincubation with SGE before virus infection stimulated HIV expression in all the cells. In U937 cells the viral release per cell was 64 times higher on permanent stimulation with 320 microg SGE relative to controls and 9 times higher following prestimulation.

[Evaluation of an intracutaneous test using a Sarcoptes mite extract solution (Acari: Sarcoptidae) as a method for detection of Sarcoptes mite-infested dogs].

Sarcoptes infestation in dogs, caused by Sarcoptes canis, is a relatively common disease in small animal practice. The parasites may induce severe allergic skin reactions. By means of clinical symptoms a presumptive diagnosis should be made. For ensurement detection of mites in skin scrapings is necessary, but it is not easy to find the parasites. Serodiagnostic methods are helpful to confirm the diagnosis. They indicate specific antibody (circulating IgE) titer. Intracutaneous test by using allergen extracts as possible third way of diagnostic methods was tested comparatively with the existing causaldiagnostic procedures in 45 dogs with suspected scabies. Preconditions of own examinations was mite antigen preparation. A mite extract solution of Sarcoptes suis was prepared and 0.1 ml were applicated intracutaneously. In 14 dogs (31.11%) allergic skin changes (Immediate reaction type 1) became apparent. The results were opposed to both other detection methods--skin scraping (4 positive findings/8.89%) and serodiagnosis (13 positive findings/30.77%).

[Allergenicity and specific protein profiles of mange mites Chorioptes bovis, Psoroptes ovis (Acari: Psoroptidae), Saroptes suis and Notoedres cati (Acari: Sarcoptidae) using SDS-PAGE and immunoblotting].

A main point of immunoparasitological research in regard to pest arthropod-infestation is biochemical and immunological characterization of antigens. Precondition of own examinations to the specific protein pattern of mange mites were in quality and amount sufficient antigen preparations in mite extract solutions. For mite separation and antigen refinement field strains of Chorioptes bovis, Psoroptes ovis, Sarcoptes suis and Notoedres cati from definitive host animals cattle, sheep, pig and cat have been used. Parasites were isolated in a migration procedure. After having applicated subepidermally a low dose of mite extract solutions in sensitized animals allergic skin changes (Immediate reaction type 1) became apparent. SDS-PAGE exhibited specific protein patterns of 4 pathogen mite species. For Chorioptes bovis 16, Psoroptes ovis 15, Sarcoptes suis 27, and Notoedres cati 36 fractions have been detected. Proteins are antigens or allergen structures to be found in saliva, faecal output or moulting products of developmental stages and other metabolites of the parasites. Protein components were transferred onto nitrocellulosis. Immunoblotting made fractions with antigen activity visible.

[Effect of Sarcocystis gigantea extract (SGE) on the replication of human immunodeficiency virus (HIV)].

Basis of this study were the previous findings regarding isolation and characterization of a Sarcocystis gigantea lectin (SGL) especially the activation of human mononuclear cells (CD3+, CD4+, CD8+, Mø, B-lymphocytes). HIV-susceptible, immortaliced cell lines (H9-, MT-4) should be investigated to examine their reactivity against SGE which contains this strong mitogen. Using lymphocyte proliferation assay a strong stimulation of noninfected CD(4+)-positive H9-cell by SGE was observed. HIV-infected H(9+)-cells showed after SGE stimulation (20-160 micrograms) an exacerbation with an optimum at day 4. The virus replication in the H(9+)-hostcells was 13 times stronger. This result could also be detected indirectly because of the higher cytotoxicity in the MT-4 cell system. Cytopathogeny was measured by MTT cellvitality assay. In parallel, the high sensitive p24 core Profile-ELISA was used to directly examine the amount of produced HIV. In case of the H(9+)-cells the virus release per cell was 5 times higher after SGE stimulation compared with control.

Production of autoantibodies against DNA and collagen after inoculation of rabbits with Trypanosoma equiperdum.

