[Quantitative evaluation for murine oocyte toxicity following intraperitoneal treatment with chemotherapeutic agents].

The ovarian toxicity induced with 12 oncostatics was evaluated using syngeneic mice, 6-week-old C57BL/6. Each drug diluted with saline to 0.2ml was intraperitoneally infused twice at 6 and 7 weeks old. Mice were sacrificed 2 weeks after the second treatment, the ovaries removed and fixed for serial sectioning, and the small oocytes of Pedersen and Peters counted. Small oocytes were destroyed in a dose-dependent fashion, and ED50, an effective dose of which produced 50% destruction of small oocytes in each mice group, was significantly divided into 4 groups of statistical difference (F = 5.77, p less than 0.0213). The smallest dose of ED50 (mg/mouse) (the strongest in oocyte toxicity): Actinomycin D 0.0064, doxorubicin 0.0184, peplomycin 0.021; the second: Bleomycin 0.107, mitomycin 0.0707, CDDP 0.120; the third: Cyclophosphamide 0.427; and the largest (the weakest): Ifosphamide 3.01, 5FU 6.17, etoposide 6.11, methotrexate 2.0 much less than, vinblastine 0.1 much less than. The most toxic included those which could attack not only DNA, but also RNA biosynthesis and the least included those which could have an effect on enzymes or proteins with no direct action on DNA. The ratio of ED50 to HUD, the single usual dose for human cancer chemotherapy, were smallest for doxorubicin (2.23), cyclophosphamide (2.59) and CDDP (5.19). Although these three are very useful oncostatics for ovarian cancer, they might have serious potential toxicity for human ovarian oocytes.

Vaginal abnormalities in ovariectomized BALB/cCrgl mice after neonatal exposure to different doses of diethylstilbestrol.

Newborn BALB/cCrgl female mice received five daily injections of various doses of diethylstilbestrol (DES), 0.0001-10 micrograms. Mice were killed at 6 days of age or at 4 months after ovariectomy at 40-42 days. Subepithelial nodules of polygonal cells in the upper (Mullerian) vagina during early postnatal life were associated with the later occurrence of ovary-independent persistent stratification with or without cornification in mice treated neonatally with 0.1-10 micrograms DES and thus are a possible predictor of this phenomenon. The thresholds for the induction of ovary-independent epithelial pegs, downgrowths and adenosis (glandular formations) were 0.1 microgram and 0.5 microgram DES/day, respectively.

Permanent chondrification in the pelvis and occurrence of hernias in mice treated neonatally with tamoxifen.

Male and female C57BL/Tw mice were given 5 daily subcutaneous injections of 100 micrograms tamoxifen (Tx), starting on the day of birth (Tx mice). In untreated fetal mice on day 18 of gestation, the greater part of the pubic and ischial bones were cartilaginous. At more than 30 days of age, however, untreated mice showed completely calcified pelvic bone, whereas in age-matched Tx mice the greater part of the junctional regions in the pelvis remained cartilaginous. Treatment with Tx starting within 5 days of age caused bladder hernia with or without cecum hernia. The pubic ligament in Tx mice at ages of 30-540 days was markedly expanded as compared with that in age-matched controls. The permanent chondrification in the pelvis was found in all mice given Tx starting within 10 days of age. By contrast, neonatal treatments of mice with other antiestrogens, clomiphene and nafoxidine (100 micrograms/day), induced neither permanent chondrification in the pelvis nor expansion of the pubic ligament nor hernia. These findings suggest that Tx has a specific effect on the symphysis pubis and some junctional regions of the developing pelvis in mice when given neonatally.

The effects of an aromatase inhibitor and a 5 alpha-reductase inhibitor upon the occurrence of polyovular follicles, persistent anovulation, and permanent vaginal stratification in mice treated neonatally with testosterone.

