The Sonic Hedgehog-Gli pathway regulates dorsal brain growth and tumorigenesis.

The mechanisms that regulate the growth of the brain remain unclear. We show that Sonic hedgehog (Shh) is expressed in a layer-specific manner in the perinatal mouse neocortex and tectum, whereas the Gli genes, which are targets and mediators of SHH signaling, are expressed in proliferative zones. In vitro and in vivo assays show that SHH is a mitogen for neocortical and tectal precursors and that it modulates cell proliferation in the dorsal brain. Together with its role in the cerebellum, our findings indicate that SHH signaling unexpectedly controls the development of the three major dorsal brain structures. We also show that a variety of primary human brain tumors and tumor lines consistently express the GLI genes and that cyclopamine, a SHH signaling inhibitor, inhibits the proliferation of tumor cells. Using the in vivo tadpole assay system, we further show that misexpression of GLI1 induces CNS hyperproliferation that depends on the activation of endogenous Gli1 function. SHH-GLI signaling thus modulates normal dorsal brain growth by controlling precursor proliferation, an evolutionarily important and plastic process that is deregulated in brain tumors.

[Super heavy oil produced water treatment by surface flow constructed wetland].

This paper adopts a new economic technology of treating super heavy oil produced water using surface flow reed wetland system. The field test was conducted at the average filtration rate of 3.33 cm/d. The average influent concentrations per year of COD, Oil, BOD5, TN were 459.16 mg/L, 27.65 mg/L, 33.52 mg/L, 13.74 mg/L, and the average effluent concentrations per year of above indices were 77.21 mg/L, 1.42 mg/L, 3.90 mg/L, 1.60 mg/L, respectively. Its respective removal ratios of COD, Oil, BOD5, TN and pH were 83.18%, 94.86%, 88.37%, 88.36%, 7.87-7.77. Analyses showed that super heavy oil produced water had no obvious effect on soil and almost no effect on growth volume and quality of reeds. Results indicated that the surface flow reed wetland could provide an efficient economical, stable new means of treating super heavy oil produced water.

[An off site petroleum-contaminated soil bioremediation technology: soil compositing in windrow].

With off-site bioremediation technology, a soil contaminated by crude oil from Liaohe Oil Field was treated on a 20 x 10 m prepared bed. 8 composting windrow units were set, each measured 8 m in length, 2 m in width, and 0.35 m in height. The results showed that when the pollutant petroleum hydrocarbon(TPH) was within the range of 4.16-7.72 g.100 g-1 soil, the total degradation rate of TPH reached 45.19%-56.74% after 53 days operation, which indicated that a technological basis would be provided for the bioremediation of oil-contaminated soil.

[mRNA and protein expression of three growth-related factors and their possible signal transduction pathways in wound healing].

OBJECTIVE: To observe the time course and tissue distribution of c-fos, c-myc and basic fibroblast growth factor(bFGF) genes and their proteins in burned skin, and explore the possible network regulation mechanisms of c-fos, c-myc and bFGF in wound healing and the role of the protein kinase C pathway involved in signal transduction among these growth-related factors. METHODS: The study was carried out on 188 Wistar rats. Animal model with 30% partial thickness scald injury was made and all rats were randomly divided into several groups: normal control (n = 8), injured control (n = 36), anti-c-fos (n = 36), bFGF (n = 36), 1-5-Isoquinolinyl-sulfony-2methylpiperazine (H7), and 2, 3-Butanedione monoxime (BDM) (n = 36). In situ hybridization, reverse transcription polymerase chain reaction (RT-PCR) and streptavidin peroxidase (SP) immunohistochemistry methods were used to detect the gene and protein expression of c-fos, c-myc and bFGF in the wounded tissues. RESULTS: (1) The first appeared induction of growth-related gene was c-fos, the second bFGF, and the last c-myc. They were peaked at 3 h, 1 d and 3 d postburn with values of 14.5 +/- 1.3, t = 13.306; 0.15 +/- 0.04, t = 3.460 and 0.11 +/- 0.05, t = 2.292, respectively (P < 0.05). (2) Treatment with c-fos antibody decreased the expression of these three growth-related factors when compared with that in the controls. There were a 80% decrease in c-myc and a 85% decrease in bFGF. (3) Treatment with H7 inhibited the gene and protein expression of c-fos and bFGF. The peak times of c-myc gene and protein were delayed to 6 hours post injury. Treatment with BDM induced an earlier expression of c-myc mRNA at 6 hours, and increased the gene and protein expressions of c-fos and bFGF. The total amount of their values was increased to 0.23 +/- 0.07, t = 2.942 and 0.69 +/- 0.07, t = 5.134 (P < 0.01). CONCLUSIONS: Thermal injury can induce a series of growth-related gene expression showing a phasic control and regional distribution. c-fos plays an important role in wound healing process. Protein kinase C pathway may repair wound.

