DRB3.2 gene polymorphism and its association with pashmina production in Changthangi goat.

PstI and TaqI restriction fragment length polymorphism (RFLP) of caprine DRB3.2 revealed three genotypes under each RFLP in Changthangi goat. The genotypic and allelic frequency in PstI RFLP was calculated as 0.07, 0.72 and 0.20 for PP, Pp and pp genotypes and 0.43 and 0.57 for P and p alleles. In TaqI RFLP the frequencies were observed as 0.11, 0.61 and 0.27 for TT, Tt and tt genotypes and 0.41, 0.59 for T and t alleles. Alignment study indicates variability of nucleotides and amino acids between alleles and different breeds of goats. The nucleotide sequences reported in this paper have been submitted to the European Molecular Biology Laboratory GenBank and the accession numbers were AY496935, AY496061 and AY496062. A significant affect (P <or= 0.05) of MHC DRB3.2 PstI polymorphism was found on the first and second year of pashmina production.

Impact of Ni(II), Zn(II) and Cd(II) on biogassification of potato waste.

A study was conducted on anaerobic digestion of potato waste and cattle manure mixture, inoculated with 12% inoculum and diluted to 1:1 substrate water ratio at 37 +/- 1 degrees C. Initially pH of substrate was found to be 4.5 to 5.0. Lime and sodium bicarbonate solutions were employed to adjust the pH to 7.5. Biogas production continued up to 10 and 7 days, when lime and sodium bicarbonate solutions were used to adjust the pH, respectively. Biogassification potential was studied in response to different ratio of waste and cattle manure. Biogas production rate was higher when potato waste and cattle manure were used in 50:50 ratio. Effect of two different concentrations (2.5 and 5.0 ppm) of three heavy metals viz. (Ni (II), Zn (II) and Cd (II)) on anaerobic digestion of substrate (potato waste--cattle manure, 50:50) was studied. At 2.5 ppm, all the three heavy metals increased biogas production rate over the control value. The percentage increase in biogas production over the control was highest by Cd, followed by Ni and Zn. In all the treatments, methane content of biogas increased with increase in time after feeding. Various physico-chemical parameters viz. total solids, total volatile solids, total organic carbon and chemical oxygen demand considerably declined after 7 days of digestion and decline was greater in presence of heavy metals as compared to control. The physico-chemical parameters revealed maximum decrease in the presence of 2.5-ppm concentrations of heavy metals with the substrate. Among all the three heavy metals employed in the study, Cd++ at 2.5 ppm was found to produce maximum biogas production rate. The use of three heavy metals to enhance biogas production from potato and other horticultural waste is discussed.

Genetic polymorphism of alpha-lactalbumin gene in riverine buffalo.

Alpha-lactalbumin (alpha-LA) is a major whey protein found in milk. Polymorphs of alpha-LA gene are reported to be significantly associated with milk production and constituent traits. Therefore, the present study was undertaken to detect polymorphism in alpha-LA at the genic level and to explore allelic variability at this locus. A total of 196 animals, belonging to four breeds of riverine buffalo viz. Bhadwari, Mehsana, Surti and Murrah were included under the present investigation. Two fragments i.e. 133 bp (Exon 1) and 159 bp (Exon 2) of alpha-LA gene were amplified by polymerase chain reaction and subsequently, single strand confirmation polymorphism (SSCP) study was carried out to identify different allelic pattern and genotypes of the animal included in the study. Both fragment of alpha-LA gene was found to be polymorphic in all the four breeds of riverine buffalo. Number of genotypes and allele varied breed to breed for both the fragments. In case of 133 bp fragment, four alleles A, B, C and D were found among different breeds of buffalo whereas in 159 bp fragment, five alleles namely A, B, C, D and E was found in different breeds. Nucleotide sequence data of different alleles showed the presence of both silent as well as functional mutation leading to variability in polypeptide chain.