Female C57BL/Tw mice were given 5 daily injection of 20 micrograms testosterone (T), 100 micrograms 4-hydroxy-4-androstene-3, 17-dione (4-HA), 100 micrograms 6-methylene-4-pregnene-3, 20-dione (6-MP), 4-HA + 6-MP, T + 4-HA, T + 6-MP, and T + 4-HA + 6-MP starting on the day of birth. The animals were ovariectomized at 30 days or 90 days of age and were killed at 150 days. The incidence of polyovular follicles (PF) at 30 days of age was significantly increased by neonatal treatment with T. By contrast, the PF incidence was significantly reduced by injections of 4-HA given simultaneously with T. Neonatally T- or T + 6-MP-treated 90-day-old mice had ovaries containing follicles and hypertrophied interstitial cells but no corpora lutea. By contrast, T + 4-HA (64%)- and T + 4-HA + 6-MP (82%)-treated mice had ovaries with corpora lutea. In the T + 4-HA + 6-MP (82%)-treated mice had ovaries with corpora lutea. In the T + 4-HA + 6-MP-treated, 150-day-old, ovariectomized mice, the number of mice showing vaginal epithelial stratification was significantly decreased as compared with T-treated mice. There were no significant differences in the number of layers, thickness, and mitotic rate of vaginal epithelium of T-treated mice compared with mice treated with T + 4-HA, T + 6-MP, or T + 4-HA + 6-MP. The present results indicate that development of PF and persistent anovulation are due to the direct action of estrogen (E) derived from T upon neonatal ovarian follicles and the neonatal hypothalamo-hypophysial system, and that T itself can induce ovary-independent vaginal changes, although 5 alpha-reduced androgen and estrogen derived from T seem to be more effective in this regard.

A comparative study of the sugar composition of lipopolysaccharides isolated from Vibrio cholerae, 'Vibrio albensis' and Vibrio metschnikovii.

A comparative study was made of the quantitative sugar composition of lipopolysaccharides (LPS) isolated from Vibrio cholerae (O1 and non O1 groups), 'V. albensis', 'V. proteus' and V. metschnikovii. The amino sugars 4-amino-4,6-dideoxy-D-mannose (perosamine) and 2-amino-2,6-dideoxy-D-glucose (quinovosamine) were present exclusively in LPS isolated from S-form O1 group of V. cholerae regardless of serotype (i.e. Ogawa or Inaba) and biotype (i.e. classical or eltor). Classical O1 group V. cholerae was distinguishable from eltor O1 group V. cholerae on the basis of the fructose content of the LPS: greater than 3% and less than or equal to 1%, respectively. Distinct differences in the sugar composition of LPS were observed between V. cholerae and 'V. albensis', 'V. proteus' and V. metschnikovii.

[Estimation of 1-hexylcarbamoyl-5-fluorouracil (HCFU) and 5-FU concentrations in sera and resected specimens from patients with ovarian tumors].

Concentrations of HCFU and 5-FU in blood and tissues of resected specimens were evaluated in 28 patients with ovarian tumor (pathologically benign 10, malignant 18) preoperatively considered to be malignant. 1) No significant difference of HCFU and 5-FU concentrations in sera, uterus and peritoneum was observed between the benign and cancer groups. 2) No significant difference of HCFU concentration in tissues of ovarian tumor was recognized between the benign and malignant groups, but as for 5-FU concentrations, 0.116 +/- 0.198 micrograms/g tissue in the cancer group was considerably higher than 0.021 +/- 0.144 micrograms/g tissue in the benign group (p = 0.085). Furthermore, the 5-FU level in ovarian cancer tissues was much higher than that in the tissues of the contralateral normal ovary, suggesting that degradation of 5-FU might be delayed in cancer tissue. 3) HCFU concentration in omental tissues from cancer patients was 0.043 +/- 0.061 micrograms/g tissue, significantly higher than that from benign patients, 0.004 +/- 0.008 micrograms/g tissue (p = 0.024). As for 5-FU concentration, no apparent difference was found, suggesting that 5-FU might be quickly eluted or metabolized in omental tissue.

[Augmentative effects of radiofrequency hyperthermia (RFH) combined with intraperitoneal (IP) administration of CDDP in ovarian cancer].