[An epidemic investigation of eperythrozoonsis in Shandong province].

OBJECTIVE: To learn the status of infection of eperythrozoonsis in Shandong province. METHODS: Seven hundred and seventy-six human blood samples, 198 domestic animal blood samples and 114 wild rat blood samples were collected. Eperythrozoon was detected in the human and animal blood stain slides under microscope. RESULTS: Overall infection rate of Eperythrozoon in human population was 7.3%. Rates were significantly different (P < 0.01) in different districts but no significant difference was found between men and women (P > 0.05). The rate among adults was higher than that of teenagers and elderly. It was also noticed that the rates of people affected with chronic diseases as hepatitis, diabetes and arthritis were higher than those without. The infection rates of goat, cows and rats were 15.6%, 3.2% and 0, respectively. CONCLUSION: Data showed that Eperythrozoon infection was intensively identified in the people and animals in Shandong province.

Expression of oncoproteins c-fos and c-jun in hypertrophic scars and chronic dermal ulcers and their regulation of basic fibroblast growth factor.

OBJECTIVE: To explore the characteristics of oncoprotein expression of c-fos and c-jun in hypertrophic scars and chronic dermal ulcers and their regulation of basic fibroblast growth factor (bFGF). METHODS: Tissues of hypertrophic scars (n = 8), chronic dermal ulcers (n = 8) and normal skin (n = 5) were taken from 21 patients with burns and chronic dermal ulcers in operation. The ABC immunohistochemical method was used to characterize the gene product expression of c-fos, c-jun and bFGF in the above tissues. RESULTS: In normal skin, both c-fos and c-jun protein expression and bFGF protein expression were observed. The signals of both oncoproteins were localized mainly in subcutaneous fibroblasts, but, positive expression of the bFGF protein was mainly in keratinocytes. In hypertrophic scars, positive expression of both oncoproteins could be found mainly in fibroblasts, but bFGF was mainly in fibroblasts and endothelial cells. In chronic dermal ulcers, endothelial cells, some of inflammatory cells and fibroblasts were positive for both of oncoproteins, but the expression of bFGF was only seen in fibroblasts and endothelial cells. CONCLUSIONS: The results indicate that the interaction between both oncoproteins and bFGF exists, and the regulating action between protooncogenes and bFGF is a major course in wound healing. The different expressions of c-fos and c-jun gene products play an important role in regulate bFGF action, thus affecting wound healing.

Expression characteristics of c-fos, c-myc and bFGF in early burn tissue.

OBJECTIVE: To investigate the sequence and characteristics of expression of proto-oncogenes, c-fos, c-myc and endogenous basic fibroblast growth factor (bFGF), in burn tissue from rats. METHODS: A partial thickness burn of 30% TBSA was created on the backs of Wistar rats. Immunohistochemistry was used to detect the expression of these proteins in normal and burned skin at 3 hours, 6 hours, 1 day, 3 days, 7 days and 14 days postburn. RESULTS: Expression of c-fos, c-myc and bFGF was induced by the burn, but these three proteins showed different expression levels in sequence and distribution. Expression of c-fos increased and peaked at 3 hours postburn. Stain positive granules were distributed mainly in the cytoplasm of basal cells and the nuclei of fibroblasts. The expression of bFGF protein increased at 6 hours and peaked at 1 day postburn; it was distributed in the cytoplasm of fibroblasts. c-myc protein peaked at 3 days postburn and was also distributed in the cytoplasm of fibroblasts. CONCLUSIONS: Thermal injury can induce c-fos, c-myc and bFGF expression and show phasic control and regional distribution. The phasic expression of these three proteins indicates that there maybe a regulatory relationship between proto-oncogenes and growth factors in early burn. This may play an important role in late tissue repair.

[The effect of bombesin on cyclin D1/CDK4 of immortalized human gastric epithelial cell line].

