Evidence for a Na+/H+ antiport in stomach smooth muscle cells.

Single smooth muscle cells were isolated from circular muscle of the canine gastric corpus by collagenase incubation. Cytoplasmic pH (pHi) of these cells was measured fluorometrically using the trapped dye 2',7'-bis(carboxyethyl)-5,6-carboxyfluorescein. Cells were examined for their Na+/H+ exchange activity after intracellular acidification. Cells acid-loaded by propionate exposure, the NH4+ prepulse technique or suspension in a Na+-depleted medium regained almost normal pHi upon exposure to a Na+ medium. The Na+-dependent alkalinization was amiloride sensitive. As well, addition of amiloride to cells suspended in a Na+ medium caused a concurrent decrease in pHi. The study indicates that a Na+/H+ antiport is present in these smooth muscle cells.

Age differences in integrated semantic memory for abstract sentences.

The Bransford and Franks [4] paradigm of linquistic abstraction was used to examine age differences in the nature of stored semantic information. Twenty-eight young (mean age 18.9 years) and 27 old (mean age 70.8 years) adults served as subjects. Specifically, age differences on two dimensions of memory were examined: (a) integration of related content from separate sentences and (b) retention of precise semantic content. Young and old were found to differ in the precision of retained semantic content. However, both age groups evidenced integration of information as indexed by a strong linear trend of recognition rate across sentence complexity and this trend did not interact with age, supporting the idea that both age groups showed comparable integration of abstract linguistic information into holistic ideas. The implications of these findings are considered in relation to current conceptualizations of age differences in memory, and new directions for research are proposed.

Relaxation of coronary artery smooth muscle cells.

The effect of adenosine and nitroglycerine on single smooth muscle cells isolated from the right coronary artery of the pig is reported. Trypan Blue exclusion and electron microscopy revealed that the cells used in this study were structurally intact. These cells were 79.2 +/- 1.6 microns long. Within 1 min of exposure to solutions containing norepinephrine, carbachol and high KCl, the cells contracted by 20-30%. The concentrations required for half maximal contraction for norepinephrine, carbachol and KCl were 0.1 to 1 nM, 0.01 to 0.1 nM and 15 to 25 mM, respectively. Adenosine and nitroglycerine inhibited the contractions caused by high (30 mM) K+ and also relaxed the cells previously contracted by high K+. Thus adenosine and nitroglycerine relax by acting directly on the smooth muscle cells.