Radiofrequency hyperthermia (RFH) was applied to 21 adnexal cancer patients in combination with IP administration of CDDP (150 to 200 mg) 2 weeks or more after surgery. The instrument used, an OMR-ON HEH 500C, was shown by rectal temperature monitoring to produce sufficient intraperitoneal temperature elevation up to 40 degrees-41 degrees C. The serum level of filterable CDDP after IP infusion of CDDP (150 mg) followed by sodium thiosulfate rescue was basically the same irrespective of whether RFH was used. It reached a peak level of 2.06 +/- 0.48 micrograms/ml (n = 8) 1h after IP infusion, and gradually declined thereafter. No serious side effect except 3 cases of II- to III-degree burns was recognized. A slight increase in the incidence of prolonged vomiting and diarrhea was found with the RFH combination. This suggests an exacerbating effect of RFH on intestinal damage following CDDP infusion. As for any suppressive effect on reaccumulation of ascites, RFH might have a slight benefit in increasing the antitumor effect of CDDP in 90% of patients.

An electron microscopic study of early pathology in chondromalacia of the patella.

For a study of the ultrastructural pathology of chondromalacia patellae, we studied biopsy specimens that had been obtained at operation in twelve young patients who had pain in the anterior aspect of the knee due to mechanical derangement and also had the pathological changes in articular cartilage that are characteristic of chondromalacia. The initial pathological finding was swelling of the superficial matrix associated with breakdown of the collagen fiber network, especially at matrix streaks in the superficial and transitional zones. Fibrillated cartilage contained surface fissures that penetrated the middle layers. Amorphous electron-dense material was found covering the internal surfaces of the fissures; this appeared to contain degraded components of the matrix. In association with these changes, there were formation of nests of cells (clusters of chondrocytes) and increased numbers of organelles in the chondrocytes. A limited repair reaction also was observed in some specimens; this was characterized by the migration of fibroblast-like cells over the surface of the cartilage. The ultrastructural observations on these specimens from patients who had chondromalacia were compatible with a pathogenesis resulting from mechanical overload.

Growth of mouse vaginal epithelial cells in culture: effect of sera and supplemented serum-free media.

Normal mouse vaginal epithelial cells isolated from ovariectomized ca. 35-d-old BALB/cCrgl mice were grown in primary culture using collagen gel matrix and a serum-free medium composed of a 1:1 mixture of Dulbecco's modified Eagle's medium and Ham's F12 (D:H) medium supplemented with insulin (IN), epidermal growth factor (EGF), cholera toxin, transferrin, and bovine serum albumin V (BSA). Three-dimensional cellular outgrowths occurred inside the collagen gel matrix. The contribution of each factor to cell growth was examined by individual addition to the basic D:H medium and by individual deletion from the complete serum-free medium. When added individually, only IN promoted growth. Deletion of IN from the complete serum-free medium markedly diminished growth; deletion of EGF or BSA slightly diminished growth. When horse, fetal bovine, or chicken serum was added to the basal D:H medium, only with increasing doses of horse serum was there enhanced cell growth. The effect of 17 beta-estradiol and diethylstilbestrol on the growth of cells was also tested, using a suboptimal medium of D:H supplemented with BSA and IN, or a minimal medium supplemented with IN alone. During the 8-d time period, addition of estrogen did not enhance cell growth in either medium. To date, we have been unable to demonstrate a mitogenic effect of estrogen; rather a dose-dependent inhibition of proliferation is seen.

Growth of mouse vaginal epithelial cells in culture: functional integrity of the estrogen receptor system and failure of estrogen to induce proliferation.

Normal mouse vaginal epithelial cells isolated from ovariectomized ca. 40-day-old BALB/cCrg1 mice were purified by Percoll density gradient centrifugation and grown in primary culture using a collagen gel matrix and a serum-free complete medium. During the 9-day culture period, a 6-fold increase in cell number was observed. Addition of estrogen to the medium did not enhance epithelial cell proliferation. In fact, all doses of estrogen (180 fM to 18 nM) were inhibitory, resulting in only a 3- to 4-fold increase in cell number by day 9 of culture. Continuous exposure to estradiol (1.8 nM) for 9 days in the serum-free complete medium resulted in a decrease in cytosolic estrogen receptors with associated nuclear accumulation of estrogen receptors. A corresponding increase of cytosolic progestin receptors was also observed, indicating that no qualitative modification of the estrogen receptor system had occurred. Thus, despite its effectiveness in specific product synthesis (progestin receptors), estrogen does not stimulate proliferation of vaginal cells in this culture system, but rather inhibits epithelial cell proliferation.

