[Effect of estradiol on stimulated theophylline and prolactin Ca2+ exit from intracellular stores of pig oocytes].

Effect of estradiol on stimulated theophylline and prolactin Ca2+ exit from intracellular stores of pig oocytes was investigated using fluorescent dye chlortetracycline. It was shown that in the presence of estradiol neithert theophylline nor prolactin stimulated Ca2+ exit from intracellular stores of oocytes. Unlike, the common action oftheophylline and prolactin, also in the presence of estradiol, stimulated Ca2+ exit from intracellular stores. Inhibition of protein kinase C inhibits Ca2+ exit from intracellular stores in common action of theophylline and prolactin. These data suggest an obvious influence of estradiol on Ca2+ exit from intracellular stores of pig oocytes stimulated by theophylline and prolactin.

[Dependence of Ca2+ release from intracellular stores on NADH and FAD levels in fertilized and unfertilized bovine oocytes].

Relation between NADH and FAD concentrations and the quantity of calcium released from intracellular stores in fertilized and unfertilized bovine oocytes was investigated using luminescent analysis. Inhibition of Ca2+ exit from intracellular stores was detected in degenerative oocytes at metaphase II and 2-cell embryos. The intensity of both NADH and FAD fluorescence increased in 2-cell degenerated embryos, whereas the increase in only NADH fluorescence intensity occurred in degenerated oocytes at metaphase II stage. Degeneration exerted no influence on NADH fluorescence intensity or Ca2+ exit from intracellular stores, whereas a decreased FAD fluorescence intensity was noted in degenerated pronuclei. The obtained data testify that in degenerated zygotes and early embryos Ca2+ release may occur from different intracellular stores.

[Peculiarities of theophylline and prolactin interaction and calcium fluctuation from intracellular stores of porcine oocytes in the presence of estradiol].

Using a fluorescent dye chlortetracycline, a study was made of the effect of estradiol on the interaction of theophylline and prolactin in the course of Ca2+ exit from intracellular stores of pig oocytes, isolated from ovaries at the stage of follicle growth. It is shown that in the presence of estradiol, prolactin does not stimulate Ca2+ exit from intracellular stores of pig oocytes. The action of theophylline similarly does not stimulate Ca2+ exit. Unlike, a joint effect of theophylline and prolactin on pig oocytes in the presence estradiol stimulated Ca2+ exit from intracellular stores of pig oocytes. These data demonstrated the influence of estradiol on theophylline and prolactin stimulated Ca2+ exit from intracellular stores of pig oocytes.

[Participation of granulosa cells in mediation of prolactin and somatotropin action on bovine oocyte-cumulus complexes in vitro].

Maturation of bovine oocyte-cumulus complexes (OCC) in media derived following granulosa cell culturing with prolactin (PRL, 50 ng/ml) and somatotropin (ST, 10 ng/ml) was studied. A medium conditioned by granulosa cells in the presence of PRL or ST exerted a stimulating effect on the proliferative activity of cumulus cells. ST introduction into the granulosa cell culture also caused a decrease in the rate of cumulus cells with degenerated chromatin at a subsequent OCC culturing. At the same time, the expansion of cumulus did not depend on hormone availability in the culture medium for granulosa cells. When OCC matured in conditioned media, a short-term inhibition of oocyte meiosis reinitiation (after 6 h of culturing) was revealed in both the experimental groups, as compared to the control. Furthermore, the addition of ST and PRL to granulosa cell culture resulted in a subsequent decline in the rate of oocytes with signs of chromosome degeneration, observed as early as by 6 h of incubation and to be retained throughout the whole period of OCC culturing. In this case the earlier resumption of meiosis was associated with a higher rate of degeneration of the nuclear material in oocytes. The results of the present study suggest that granulosa cells may mediate, at least in part, PRL and ST impacts on in vitro maturation of bovine OCC, with no contact between OCC and granulosa cells being required for hormonal signaling.

[Effect of progesterone on theophylline and prolactin stimulated Ca2+ exit from intracellular stores of pig oocytes].

Effect of progesterone on theophylline and prolactin stimulated Ca2+ exit from intracellular stores of pig oocytes was investigated using a fluorescent dye chlortetracycline. It is shown that in progesterone treated oocytes prolactin in concentration 50 ng/ml inhibits Ca2+ exit from intracellular stores of pig oocytes. Theophylline exerts the effect on prolactin Ca2+ exit from intracellular stores of pig oocytes. Employment of protein kinase C inhibitor cancelled inhibitory effect of prolactin and theophylline on Ca2+ exit from intracellular stores of pig oocytes. Ca2+ exit from intracellular stores of pig oocytes caused a joint influence of prolactin and GDP, and that of theophylline and GTP. The influence of protein kinase C inhibitor cancelled the stimulating effect of prolactin and GDP on Ca2+ exit from intracellular stores of pig oocytes also did not render any influence on the action of theophylline and GTP. These data suggest the influence of progesterone on theophylline and prolactin stimulated Ca2+ exit from intracellular stores of pig oocytes.

