Fortimicins A and B, new aminoglycoside antibiotics. I. Producing organism, fermentation and biological properties of fortimicins.

A culture of Micromonospora species MK-70 was found to produce two new antibiotics, fortimicins A and B. Antibacterial and paper chromatographic data on an eluate from IRC-50 treatment of fermentation beers indicated that fortimicins A and B are new antibiotics with broad-spectrum, basic and water-soluble properties. Fortimicin A exhibited potent, unique, broad-spectrum antibacterial activity against Gram-positive and negative bacteria both in vitro and in vivo, while fortimicin B was only weakly active.

Fortimicins A and B, new aminoglycoside antibiotics. II. Isolation, physico-chemical and chromatographic properties.

The aminoglycoside antibiotics fortimicins A and B produced by a naturally occurring strain Micromonospora sp. MK-70 were isolated from its fermentation beer. Fortimicins A and B were isolated as water-soluble, basic, white amorphous powders having molecular formula C17H35N5O6 and C15H32N4O5, respectively. Acid hydrolysis of fortimicin A indicated that it has one mole of glycine in its molecule while fortimicin B has not. Paper chromatography, silica-gel and carbon thin-layer chromatography revealed that fortimicins A and B are novel aminoglycoside antibiotics.

New antibiotic XK-62-2 (sagamicin). II Taxonomy of the producing organism, fermentative production and characterization of sagamicin.

Cultures of Micromonospora species, strains MK 65 and MK 62, were found to produce a new antibiotic XK-62-2 (Sagamicin). Antibacterial and paperchromatographic data on an eluate from IRC50 treatment of fermentation beers indicated that XK62-2 is a new antibiotic with broad spectrum, basic, and water-soluble properties, probably closely related to the gentamicin C group of antibiotics.

A rat model of postthoracotomy pain: behavioural and spinal cord NK-1 receptor assessment.

PURPOSE: To develop a new rat model of postthoracotomy pain for investigating its mechanisms and clarifying neurochemical changes. METHODS: Male Wistar rats were randomly assigned to three groups that underwent either fourth and fifth intercostal nerve ligation, cutting of the fourth and fifth ribs, or a sham operation in which only pleura was cut. For behavioural response assessment during the following month, pinch and touch were used as mechanical stimuli, and acetone was used as a cold thermal stimulus. In addition, (125)I-substance P autoradiography was used to determine neurokinin (NK) receptor density in spinal cord laminae I and II at one to six weeks after surgery. RESULTS: In rats with nerve ligation, hypersensitivity to noxious and non-noxious stimuli continued throughout the month. The "mirror phenomenon" was observed. The lowest threshold was obtained in the dorsomedial portion of the T4 dermatome on the side of surgery. In rats with rib cutting, a lowered threshold to noxious and non-noxious stimuli was observed for two weeks. In rats with sham operations, hypersensitivity was seen only at postoperative day one. NK-1 receptor density on the side of operation increased significantly in rats with nerve ligation from day seven to 28. Receptor density was highest on day 14 (22.97 +/- 1.04 fmol x mg(-1) tissue vs. control, 16.22 +/- 0.43), representing a 50% receptor excess on the side of ligation compared to the contralateral side. CONCLUSION: Intercostal nerve damage induces long-term postthoracotomy pain and an increase of spinal NK-1 receptors in rats. This model may be useful for investigation of postthoracotomy pain.

Morphometric development of the human fetal auditory system: inferior collicular nucleus.

The development of the human inferior collicular nucleus was studied on serial sections of the brains of 9 fetuses at 12-34 weeks of gestation and an adult of 63 years using an electronic planimeter with a computer. Morphometric analysis of the development of the inferior collicular nucleus showed that its development accelerates after 18 weeks of gestation in terms of the columnar volume and length, size of neurons and circularity ratio. Large-sized neurons 2-3 times the area of small-sized neurons appeared after 18 weeks of gestation. The ratio of large neurons to the total ranged between 3 and 6% in the inferior collicular nucleus. The development of the inferior collicular nucleus was similar to that of the ventral cochlear nucleus and the medial superior olivary nucleus.

Intravenous gammaglobulin therapy of Satoyoshi syndrome.

Satoyoshi syndrome is a very rare disease of unknown etiology, characterized by intermittent painful muscle spasms, alopecia, multiple epiphyseal changes, diarrhea and endocrine disorders. We administered intravenous gammaglobulin to a 7-year-old girl with Satoyoshi syndrome. Frequency of muscle spasms and the titers of antinuclear antibody and anti-DNA antibody decreased. This is the first report of gammaglobulin therapy of Satoyoshi syndrome. We suggest that this illness could be related to an autoimmune mechanism.