Postnatal development of uterine abnormalities in mice exposed to DES in utero.

Postnatal development of uterine abnormalities was examined in the offspring of mice given 4 daily subcutaneous injections of 2,000 micrograms diethylstilbestrol (DES) and of oil vehicle alone from days 15 to 18 of gestation. The prenatally DES-exposed mice (DES mice) exhibited a disorganization of the inner circular musculature of the uterus as early as at 5 days of age. Invaginations of the luminal epithelium into the stroma which began to form in 5-day-old control and 10-day-old DES mice developed into uterine glands at 10 and 15 days, respectively. The number of glands per uterine section was significantly reduced in DES mice as compared with the controls at 10-30 days. Stratification with or without squamous metaplasia (ST) was found in the luminal epithelium of DES mice at 20 days. Five groups each of DES mice and the controls ovariectomized (OVX) at 10 days were given 5 daily subcutaneous injections of 10(-4), 10(-3), 10(-2), 10(-1), and 1 microgram 17 beta-estradiol (E2) starting at 25 days. ST was never observed in OVX mice given prepubertal injections of any E2 dose, whereas in DES OVX mice aged 30 days, ST was produced by prepubertal injections of E2 doses higher than 10(-2) micrograms/day. Uterine responsiveness, weight gain and gland formation were significantly reduced in DES mice given prepubertal E2 injections compared to the controls.

Pathways to chronic inflammation in rheumatoid synovitis.

Postcapillary venules resembling the high endothelial venules (HEVs) of lymphoid tissues have often been observed at sites of chronic inflammation. We have therefore postulated that such venules may be an important site of lymphocyte migration into rheumatoid synovial membrane and that inflammatory cell products may act on endothelial cells (ECs) to increase lymphocyte emigration. Electron microscopic examination of rheumatoid synovial membranes showed that a strong correlation existed between the proportion of lymphocytes in perivascular tissue and the height/base ratio of the ECs in those areas. In addition, binding experiments showed that peripheral blood mononuclear cells preferentially bound to ECs in sections of rheumatoid synovial membrane that had the morphological appearance of HEVs. In vitro binding experiments, in which lymphocyte adhesion to human umbilical vein EC monolayers was measured, showed that adhesion was enhanced by preincubation of the ECs with interferon-gamma or interleukin 1 (IL 1). The central role of IL 1 in increasing lymphocyte migration into the rheumatoid synovial membrane was also supported by the findings that IL 1 is chemotactic for lymphocytes, ECs can secrete IL 1, and IL 1 activity is readily detectable in synovial fluids of rheumatoid arthritis patients.

[Side effects and anti-cancer effects following intraperitoneal (ip) administration of mitomycin C (MMC)].

Nineteen patients with gynecological cancers were initially treated intraperitoneally with MMC at a dose of 20 to 52 mg/body and then intravenously with CAP regimen. The following side effects were observed in patients intraperitoneally administered doses of MMC greater than 25 mg/m2; leucocytopenia 7/12, thrombocytopenia 5/12, liver damage 8/12, renal damage 2/12, ileus 3/12 and severe anorexia 2/12. Sixteen patients were evaluated for their prognoses more than 1 year after treatment. Seven of them are well with no sign of recurrence, but 9 suffered recurrence with or without peritonitis carcinomatosa (PC). The incidence of PC among these 9 cases was not linked with clinical staging, complete or incomplete resection or the dose of MMC administered, but rather with the respective histopathology; mesonephroid 2/2, metastatic 2/2, mucinous 1/1, serous 0/3, endometrial corpus 0/1. The results suggested that the former 3 kinds of cancer did not respond to i.p. infusion of MMC, whereas the latter 2 were sufficiently sensitive to MMC to prevent the occurrence of PC.

Electron microscopic study of rheumatoid synovial vasculature: intimate relationship between tall endothelium and lymphoid aggregation.

