Immunological studies on herpetic keratitis: effect of IL-2 on HSV-specific CTL.

The effect of interleukin 2 (IL-2) on cytotoxic T lymphocytes (CTL) was examined in mice with corneal infection of herpes simplex virus (HSV). The corneas of the mice were inoculated with HSV after scratching the corneal epithelium. Ten days after the inoculation, CTL were induced in vitro from spleen cells of the mice. IL-2 was added for the initiation of CTL induction. CTL induced without IL-2 served as controls. After the culture, viable cells were counted and cytotoxicity of CTL was measured by 3H-proline releasing assay. IL-2 promoted proliferation of HSV specific CTL without increasing the cytotoxicity of HSV specific CTL.

Immunosorbent consisting of DNA immobilized on oxirane-activated sepharose.

The preparation of adsorbents for DNA antibodies is described. The degree of immobilization of native DNA on Sepharoses activated with epichlorohydrin or bisoxirane was investigated as a function of pH, temperature, time, concentration of DNA, and oxirane content in the supports. The maximum amount of DNA bound was obtained after 8 h at 40-50 degrees C at pH 11-11.5. The amount bound was increased by raising either the concentration of DNA or the oxirane content of the supports, and could reach 300 mg/g dry support. The immobilized DNA was applied to the adsorption of DNA antibodies using either commercial human serum with anti-native DNA activity or the sera of patients with systemic lupus erythematosus. The amount of antibody adsorbed depended on the amount of DNA. The thermal stability of the immobilized DNA was also examined. After heating at 80 degrees C, the leakage of DNA was slight and the adsorption of antibodies was not affected.

Histological changes of muscle in a patient with pyruvate dehydrogenase deficiency.

Histological changes of muscle from a 17-month-old boy with pyruvate dehydrogenase deficiency are presented. The patient had muscle hypotonia, mental retardation, seizures, lactic acidosis and hyperalaninemia. Deficient activity of the pyruvate dehydrogenase complex was found in his platelets (about 25% of normal) and of pyruvate dehydrogenase in his biopsied muscle (about 5% of normal). A muscle biopsy specimen showed an increased proportion of type IIC fibers (24%), fiber-type grouping and lipid droplet accumulation.

Studies on the phenotyping system of hyperlipoproteinemias. Evaluation of a new method by enzymic staining of lipids in serum lipoproteins separated by electrophoresis on agarose.

A new electrophoresis of lipoproteins was examined as a means of defining hyperlipoproteinemia phenotypes. The serum lipoproteins of 119 patients with hyperlipoproteinemia whose serum total cholesterol (TC) concentration was more than 250 mg and/or whose triglyceride (TG) concentration exceeded 180 mg per 100 ml were determined by using a method of electrophoretic separation of lipoproteins on agarose and enzymic staining of each lipid component separately. Based on the lipoprotein-lipid profile with the acid of various parameters for distinction, i.e., Type II A, LDL-C greater than or equal to 180 mg/dl: Type IV, VLDL-TG greater than or equal to 130 mg/dl: type II B, LDL-C greater than or equal to 130 mg/dl, 119 patients were classified into Fredrickson's phenotypes of hyperlipoproteinemia. The percentages of phenotypes with those hyperlipoproteinemias were: type II A, 31.1%; II B, 33.6; IV, 29.4%; and V, 5.9% respectively. But types I and III were not observed in this survey. The characteristics of the lipoprotein profiles of each phenotype were clearly identified, and they were: type II A, high peak LDL-C and decrease of HDL-C fraction; II B, high peak of LDL-C and VLDL-TG; IV, high peak of VLDL-TG; V, prominence of chylomicrons and high VLDL-TG peak. The serial observations of the lipoprotein-lipid profiles were very useful for detecting the progressive changes in lipoproteins which occurred during the course of diseases, particularly in cases of secondary hyperlipoproteinemia. In conclusion, the new lipoprotein-lipid profile based on the agarose electrophoresis-enzymic staining method not only offers great help in classification of hyperlipoproteinemia but has contributed to therapeutic management.

Identification and characterization of a new variant of apolipoprotein E (apo E-Kochi).

A new variant of apolipoprotein E (apo E), named apo E-Kochi, was identified in the sera of a 29-year-old male with hyperlipoproteinemia as characterized by a broad-beta band. The characteristic double bands of apo E were seen in the isoelectric focusing gel of very low density lipoprotein from the proband and three members of his family. Of the double bands from the probands, the more cationic component was identical to ordinary apo E3 and the other anionic band was located at approximately a distance of one-half charge to the anode side. This anionic band is a new electrophoretical isoform of apo E (apo E-Kochi), and the molecular weight by sodium dodecyl sulfate electrophoresis and its antigenicity against anti apo E serum are the same as apo E3. Sequence analysis of lysyl endopeptidase fragments showed that apo E-Kochi differs from normal apo E3 at residue 145, where an arginine residue is substituted for histidine.