OBJECTIVE: To investigate the regulatory effect of bombesin on the growth of immortalized human gastric epithelial cell line GES-1, as well as on the cell cycle-regulating elements cyclin D1 and CDK4. METHODS: GES-1 cells were treated with bombesin, alone or together with an bombesin antagonist. The growth effect was evaluated by MTT method, cyclin D1 expression by flow cytometry, CDK4 expression by Western blot plus enhanced chemoluminescence and CDK4 activity by immunoprecipitation with antibody-coated Sepharose beads. RESULTS: Bombesin at 10(-7) mol/L significantly stimulated the growth of GES-1 cells, increased cyclin D1 expression which was inhibited by bombesin antagonist. Bombesin also increased CDK4 protein expression and CDK4 activity. CONCLUSION: In an immortalized human gastric epithelial cell line, bombesin promotes cell cycle progression via an increase in the expression of cyclin D1/CDK4 and CDK4 activity.

[The relationship between eosinophils, activated T lymphocyte, leukotreine and the exercise-induced asthma].

OBJECTIVE: To explore the relationship between eosinophils, T lymphocyte, leukotreine (LT) and exercise-induced asthma (EIA). METHODS: In 32 asthmatic patients (13 with EIA and 19 asthmatic without EIA) and 8 normal persons, serum eosinophil cationic protein (ECP) and actived T lymphocyte (CD(25)(+)%) in peripheral blood were measured at the time before and 10, 60 minutes after exercise testing. The maximum minute ventilation (V(E)) were also measured. Another 22 non-smoking asthmatic patients with EIA and 10 normal subjects were enrolled for measuring urinary leukotriene E(4) levels before and 2 hours after exercise. EIA patients received zafirlukast 20 mg twice a day for three days. Standardized exercise challenge were performed after 72 hours of medication. RESULTS: There was a linear relationship between ECP, CD(25)(+)% and the forced expiratory volume in one second (FEV(1)) in asthmatic group (r = -0.79 and -0.61 all P < 0.01). Expressions of IL-4 mRNA, IL-5 mRNA CD(25)(+)% and serum ECP levels showed no significant difference at all testing points between two asthmatic groups, meanwhile, V(E) also showed no difference among three groups. Urinary LTE(4) levels in EIA group increased significantly two hours after exercise. Zafirlukast significantly reduced AUC(0 approximately 60 min) and the percentage of maximum fall in FEV(1) and accelerated the falling FEV(1) to return to pre-exercise baseline. CONCLUSIONS: Hyperventilation during exercise may not be the cause leading to EIA, neither did the activation of T lymphocytes, IL-4 and IL-5 gene expression nor ECP. Leukotriene may play an important role in the pathogenesis of EIA.

[Clinical investigation of burn wound infection with type I herpes simplex virus].

OBJECTIVE: To elucidate the clinical features of burn wound infection with type I herpes simplex virus (HSV -- 1), so as to propose the possible therapeutic measures. METHODS: Burn wound infection with HSV -- 1 was diagnosed in 7 cases by means of bacterial culture, PCR detection of virus, immunohistochemical staining and pathomorphological section with the exclusion of other causes. In addition, the clinical signs and the features of the development of the infection in 62 cases were recorded and analyzed. RESULTS: In the burn patients with the infection, the burn wounds were covered by tumor-like masses with the cauliflower appearance. In the 7 cases who underwent pathomorphological examination, 5 with positive HSV -- I. Epithelial cells exhibited balloon -- like appearance, and reticular degeneration, and inclusion bodies were found in balloon-like cells under microscope examination. In the late stage, there revealed obvious proliferation of epithelial prickle cell layer in shape of pseudopapillomatous mass with the obvious liquefaction of basal cells. The sufferers exhibited evident local signs with mild systemic reaction. The management of the wound included wet compress with the addition of neomycin and pentacyclic guanosine. CONCLUSION: With the employment of PCR virus detection and immunohistochemical staining, burn wound infection with HSV -- I could be identified. Non -- surgical treatment could be effective.

Purification of human prothrombin from Nitschmann fraction III using DEAE membrane radial flow chromatography.

A DEAE membrane radial column (4x5 cm) was used to purify human prothrombin from Nitschmann fraction III and performances of the column were investigated. Sample flow-rate of 20-30 ml/min (i.e., 24-36 bed volumes/h) and elution velocity up to 40 ml/min (i.e., 48 bed volumes/h) were obtained without compromising the separation efficiency. Its breakthrough capacity was one-third to half that of the commercial DEAE Sepharose Fast Flow media. It was concluded that the novel column is an attractive alternate to traditional axial column.