A relationship has been demonstrated between the tallness of the endothelial cells (EC) of the postcapillary venules (PCV) of rheumatoid (RA) synovial membrane and the cellular composition of adjacent perivascular mononuclear cell infiltrates. A strong positive correlation between EC tallness and the number and percent of perivascular lymphocytes was observed. In contrast, EC tallness showed negative correlations with percent perivascular plasma cells, macrophages and fibroblasts. None of the preceding correlations were observed in the case of pericapillary infiltrates. The findings obtained indicate that the PCVs of the RA synovial membrane, from which lymphocytes emigrate to form perivascular lymphoid aggregates, resemble the tall endothelial PCVs of normal lymphoid tissue. They suggest that chronic inflammatory tissue and normal lymphoid tissue share common mechanisms of lymphoid emigration.

Occurrence of genital tract abnormalities and bladder hernia in female mice exposed neonatally to tamoxifen.

Three groups of female C57BL/Tw mice given 5 daily injections of 2, 20 or 100 micrograms tamoxifen (Tx) starting on the day of birth were killed at 35 and 150 days of age. About a half of the mice killed at 150 days had been ovariectomized at 90 days. Uterine hypoplasia, myometrial involution and suppression of the uterine-gland genesis were found in the 2 age-groups of Tx-treated mice. Vaginal hypoplasia and hypospadia were common abnormalities in 150-day-old Tx-treated mice. Vaginal adenosis was encountered in 35-day-old mice treated neonatally with 20 or 100 micrograms Tx, but not in 150-day-old group. Permanent proliferation of vaginal epithelium was not induced by Tx. More than 80% of oocytes in small follicles were degenerated in Tx-exposed mice at 150 days, but not so in those at 35 days. Ovaries of neonatally Tx-exposed mice lacking corpora lutea made no luteinizing response to human chorionic gonadotropin injected prepubertally. Urinary-bladder hernia with or without caecum hernia frequently occurred in 150-day-old mice given 20 or 100 micrograms Tx. The present study revealed that neonatally administered Tx causes various abnormalities in gonad and genito-urinary tract of female C57BL/Tw mice.

Electron microscopic study of HLA-DR and monocyte/macrophage staining cells in the rheumatoid synovial membrane.

We examined synovial membrane samples from 6 rheumatoid arthritis (RA) and 3 osteoarthritis patients and from 1 normal subject, by an immunoelectron microscopic technique using anti-HLA-DR (anti-Ia) and anti-monocyte/macrophage (63D3) monoclonal antibodies. In the lining layer, the type A macrophage-like cells were strongly DR+ and 63D3+, whereas the type B fibroblast-like cells were almost completely negative. Lymphocyte-rich areas (containing more than 90% densely packed lymphocytes) showed weak and patchy DR staining of the lymphocytes. In these areas, 3-5% of the cells were macrophage-like cells which were 63D3-, a type of staining compatible with that of the interdigitating cell (IDC). In the plasma cell-containing (transitional) areas, many strongly DR+ macrophage-like cells were observed in close contact with lymphocytes and plasma cells. Ten to twenty percent of these cells were 63D3-, which suggests that they were IDC. Cells with the structural appearance of IDC were most frequently seen in those transitional areas which contained elevated concentrations (50-70%) of lymphocytes. In uninfiltrated interstitial areas, approximately 50% of the cells stained strongly with both anti-DR and 63D3 antibody, indicating that they were cells of monocyte/macrophage lineage, presumably histiocytes. This investigation has demonstrated the presence of the DR antigen in the RA synovial membrane on 1) phagocytic cells of the lining area, 2) lymphocytes and small numbers of IDC-like cells in dense, lymphocyte-rich areas, 3) large numbers of macrophage-like cells, of which some had the morphologic appearance of IDC, in transitional or plasma cell-containing areas, and 4) histiocytic cells in uninfiltrated interstitial areas. The observation of large numbers of DR+ macrophages and IDC-like cells in close contact with lymphocytes and plasma cells in the RA synovial membrane emphasizes their role in an active immune response. The observation of substantial numbers of potentially immunocompetent, DR+ histiocytic cells in uninfiltrated regions of the synovial membrane suggests that such cells may play a role in the progression of the synovial inflammatory reaction.
