Cytopathic effect of heat-labile toxin of Bordetella parapertussis on aortic smooth muscle cells from pigs or guinea pigs.

The in vitro effect of the heat-labile toxin (HLT) of Bordetella parapertussis on HeLa, baby hamster kidney (BHK), chinese hamster ovary (CHO), myeloma (Pa-NS-1), human embryonic lung (HEL-R66) cells, erythrocytes, adipocytes and lymphocytes from guinea pigs, mice or rats, or aortic smooth muscle cells from pigs or guinea pigs was examined. Within 8, 6, 4, 2, and 2 hr after the exposure to 1, 3, 10, 30, and 100 MNDs/ml of HLT, respectively, the cultured smooth muscle cells only showed a cytopathic change. When the cells exposed to HLT were washed out within 60 min post-exposure, the change could be induced with an extend period of lag. Histamine, KCl or norepinephrine caused similar change in the cells, but the period of lag was within 30 min. The HLT activity was neutralized by an anti-B. parapertussis- or B. bronchiseptica-HLT guinea pig IgG. HLT had no effects on any other cells tested.

ATP-dependent glutamate uptake into synaptic vesicles from cerebellar mutant mice.

The ATP-dependent glutamate uptake system in synaptic vesicles prepared from mouse cerebellum was characterized, and the levels of glutamate uptake were investigated in the cerebellar mutant mice, staggerer and weaver, whose main defect is the loss of cerebellar granule cells, and the nervous mutant, whose main defect is the loss of Purkinje cells. The ATP-dependent glutamate uptake is stimulated by low concentrations of chloride, is insensitive to aspartate, and is inhibited by agents known to dissipate the electrochemical proton gradient. These properties are similar to those of the glutamate uptake system observed in the highly purified synaptic vesicles prepared from bovine cortex. The ATP-dependent glutamate uptake system is reduced by 68% in the staggerer and 57-67% in the weaver mutant; these reductions parallel the substantial loss of granule cells in those mutants. In contrast, the cerebellar levels of glutamate uptake are not altered significantly in the nervous mutant, which has lost Purkinje cells, but not granule cells. In view of evidence that granule cells are glutamatergic neurons and Purkinje cells are GABAergic neurons, these observations support the notion that the ATP-dependent glutamate uptake system is present in synaptic vesicles of glutamatergic neurons.

Silver-banded karyotypes of the rainbow trout and the brook trout and their hybrids: disappearance in allotriploids of Ag-NORs originated from the brook trout.

Silver-banded karyotypes of the rainbow trout Salmo gairdneri, the brook trout Salvelinus fontinalis, and their hybrids were described. Diploid-type and triploid-type hybrids were obtained. Triploid-type hybrids had two maternal genomes of the rainbow trout and one paternal genome of the brook trout. The rainbow trout had one pair of M or SM with Ag-NORs near the satellite. In the brook trout, Ag-NORs were observed in four pairs of chromosomes, but each cell had different number of chromosomes with Ag-NORs. In all observed cells of triploid-type hybrids, Ag-NORs originated from the brook trout were not recognized, whereas those from the rainbow trout were found.

[Anti-cancer efficacy of peplomycin, adsorbed on activated carbon particles, against lymph node metastases in mice].

PEP-CH is comprised of activated carbon particles that adsorb peplomycin. Mice which were inoculated with 5 x 10(5) MH 134 tumor cells subcutaneously opposite the foot-pad of the left hind paw on day 0, received a subcutaneous injection of peplomycin, 0.25 mg/mouse, in the form of PEP-CH or a peplomycin aqueous solution administered at the foot-pad of the left hind paw on day 10. The left popliteal lymph nodes then were transferred intraperitoneally to normal mice on day 14. In the PEP-CH group, the survival time of the recipients was statistically and significantly long, and the transferred tumor cell number, estimated with a calibration curve, was markedly small when compare to mice in the other treatment groups.

Reduced pulmonary toxicity of peplomycin in a new drug-delivery system.

