Microbial mutation studies with tetrachlorvinphos (Gardona)).

The mutagenic activity of tetrachlorvinphos was investigated in agar-layer cultures of Escherichia coli WP2 and WP2 uvrA, Salmonella typhimurium TA1535, TA1538, TA98 and TA100. Assays were carried out both in the presence and in the absence of S9 fractions of liver homogenates from rats and mice, both from untreated animals, and from animals pre-treated with Aroclor 1254. The induction of mitotic gene conversion by tetrachlorvinphos was studied in stationary phase cultures of the yeast Saccharomyces cerevisiae D4. No mutagenic effects, as determined by reverse gene mutation, were detected in vitro in the bacterial/mammalian microsome assay when a range of bacterial tester strains were exposed to tetrachlorvinphos at amounts up to 2000 micrograms per plate, either in the absence or in the presence of S9 fractions from non-induced or Aroclor-induced mouse or rat livers. Tetrachlorvinphos did not increase the mitotic gene conversion frequency in stationary phase cultures of the yeast, Saccharomyces cerevisiae D4.

Genetic toxicology testing of 41 industrial chemicals.

41 compounds or mixtures of diverse structure and application have been tested for genotoxic activity. The materials were tested in bacterial mutation assays, in Saccharomyces cerevisiae JD1 for mitotic gene conversion and in a cultured rat-liver cell line for structural chromosome damage. 11 compounds were bacterial mutagens, 4 induced mitotic gene conversion in yeast and 5 were positive in the chromosome assay. 5 of the materials were positive in bacteria only and 2 compounds induced chromosome damage in cultured cells in the absence of mutation in bacteria or gene conversion in yeast. The materials were tested over a 5-year period and the performance and evolution of the 3 assays during this time is evaluated. The results are considered in relation to the structure of the chemicals and the genotoxicity of related compounds.

The induction of mitotic gene conversion in the yeast, Saccharomyces cerevisiae JD1 by 4-chloromethylbiphenyl (4CMB), benzyl chloride (BC) and 4-hydroxymethylbiphenyl (4HMB).

The induction of mitotic gene conversion by 4CMB, BC and 4HMB was studied in both log-phase and stationary-phase cultures of the yeast, Saccharomyces cerevisiae JD1. Assays were performed both in the presence and in the absence of S9 microsomal fraction obtained from a liver homogenate from rats pretreated with Aroclor 1254. Exposure of both stationary-phase and log-phase cultures to 4CMB and BC resulted in an increase in mitotic gene conversion, both in the presence and in the absence of a microsomal activation system; the magnitude of response was greater in stationary-phase cultures. 4HMB did not increase the gene conversion frequency in log-phase or stationary-phase cultures.