Measurements of N2O and CH4 from the aerated composting of food waste.

Emissions of N2O and CH4 from an aerated composting system were investigated using small-scale simulated reactors. The results show relatively high emissions of N2O at the beginning of composting, in proportion to the application amount of food waste. After 2 days, the N2O emission decreased to 0.53 ppmv on average, near to the background level in the atmosphere (0.45 ppmv). The addition of composted cattle manure increased N2O emissions not only at the beginning of composting, but also during the later period and resulted in two peak emission curves. Good correlation was observed between the N2O concentration at the air outlet and NO2- concentration in waste, suggesting a generation pathway for N2O from NO2- to N2O. Methane was only detected in treatments containing composted cattle manure. The high emission of methane illustrates the involvement of anoxic/anaerobic microorganisms with the addition of composted manure. The result suggests the existence of anoxic or anaerobic microsite inside the waste particles even though ventilation was employed during the composting process.

Enzymes of vancomycin resistance: the structure of D-alanine-D-lactate ligase of naturally resistant Leuconostoc mesenteroides.

BACKGROUND: The bacterial cell wall and the enzymes that synthesize it are targets of glycopeptide antibiotics (vancomycins and teicoplanins) and beta-lactams (penicillins and cephalosporins). Biosynthesis of cell wall peptidoglycan requires a crosslinking of peptidyl moieties on adjacent glycan strands. The D-alanine-D-alanine transpeptidase, which catalyzes this crosslinking, is the target of beta-lactam antibiotics. Glycopeptides, in contrast, do not inhibit an enzyme, but bind directly to D-alanine-D-alanine and prevent subsequent crosslinking by the transpeptidase. Clinical resistance to vancomycin in enterococcal pathogens has been traced to altered ligases producing D-alanine-D-lactate rather than D-alanine-D-alanine. RESULTS: The structure of a D-alanine-D-lactate ligase has been determined by multiple anomalous dispersion (MAD) phasing to 2.4 A resolution. Co-crystallization of the Leuconostoc mesenteroides LmDdl2 ligase with ATP and a di-D-methylphosphinate produced ADP and a phosphinophosphate analog of the reaction intermediate of cell wall peptidoglycan biosynthesis. Comparison of this D-alanine-D-lactate ligase with the known structure of DdlB D-alanine-D-alanine ligase, a wild-type enzyme that does not provide vancomycin resistance, reveals alterations in the size and hydrophobicity of the site for D-lactate binding (subsite 2). A decrease was noted in the ability of the ligase to hydrogen bond a substrate molecule entering subsite 2. CONCLUSIONS: Structural differences at subsite 2 of the D-alanine-D-lactate ligase help explain a substrate specificity shift (D-alanine to D-lactate) leading to remodeled cell wall peptidoglycan and vancomycin resistance in Gram-positive pathogens.

Oligodendrocyte lineage and the motor neuron connection.

One of the more surprising recent discoveries in glial biology has been that oligodendrocytes (OLs) originate from very restricted regions of the embryonic neural tube. This was surprising because myelinating OLs are widespread in the mature central nervous system, so there was no reason to suspect that their precursors should be restricted. What we now know about early OL development suggests that they might have as much (or more) in common with ventral neurons-specifically motor neurons (MNs)-as with other types of glia. This has implications for the way we think about glial development, function, and evolution. In this article we review the evidence for a shared MN-OL lineage and debate whether this is the only lineage that generates OLs. We decide in favour of a single embryonic lineage with regional variations along the anterior-posterior neuraxis.

Chromatography of human prothrombin from Nitschmann fraction III on DEAE Sepharose Fast Flow using axial and radial flow column.

An axial column (3 x 2.6 cm) and a radial flow column (3.5 x 5 cm) packed with DEAE Sepharose Fast Flow media was evaluated for the separation of human prothrombin from Nitschmann fraction III. Under radial flow conditions, a sample flow rate up to 14 mL/min (approximately 18 bed vols/h) was achieved. Breakthrough capacity was determined and both columns had almost the same breakthrough capacity per mL media, indicating that the sample loading was independent of radial column geometry.

Gibberellin signal transduction.

Recent studies using biochemical and genetic approaches have identified a number of components, including several negative regulators, of the gibberellin (GA) signal transduction pathway in higher plants. The basal state of GA signaling is likely to be repressive, and the GA signal seems to activate the pathway by de-repression to allow GA-stimulated growth and development.
