Parallel studies in parotid and pancreatic function and histology were carried out in Thomas fistula dogs with irradiation-induced chronic pancreatitis. After completion of base line studies of parotid and pancreatic secretion, four dogs were subjected to 2,400 rads tumor dose over two weeks (with a nominal single dose of 1,175 rets); two additional dogs served as control. Sequential studies were performed on pancreatic secretion in response to secretin and on parotid secretion in response to urecholine. After a short-lived period of hypersecretion. There was a progressive reduction in pancreatic secretory rate, bicarbonate and enzyme outputs (greater than 90% after three months). Parotid secretion increased over this period but subsequently decreased, so that at eight months there was a marked reduction both in pancreatic and parotid function. Whereas pancreatic histology showed diffuse interstitial fibrosis and reduction of acinar tissue, no histologic changes were noted in the parotid glands. Although the mechanisms are unclear, alterations in parotid secretion may be of value in the diagnosis of chronic pancreatitis, in the evaluation of its severity, and may yield information on the duration of the disease.

Mature female mice (ICR-JCL), 8 to 12 weeks of age, were artificially inseminated at 8:30-9:30 a. m. on the day of estrus vaginal smear (about 3-7 hr after ovulation) with 3.18 X 10(6), 1.83 X 10(6) and 1.15 X 10(6) sperms from four, two and one cauda epididymidis, respectively, of adult males which were suspended in 50 microliter of a modified Krebs-Ringer-bicarbonate solution and incubated at 37 degrees C was under 5% CO2 in air for an hour. Immediately after insemination, pseudopregnancy induced by an artificial penis and a vaginal tampon. Out of 13 females inseminated with 3.18 X 10(6) sperms, 9 females showed placental signs and 8 of them gave birth to the mean 10.5+/-2.20 (M+/-S.D.) young at term. Four of 13 females having received 1.83 X 10(6) sperms became pregnant giving birth to the mean 4.3+/-2.1 (M+/-S.D.) young at term. On the other hand, 5 out of 6 females failed to become pregnant following insemination with 1.15 X 10(6) sperms, and only one showed a placental sign and gave birth to twelve young at term. It is concluded that the conception rate and litter size are both dependent on the number of sperms inseminated and that more than 3 X 10(6) sperms are necessary to get the conception rate and the litter size comparable to those in natural mating.
Effects of duodenal alkalinization on pancreatic secretion were evaluated in seven dogs including two antrectomized dogs with chronic duodenal fistulas. Continuous intravenous injection of secretin or secretin plus CCK PZ was administered as a stimulus for the pancreatic secretion. Intraduodenal infusion of alkaline solutions such as pancreatic juice (pH 8.6), bile (pH 8.1), bicarbonate solution (160 mEq./I., pH 8.6) and TRIS buffer solution (0.3 M, pH 9.5) caused a moderate increase in protein output and a modest or no increase in flow but no change in bicarbonate output. During infusion of pancreatic juice, varied doses of secretin (0, 0.015, 0.03, 0.06 u./kg./min.), or secretin (0.03 u./kg./min.) plus CCK-PZ (0.05 u./kg./min.) were administered intravenously. For example, under the stimulation of secretin (0.03), flow increased modestly (17.7%), an increase which was of statistical significance. Protein output also showed a significant increase of 35.7%. There was, however, no response of the pancreas to the infusion of pancreatic juice when the gland was stimulated by secretin plus CCK-PZ. Antrectomy did not alter the effect of alkaline intraduodenal infusions on pancreatic secretion.

Complementary and genomic DNAs isolated from the fibroblasts of 10 Japanese (7 late infantile, 2 juvenile, and 1 adult form of the disease) and one Caucasian patient with Niemann-Pick disease type C were analyzed for mutations in the NPC1 gene. Fourteen novel mutations were found including small deletions and point mutations. A one-base deletion and a point mutation caused splicing errors. The mutations were not clustered in any particular region of the gene and were found both in and out of the transmembrane domains. Three patients were homozygous, five were compound heterozygous, and the remaining three were suspected of being compound hetrozygous with an unknown error in one of their NPC1 alleles. Of the 14 mutations, the G1553A substitution that caused a splicing error of exon 9 appeared to be relatively common in Japanese patients, because two patients were homozygous and one patient was compound heterozygous for this mutation.
It was recently reported that TGF-beta 1 either had no significant effect on or increased the survival of dopaminergic neurons in culture. TGF-beta 2 and TGF-beta 3 were reported to cause increased survival or to greatly inhibit survival. The transforming growth factors are a highly pleiotropic group of compounds, and the above results suggest that their actions may be critically dependent on the conditions of the assay. We have therefore tested these compounds under optimal conditions of culture, in a medium containing a low (2.5%) concentration of fetal bovine serum. TGF-beta 2 and 3 inhibited neuronal (MAP2-pos) survival only at the highest concentration (10 ng/ml) tested, while inhibition of survival of dopaminergic neurons was observed at 1.0 and 10 ng/ml. These results therefore suggest that the inhibitory action of TGF-beta 2 and 3 on the survival of dopaminergic neurons in culture, under the experimental conditions outlined, may be relatively specific.

Among 145 patients with carcinoma of the pancreas who had been admitted to three institutions between 1976 and 1985, 117 were entered into a prognostic factor analysis system according to Cox's proportional hazards model. The treatment was classified into four modalities: bypass surgery, single-dose intraoperative radiotherapy (IOR), IOR combined with external irradiation, and resection with or without various radiations. The most important prognostic factors for treatment were identified as location of tumor, TNM stage, and relief of back pain. Adjusting the imbalances among the treatment groups using Cox's model, the superiority of IOR with external irradiation was confirmed for both bypass and single-dose IOR. With this adjustment, the survival rates proved to be more meaningful than the simple Kaplan-Meier estimates.