The aim of the study was to investigate the induction of autoantibodies in rabbits after the inoculation with Trypanosoma equiperdum. Eight female rabbits were inoculated s.c. with 5 x 10(6) parasites of Trypanosoma equiperdum. Four of these animals were treated with Diminazen (Berenil) at a dosage of 28 mg per rabbit by intramuscular injection 35 days p. i. Two additional non-infected rabbits served as negative controls. 9 days p.i. all infected animals possessed serum antibodies against Typanosoma equiperdum. Titers peaked to 1:2048, measured between 14 and 30 days p.i. and were persistent until the end of examinations. In parallel, there was an increase of anti-dsDNA and anti-collagen autoantibodies, but not of autoantibodies against recombinant U1RNP and La antigens. In comparison to patients with systemic lupus erythematosus, the anti-DNA binding of rabbit's sera was reduced more strongly in presence of increasing salt concentrations. In addition, the sera of inoculated rabbits recognized several unknown bands of a cell extract in immunoblotting. The parallel increase of antibodies against Trypanosoma, control foreign antigens, DNA and collagen suggests a polyclonal stimulation of the immune system. It was concluded that this rabbit model seems to be sufficient to investigate the mechanisms which are able to induce autoimmune phenomena.

Prevalence of ticks in relation to their role as vector of Borrelia burgdorferi under autochthone conditions.

A total of 59 (78.7%) of 75 areas were found to be inhabitated by ticks; all the 4335 collected ticks were identified as Ixodes ricinus. Both nymphs and adults were found to be more active during the late spring and early summer months. Ticks of 29 recreation areas were found to be carrying spirochaetes. Considering the total number of 75 recreation areas examined and the total number of 59 areas inhabitated by ticks the percentage was 38.7 and 49.2 respectively. The positivity rate by dark field microscopy was 16.7% in 653 tick samples (single nymphs 8.8%, pooled nymph samples 21.5%, single females 10.9% and single males 10.8%). Overall positivity rate by culture examination of 245 samples was 22.8%. It was maximal in nymphs (24.7%) followed by males (21.1%) and females (19.7%). 10.2% of the 177 samples, found negative by DFM, were positive by this method. A positivity rate of 23.9% in 96 tick samples was found by IFAT. A total of 56 spirochaete-strains were obtained of which 51 could be maintained further; 50 strains were identified as Borrelia burgdorferi.

[Helminth control in sheep and goat flocks].

Pasture helminthoses of sheep and goats should permanently be under parasitological control. Starting from 6 principles the economically most important pasture helminthoses--Trichostrongylidosis, Dictyocaulosis, Protostrongylinosis, Monieziosis, Fasciolosis and Dicrocoeliosis--are reviewed under the aspect of control strategies. The controlled use of highly effective anthelmintics of different compound classes is suitable in regulating helminth populations. The problems of anthelmintic resistance and anthelmintic residues are mentioned.

[Risk for humans and animals by excretion of helminth eggs].

In presence of large human and animal populations on a small area potential danger is given for frequent occurrence of helminth infections. Important helminthosis of men and animals in Middle Europe are represented, particularly the correlations between hosts and parasitic agents in epidemiological point of view. The represented helminth species are such of man as final host which are dangerous for men and animals, and such of animals which are dangerous also for both man and animals. On the one hand there is a lot of new scientific findings, on the other hand there are gaps in knowledge, concerning species of nematodes, cestodes and trematodes with reference to living expectancy, reproduction potential, duration and dynamic of elimination as well as resistance of exogenous helminth stages and finally methods for proof of pathogens in the biotope as a requirement for purposeful control with ecologically acceptance.

[Autoimmune phenomena during Trypanosoma equiperdum inoculation of the laboratory mouse].

In the course of the Trypanosoma equiperdum-infection of mice an increase of IgM antibodies against the autoantigens dsDNA, keratin and collagen as well as against a protozoan foreign antigen consisting of Sarcocystis gigantea-extract could be observed with a maximum level between 4th and 8th day p. i. The IgG-antibodies did not significantly change during the investigation time. A splenomegaly appeared after the infection. The weight of spleen was four times higher than normal. It was suggested that splenomegaly as well as induction of antibodies against several autoantigens and a foreign antigen were due to a polyclonal activation of lymphocytes.

[The use of cryopreserved swine blood for in vitro feeding of the soft tick Ornithodoros moubata].

