Chondroitin sulfate isomers (ChS isomers) were digested with chondroitinase ABC, and the unsaturated disaccharides produced were converted into fluorescent dansylhydrazine derivatives and analyzed by HPLC. The proposed method was applied to the determination of relative amounts of ChS isomers in normal human urine and analytical results showed that chondroitin-6-sulfate and chondroitin-4-sulfate were excreted in large amounts. Moreover, this method was used for the identification of urinary ChS isomers which were extracted from a cellulose acetate strip after carrying out electrophoresis.
A retrospective study was made of 433 patients with colorectal cancer who were operated on between January, 1971 and September, 1986. Two-hundred and forty-eight synchronous polyps (226 adenomas and 22 carcinomas) were found in 128 patients (29.6%). The incidence of polyps varied according to the patient's age, sex, family history of colorectal cancer, and also according to the macroscopic classification, depth of infiltration, and the number of the primary tumors. Of these, macroscopic classification seemed to be the most important factor determined by multivariate analysis. The incidence of synchronous polyps of the protuberant type were the highest (50%), as compared to the ulcerating type (28%) and the infiltrating type (19%) (p less than 0.03).
A simple and accurate HPLC method for the determination of orthophosphate in the presence of large amounts of organophosphates is described. The method is based on the formation and separation of the molybdenum orthophosphate complex. In order to prevent the hydrolysis of organophosphates, the sample was deproteinized with silicotungstate in acetate buffer (pH 4.0) under ice-cooling and then treated with ammonium molybdate in maleate buffer (pH 7.0). The sample was injected onto Styragel 60 A column (5 mm ID x 100 mm) with 38% (v/v) acetonitrile containing 0.3 M sulfuric acid as eluent. Detection was at 310 nm. The method was applied to the determination of orthophosphate in liver, kidney, spleen and mouse blood.
A method for the determination of cyanide and thiocyanate in blood plasma and red cells of humans was established. It involved high-performance liquid chromatography and fluorometric detection by the König reaction. Calibration curves for cyanide and thiocyanate were linear in the range 1-200 pmol and 2-300 pmol, respectively. Clean-up methods for the determination of cyanide and thiocyanate in red cells were also developed. These methods were applied for the determination of cyanide and thiocyanate in the blood of smokers and non-smokers.

Two radiochemical esterolytic assays for urokinase are described. One assay is based on the urokinase-dependent hydrolysis of Nalpha-acetyl-glycyl-L-lysine [3H]methyl ester and the other on the urokinase-dependent activation of plasminogen and assay of generated plasmin with Nalpha-tosyl-L-arginine [3H]methyl ester. The assays are performed in tubes placed in liquid scintillation counting vials. At the end of the experiment generated [3H]methanol is extracted into the liquid scintillation cocktail and counted. Unhydrolyzed substrate largely remains in the aqueous phase and contributes only a small fraction of the counts. This facile separation of 3H-labeled alcohol from the ester substrate allows the simple and highly sensitive assay for urokinase. The assays give results in good agreement with the classical fibrin plate assay.