[Prolactin-binding activity of bovine granulosa cells on luteinization and exposure to somatotropin in vitro].

The influence of luteinization and bovine somatotropin (ST, 5-50 ng/ml) during cultivation of bovine granulosa cells on their ability to bind [125I]-labeled bovine prolactin (PRL) was studied. On the second day of cultivation in serumfree medium, granulosa cells from immature antral follicles underwent spontaneous luteinization, in both the absence and presence of ST. The level of [125I]-PRL specific binding to cells increased after two days of cultivation, with a negative correlation being revealed between estradiol production by the cells and their PRL-binding activity. At the same time, the addition of ST to the culture medium had no effect on the level of [125I]-PRL specific binding to native and luteinizing granulosa cells. The findings suggest a stimulatory influence of the luteal differentiation process on the PRL-binding activity of bovine granulosa cells, this influence is independent of the action of ST.

[The influence of guanine nucleotides and protein kinase C on Ca2+ from intracellular stores of pigs oocytes stimulated by theophylline and cyclic AMP].

Effect of guanine nucleotides and protein kinase C on Ca2+ exit from intracellular stores of pig oocytes, stimulated by theophylline and dbcAMP, was investigated using fluorescent dye chlortetracycline. Effect of cAMP on Ca2+ exit from intracellular stores of pig oocytes was not associated with activation of protein kinase C. In calcium-free medium, cAMP does not stimulate Ca2+ exit from intracellular stores of pig oocytes treated with GDP. In the presence of GDP, inhibition of protein kinase C activates Ca2+ exit from intracellular stores of pig oocytes on the action of cAMP. These data suggest the existence of different effects of guanine nucleotides on Ca2+ exit from intracellular stores of pig oocytes stimulated by cAMP.

[The influence of somatotropin, prolactin and insulin on granulosa cells from bovine atretic follicles].

An influence of somatotropin, prolactin and insulin on destructive processes in bovine granulosa cells from small antral follicles following atresia in vivo was studied in vitro. As compared to control, the addition of the studied hormones to serum-free suspension system was shown to result in increase in number of cells without signs of chromosome degeneration after 24 and 48 hrs of incubation. The revealed inhibitory action of somatotropin, prolactin and insulin on chromatin degeneration in granulosa cells was not due to the hormonal influence on proliferative activity of the cells. The stimulatory action of insulin on the viability and estrogen-secretory activity of granulosa cells cultured for 1 day was also found. At the same time, somatotropin and prolactin did not affect the estradiol and progesterone production by the cells. The data obtained suggest that the inhibitory action of somatotropin and prolactin on destructive processes in cultured granulosa cells is not related to the hormonal regulation of the steroidogenic activity of the cells, whereas the similar action of insulin may be partially due to its stimulatory influence on the estradiol secretion.

[Method for determining dopamine and morphine binding sites in lymphocytes from human peripheral blood].

A histochemical method was designed to detect the regions of binding the dopamine and morphine in human peripheral blood lymphocytes. It is based on incubating the suspension of lymphocytes and conjugated dopamine or morphine with bull serum albumin (BSA) marked by horse-radish peroxidase. After incubation, smears are prepared from the lymphocyte suspension, which are stained by diaminobenzidine in the presence of hydrogen peroxide for peroxidase. The light microscope with oil immersion is used to count the number of lymphocytes (from among 100 hundred of them), which contain the peroxidase granules. Smears from the lymphocyte suspension, which were incubated with the BSA-peroxidase conjugate, were controls. The binding of peroxidase-marked ligands of dopamine and mu-opioid receptors with lymphocytes was oppressed by the dose-dependant preliminary incubation with antagonists (haloperidol, naloxone), on the basis of which the presence of the ligand-receptor interaction can be suggested. The number of bindings of dopamine and morphine in lymphocytes was shown to be reliably higher in the alcoholic-intoxication state versus the healthy subjects without any signs of alcohol consumption. The designed method is simple enough in use and does not require any special equipment for the receptor detection in a moderate blood quantity.

[Involvement of protein kinase C in the regulation of Ca2+ exit from intracellular stores of prolactin stimulated pig oocytes].

Involvement of protein kinase C in the regulation of Ca2+ exit from intracellular stores of pig oocytes activated by prolactin was investigated, using the fluorescent dye chlortetracycline. In the presence of extracellular calcium, the inhibitor of protein kinase C Ro 31-8220 increased calcium exit from intracellular stores in pig oocytes after prolactin treatment. In calcium-free medium, Ro 31-8220 exerted effect on calcium release from intracellular stores. In calcium-free medium, prolactin did not stimulate calcium release from intracellular stores of oocytes in the presence of thimerosal, while in the presence of protein kinase C inhibitor, prolactin increased Ca2+ content from intracellular stores in such oocytes. These data suggest a direct involvement of protein kinase C in the processes of regulation of Ca2+ exit from intracellular stores of pig oocytes stimulated by prolactin.