Porcine stored blood has been used in the in vitro feeding of the soft tick Ornithodoros moubata through a Parafilm membrane. The efficiency of the feeding decreased after storage of 5 weeks by -20 degrees C. Through addition of 10(-3) M ATP/l the feeding results with frozen blood for duration of storage up to 200 days well agreed with the results of feeding tests with fresh porcine blood: The nymphs (N)1-3 fed the 4-5fold, the N4-6 the 2.7fold and the adults (A) the 1.9fold of their body weight. The feeding rate was 98.6% (N1-3); 92.7% (N4-5) and 81.9% (A). The mortality rate was under 10%. Females laid 130 eggs after blood meal.

[The book about equery, by Jordanus Ruffus (13th century), in Ms 78 C of the copper engraving collection, Berlin].

One of the italian translations of the "Medicina equorum" written by the major stablemaster of the Emperor Frederic II. of Hohenstaufen, JORDANUS RUFFUS, dates to the late 13th century. It forms part of a manuscript kept in the State Library of Berlin-West. It is the only copy of the "Medicina equorum" which depicts the treatments of the different horse diseases in a very instructive manner. A translation into German of this italian manuscript was carried out recently in our Institute. This paper emphasizes the historical value of this opus and makes comments on the illustrations.

Activation of human CD4+ and CD8+ cells by Sarcocystis gigantea lectin.

As recently reported, Sarcocystis gigantea lectin (SGL) is a powerful mitogen and a polyclonal activator (Syn. S. ovifelis) of human peripheral B-cells. In the present study we investigated the reactivity of human T-helper (CD4) and T-suppressor (CD8) cells to SGL. Mononuclear cells (MNCs) from five newborns and six adults were examined cytofluorometrically for the expression of cell-surface differentiation and activation antigens using a set of seven monoclonal antibodies. In all, 96% of cord-blood and 81% of adult CD4 cells expressed receptors for interleukin-2 (Tac+) after 64 and 164 h microgram/ml). The percentages of neonatal and adult Tac+ CD8 cells amounted to 67% and 59%, respectively. Major histocompatibility complex (MHC) class II antigens identified using the monoclonal anti-human leukocyte antigen (HLA)-DR antibody L243 were expressed on 30% (CD4) and 44% (CD8) of adult T-cells. Neonatal HLA-DR+ T-lymphocytes were not detectable. In parallel, functional tests were performed to examine cell proliferation and MNC antibody production.

[Livestock manure as a vector for parasites--a report of experiences].

Analysing the results of own investigations and informations in literature a review is given about the role of manure specially in stables of high-intensity cattle and pig production as a vector for exogenic parasite stages. In the course of investigations--in most cases simulating practice conditions--so-called indicator germs were used as test material. In cattle stables manure is significant as a vector for Eimeria species, Cryptosporidium parvum, Sarcocystis species, Taenia saginata and Fasciola hepatica. In pig stables manure must be classified as a reservoir of infections with Toxoplasma gondii, Sarcocystis species and Ascaris suum; moreover it represents a favourable culturing medium for stable flies. Possibilities to overcome the problems are discussed under parasitological field of view.

Parasitology in East Germany--roots, period of the G.D.R., future.

The paper starts with a historical reflection on the region and on famous scientific personalities which worked up parasitological problems on the territory of East Germany. Native scientists like J. L. Frisch, C. A. Rudolphi, R. Leuckart, A. C. Gerlach, R. Virchow, R. Koch, R. von Ostertag, W. Nöller, R. Wetzel made in fundamental contributions to the growth of knowledge in General and Special Parasitology. The parasitological institutions existing in East Germany are represented briefly. The parasitologists of East Germany, nearly 160,--biologists, physicians and veterinarians--among them more than 50 specialized parasitologists were associated in the Parasitological Society of the G.D.R. since 1961. A review is given of actual parasitological problems of agricultural and other domestic animals, fair game animals, of bees and freshwater fish as well as of the most important parasitoses of man frequent in East Germany, under the aspects of epidemiology, diagnosis and control. The rapid evolution of an intensive animal production under large scale management conditions required a permanent control of infestation with pathogenic protozoans, arthropods and helminths. Some selected results achieved in research and practice are represented. The epidemiological situation referring to parasitic zoonoses is favourable at present. Since 1970 a group of experts from East Germany working successfully in Mongolia on the systematic control of ectoparasites of farm animals together with Mongolian parasitologists. It is considered to be necessary to make an international agreement in parasitological research in future more than till now.
