Relationships between milk levels of hormones and growth or puberty of offspring in mice.

Milk levels of prolactin, growth hormone (GH) or progesterone on days 12-13 of lactation and their relationships with the growth or the puberty of offspring were examined in primiparous SHN mice. Milk prolactin level had negative correlations with the body weights on days 12 and 20, the growth rate between days 0 and 20 or the age and the body weight at vaginal opening of offspring. There was a significant correlation between milk GH level and the growth rate between days 0 and 12 of offspring. A positive correlation was also found between milk progesterone level and the age at vaginal opening. These results suggest that hormones in milk would have effects on the growth or the puberty of offspring.

Mephenytoin hydroxylation in the Cuna Amerindians of Panama.

1 Mephenytoin p(4')-hydroxylation, which is deficient in 3-5% of Caucasians, was examined in 96 Cuna Amerindians of Panama. 2 Attempts were made to exclude poor compliance with urine collection and ingestion of the drug dose since the assignment of phenotype was based upon urinary recovery of the metabolite. These involved the measurement of the urinary recovery of sparteine, added to the ingested capsule, and of the renal excretion of creatinine. 3 Of the 90 Cunas deemed to be reasonably complaint, none of them appeared to be deficient in p(4')-hydroxylation of mephenytoin.

Augmentation of the response of mouse uterine epithelial cells to estradiol by uterine stroma.

The effect of estrogen on the in vitro growth of mouse uterine epithelial cells was assessed. Epithelial cells from the immature mouse uterus were successfully cultivated in a 1:1 mixture of Dulbecco's Modified Eagle's Medium and Ham's F-12 supplemented with insulin (5 micrograms/ml), transferrin (10 micrograms/ml), hydrocortisone (10(-7) M), BSA (2 mg/ml), and fetuin (1 mg/ml). Addition of 17 beta-estradiol in the range of 1-100 nM did not significantly change the total DNA content of the epithelial cells. A binding component of [3H]estradiol by cultured uterine cells was shown to be specific, saturable, and of high affinity. Kd values for specific binding by epithelial and stromal cells were 1.0-1.7 x 10(-10) M. Maximal specific binding was 0.74 and 2.3 fmol/micrograms DNA for epithelial and stromal cells, respectively. Treatment of epithelial and stromal cells for 4 days with 10 nM estradiol led to a 2- to 6-fold increase in progesterone receptor concentration. Treatment of epithelial and stromal cells in mixed culture for 4 days with 10 nM estradiol resulted in a significant increase in total DNA. That epithelial-stromal contact was critical for estradiol stimulation was shown by the fact that if the cell types were separated into two compartments which still allowed free media mixing, total DNA was not enhanced by estradiol. These observations are organized into a model for mitogenic action of estradiol that seems to reconcile observed disparities in the action of the hormone in vivo and in vitro.

Accuracy of digital palpation for size assessment of the prostate evaluated by transrectal sonography.

In order to clarify how precisely digital palpation gives information on prostatic size, evaluation of the size measurement of the prostate by means of transrectal sonography was performed on 1,543 subjects with various prostatic disorders. There was little agreement between the assessment of prostatic size by digital palpation and the ultrasonic measurement. The larger the prostate the less accurate was digital palpation. The prostatic size assessed by digital palpation was particularly dispersed widely in the prostate of 'hen's egg' size. It was proved that the size assessment by digital palpation was only effective statistically in a two-step differentiation, 'hen's egg' size and smaller. More complicated size classification was of little clinical value in digital palpation.

Quantitative measurement of renal function using Ga-68-EDTA.

We studied renal function using a positron emission tomography with C15O gas and 68Ga-EDTA in 6 normal subjects (male, 21-77 years old). The blood volume (ml/100 g (kidney] was 19.2 +/- 1.8 for the one 21 years old subject and 11.8 +/- 2.0 for the rest (65-77 years old). The glomerular filtration rate (ml/min/100 g (kidney] was 57.8 +/- 1.1 for the 21 years old subject and 30.4 +/- 2.8 for the rest.

Cytogenetic findings and clinical features in acute leukemia and transient myeloproliferative disorder in Down's syndrome.

Cytogenetic, immunologic, and electron microscopic studies were performed on the blast cells of 28 pediatric patients with Down's syndrome, 13 with acute leukemia (DS-AL) and 15 with transient myeloproliferative disorders (DS-TMD). Clonal chromosome abnormalities were found in the cells of all patients with DS-AL but not those with DS-TMD. The younger ages and higher hemoglobin concentrations, platelet counts, and WBC counts of DS-TMD patients provided a clinical contrast with the frankly leukemic cases. Myelodysplastic syndrome, characterized by a small percentage of leukemic blast cells, was observed in 11 of the 13 patients with DS-AL compared with none in the DS-TMD group. Electron microscopy disclosed a positive platelet peroxidase reaction in each of the 11 DS-TMD patients and in nine of the 13 DS-AL patients. Immunologic studies revealed antiplatelet-megakaryocyte antigens on the blast cells of the majority of patients in both study groups. Our findings suggest that the blast cells in cases of DS-AL and DS-TMD arise from cells of the megakaryocytic lineage or from a myeloid progenitor with the capacity for megakaryocytic differentiation. The high risk of the development of AL in patients with DS who are less than 3 years old may be related to increased megakaryocyte proliferation in this age group.

[Percutaneous nephroureterolithotomy in two city hospitals: review of 102 cases].

The authors experienced 102 cases of percutaneous nephroureterolithotomy (PNL) in the urological clinic at two city hospitals between May, 1984, and July, 1986. Percutaneous access for renal pelvis was performed under interventional ultrasound and the disintegration of stones was mainly performed by an electric drill system. Stones were successfully removed in all the cases, although residual stones were recognized in 16 cases (15.7%). Complications were rather minor and were treated conservatively without open surgery. PNL is a very effective and promising method not only for university hospitals but also for the urological clinic in city hospitals.

Kinetic evaluation of the effect of LHRH analog on prostatic cancer using transrectal ultrasonotomography.

The size of the prostate was measured in 11 patients with the prostatic cancer by means of transrectal ultrasonotomography after LHRH analog ([Ser(Bu)6] LHRH) treatment. Prostate volume changed parallel to serum testosterone (T) level and decreased by 17-62% compared with baseline after LHRH analog treatment for 4 months. Two different patterns were observed for the change in prostatic volume. Type A expanded transiently and then decreased gradually, while type B decreased continuously from the beginning with no expansion. After treatment for 4 months, the prostatic volume of type A was significantly less than that of type B. It is suggested that prostatic cancer showing a type A regression curve may be more sensitive to T, resulting in a better prognosis than type B.

On phenotyping with isoniazid: the use of urinary acetylation ratio and the uniqueness of antimodes. Study of two Amerindian populations.

Various conventions have been used to express the activity of polymorphic hepatic N-acetyltransferase of isoniazid. Among them, two of the most common are the "percentage of acetylisoniazid" and the "inactivation index." A third alternative convention is proposed, the "molar acetylation ratio," which showed the most clear-cut bimodal distribution when applied to the results obtained for Cuna and Teribe Amerindians living in Panama. Through this method a unique antimode was assigned to each Amerindian group and the same frequency of slow acetylators (24% to 29%) was found, unlike the results obtained by conventional approaches and antimodes derived from white populations.