[Effect of the protein kinase C inhibitor on changes in Ca(2+) level in pig granulosa cells induced by prolactin].

The effect of the PKC inhibitor on Ca2+ responses to prolactin in the pig granulosa cells was studied using fluorescent dye and chlortetracycline. The effect was shown to be connected with activation of the PKC. The Ro 31-8220 increased penetration of extracellular calcium and exit of calcium from intracellular stores. The data obtained suggest an involvement of the PKC in changes of calcium contents in the pig granulosa cells activated by prolactin.

[Reciprocal modulation of regulatory action of prolactin and somatotropin on DNA synthesis in granulosa cells of cows].

A comparative analysis of the individual and joint influence of bovine prolactin (BPRL) and bovine somatotropin (BST) on DNA synthesis in cultured bovine granulosa cells from follicles of 3-5 mm in diameter was conducted, and a possibility for regulation of BPRL binding to receptors on the cells by BST was examined. Despite a unidirectional growth-promoting action of BPRL and BST on granulosa cells, their joint mitogenic effect on the cells was not additive at low concentrations of BPRL. The addition of BST to the culture medium did not alter the biphasic character of dependence of DNA synthesis in the cells on BPRL concentration, but increased the maximum effective concentration of the latter. When culturing granulosa cells with BST, the absence of its influence on the level of BPRL specific binding to the cells was shown. This fact suggests that BST modulating action on the mitogenic effect of BPRL is not a result of a change in the number of free receptors for PRL on granulosa cells. The results obtained are discussed in relation to the notion of similarity of receptor functional properties and intracellular signal ways for PRL and ST.

[Effect of the protein kinase C inhibitor Ro 31-8220 on Ca2+-responses, induced by somatotropin, in swine granulosa cells].

Somatotropin effect on Ca2+ responses in pig granulosa cells from antral follicles was investigated using fluorescent dye Fluo-3 AM and chlortetracycline. Ro 31-8220 increased the entry of extracellular calcium and the exit of calcium from intracellular stores. In Ca-free medium Ro 31-8220 exerted no influence on the level of calcium in granulosa cells. The effect of somatotropin on pig granulosa cells is associated with PKC activation. These data suggest the involvement of PKC in the changes of calcium in pig granulosa cells activated by somatotropin.

[Changes in the content of prolactin and somatotropin and the level of their binding with cells in the growing antral follicles of cows].

The hormone concentrations in the follicular fluid rise along with an increase in follicular diameter whereas the level of their binding to cells declines. The findings suggest that two basic parameters characterizing the degree of prolactin and somatotropin actions on granulosa cells develop in different directions in the course of antral follicle maturation.

[The effect of prolactin on DNA synthesis in cultured bovine granulosa cells].

The influence of bovine prolactin (BPL) on DNA synthesis in cultured cow granulosa cells in various media has been studied. BPL exerted a mitogenic effect on granulosa cells of follicles 3-5 mm in diameter, with the effect depending on hormone concentration in the cultural medium. In the medium with bovine growth hormone the maximum effective concentration of BPL was 100 times more than in other used media. The degree of BPL influence on granulosa cell proliferation depended on the degree of cell differentiation. In cultures of granulosa cells taken from follicles 1-2 and 3-5 mm in diameter, while the BPL concentration increased from 500 ng/ml to 5 micrograms/ml in the serum free medium, and from 50 ng/ml to 500 ng/ml in the medium with 5% fetal bovine serum, the stimulation of DNA synthesis in cells decreased. The growth-promoting influence of BPL on granulosa cells of follicles 6-10 mm in diameter did not depend on its concentration in the cultural medium. The maximum mitogenic hormonal effect decreased with the growth of a follicle. This fact is in accord with the recent notions of a progressive reduction of granulosa cell proliferative activity during follicular maturation. The results obtained are discussed in terms of a hypothesis that prolactin can influence granulosa cell secretion of a regulator of meiosis and folliculogenesis, i.e. an inhibitor of oocyte maturation.

[Lactogenic receptors on cow granulosa cells].

Bovine prolactin (BPRL) binding to cow granulosa cells depending on the antral follicular diameter has been studied. The data obtained point to the existence of one type of low affinity PRL-binding sites on granulosa cells. No reliable differences in the parameters of BPRL interaction with lactogenic receptors in the dynamics of folliculogenesis have been found. The recombinant bovine growth hormone and BPRL competitive binding while interacting with lactogenic receptors has been shown that suggests action of these hormones at over the same binding sites on cow granulosa cells.

[Risk factors and transmission routes of the causative agents of campylobacteriosis in commercial poultry plants].

The study, carried out in two regions of the USSR and aimed at estimation of the contamination of products supplied by industrial poultry complexes (IPC), revealed that the contamination of these products was closely related to the Campylobacter contamination of the personnel of IPC. The causes of high Campylobacter contamination of the products of IPC at all technological stages of their production are described. The species, serovars and biovars of Campylobacter strains isolated from different sources were determined, which made it possible to carry out the specific and intraspecific differentiation of these strains.