Studies on the relationships between the synonymous codon usage and protein secondary structural units.

The relationship between the synonymous codon usage and protein secondary structural elements (alpha helices and beta sheets) were reinvestigated by taking structural information of proteins from Protein Data Bank (PDB) and their corresponding mRNA sequences from GenBank for four different organisms E. coli, B. subtilis, S. cerevisiae, and Homo sapiens. It was observed that synonymous codon families have non-random codon usage, but there does not exist any species invariant universal correlation between the synonymous codon usage and protein secondary structural elements. The secondary structural units of proteins can be distinguished from the occurrences of bases at the second codon position.

Role of the cholinergic system in the modulation of ventricular arrhythmias induced by subepicardial epinephrine in the dog.

The modulating role of the cholinergic system in epinephrine (EP)-induced ventricular arrhythmias (VAs) was studied in anesthetized (alpha-chloralose and pentobarbitone sodium) dogs. Unifocal VAs were produced by subepicardial infusion of EP (4 X 10(-3)M) in 2.5 X 10(-3) M CaCl2-0.9% NaCl at a rate of 10 microliter/min for a maximum period of 10 min at a time. One of the interacting drugs acetylcholine Cl (ACh), carbachol Cl (CCh), physostigmine salicylate (PHY), DL-propranolol HC1 (PROP), quindine HC1 (QD), lidocaine HC1 (LD), and atropine sulfate (AT) was given in an equimolar concentration (4 X 10(-3)M) along with EP. The effects of bilateral vagotomy (VT) were also studied. The time of onset, frequency, and duration of VAs were recorded. Reproducible cognizable VAs (less than 10%) were produced by EP in 38 of 43 experimental dogs. Focal infusion of the cholinomimetic agent ACh or CCh marketedly inhibited the arrhythmogenic activity of EP at the focal site. The effect of the anticholinesterase agent PHY was similar. These experiments showed that exogenous administration of a cholinomimetic agent or focal facilitation of endogenous cholinergic influence had an antiarrhythmic activity against EP. On the other hand, bilateral VT or focal infusion of AT markedly potentiated the arrhythmogenic potentiality of EP. Thus, it appeared that removal of endogenous influence facilitated EP-induced arrhythmias. The antiarrhythmic activities of the beta-adrenoceptor-blocking agent PROP and the membrane-stabilizing agents LD and QD against EP were confirmed using the subepicardial infusion technique. These studies show that the presence of cardiovagal tone or its facilitation inhibits, and its blockade increases, the propensity to VAs in situations in which the adrenergic system may be involved primarily or otherwise. The receptors involved appear to be muscarinic in nature.

Variability of milk fat globule membrane protein gene between cattle and riverine buffalo.

A study on butyrophilin (BTN) gene was conducted to detect variability at nucleotide level between cattle and buffalo. Hae III PCR-RFLP was carried out in crossbred cattle and it revealed polymorphism at this locus. Three genotypes namely, AA, BB and AB and two alleles were observed with frequencies 0.78, 0.17, 0.04 and 0.87, 0.13, respectively. The sequences of different cattle, buffalo and sheep breeds have been reported in the EMBL gene bank with accession numbers: AY491468 to AY491475. The nucleotides, which have been substituted from allele A to B, were found to be C to G (71st nucleotide), C to T (86th nucleotide), A to T (217th nucleotide), G to A (258th nucleotide), A to C (371st nucleotide) and C to T (377th nucleotide). The nucleotide substitution at 71st, 86th and 377th position of the fragment were expected to be a silent mutation where as nucleotide changes at 217th, 258th and 371st positions were expected to be substituted by lysine with arginine, valine with isoleucine and leucine with proline in allele B. The differences of nucleotides and amino acids between cattle, buffalo and sheep breeds have been revealed and on the basis of nucleotide as well as protein variability the phylogenetic diagram have been developed indicating closeness between cattle and buffalo.