Pulmonary toxicity was examined by means of Matsuda and Takahashi's procedure in peplomycin solution, and two types of a new dosage form of peplomycin (PEP-CH), which has peplomycin adsorbed on to activated carbon particles. One was PEP-CH IP, and is designed for intraperitoneal administration. The other was PEP-CH IM, for intramuscular administration. Male mice of ICR strain received a bolus injection of 50 mg kg-1 peplomycin in the form of aqueous solutions PEP-CH IP and PEP-CH IM. The survival rate was 100% after 5 weeks in both groups that were administered PEP-CH. The rate was 50% for the group given peplomycin solution intraperitoneally, and 70% for the group given peplomycin solution intramuscularly. Five weeks after administration the mice were killed, and the grade and the incidence of pulmonary fibrosis were evaluated histologically. The grade and incidence of pulmonary fibrosis were zero in the two groups given PEP-CH. The grade was 0.33 and 1.76, and the incidence was 60% and 86%, respectively, in the group given peplomycin solution intraperitoneally and in the group given peplomycin solution intramuscularly.

Metabolic effects of 3'-deoxyadenosine (cordycepin) and 2-halo-3'-deoxyadenosine on repair of X-ray-induced potentially lethal damage in Chinese hamster V79 cells.

Using cultured Chinese hamster V79 cells, an attempt has been made to examine the phosphorylation of cordycepin and its 2-halo derivatives (2-chloro-3'-deoxyadenosine, 2-bromo-3'-deoxyadenosine, and 2-iodo-3'-deoxyadenosine) by adenosine kinase, within the cells, and to correlate it with their cytotoxicities and abilities to inhibit the repair of X-ray-induced potentially lethal damage (PLDR). Of all compounds, only cordycepin was found to be phosphorylated and showed both potent cytotoxicity and ability to inhibit PLDR.

Restriction fragment length polymorphism of bcr in Japanese patients with hematological malignancies.

We surveyed DNAs from patients with various hematological malignancies by Southern blot hybridization to analyze bcr rearrangements, and detected a new restriction fragment length polymorphism (RFLP) of the breakpoint cluster region at a BamHI site in three patients. By using a 1.2-kb HindIII-BglII 3' bcr probe, unusual BamHI restriction enzyme fragments (1.9 kb and 1.4 kb) were detected from the DNAs of three patients with hematological malignancies. DNAs from cultured fibroblasts derived from the skin of a patient, as well as from peripheral leukocytes of the father of a patient and the mother of another patient, showed identical 1.9- and 1.4-kb additional bands, besides a 3.3-kb germline band, establishing that polymorphism, rather than gene arrangement, was responsible for these additional restriction enzyme fragments. However, RFLP was not detected in the DNAs of 40 normal unrelated individuals.

[Clinical evaluation of cefpodoxime proxetil in the treatment of skin and soft tissue infections. A double blind comparison of cefpodoxime proxetil and cefaclor].

In order to objectively evaluate the effectiveness, safety and usefulness of the new oral cephem cefpodoxime proxetil (CS-807, CPDX-PR) for the treatment of skin and soft tissue infections, a double-blind comparative study was undertaken using cefaclor (CCL) as the control drug. CPDX-PR and CCL were administered for 7 days at daily doses of 400 mg (divided into 2 portions) and 750 mg (divided into 3 portions), respectively. A total of 243 patients (118 in the CPDX-PR group and 125 in the CCL group) was treated in this study. The effectiveness, safety and usefulness were evaluated in 222 (106 in the CPDX-PR group and 116 in the CCL group), 234 (113 in the CPDX-PR group and 121 in the CCL group) and in 223 patients (107 in the CPDX-PR group and 116 in the CCL group), respectively. There were no differences in patients' backgrounds between the 2 groups, except for the presence or the absence of surgical treatments. The results we obtained are summarized below: 1. In the evaluation of clinical efficacy by the subcommittee, excellent, good, fair and poor efficacy were observed in 36, 43, 17 and 10 patients in the CPDX-PR group, respectively; the efficacy rate was, therefore, calculated to be 74.5%. As for the CCL group, respective results were observed in 50, 39, 17 and 10 patients, indicating an efficacy rate of 76.7%. There was no significant difference between the 2 groups. Improvement rates judged by physicians in charge were 80.2% in the CPDX-PR group and 88.8% in the CCL group. Moreover, no significant difference in diseases or severity were found between the 2 groups. 2. As for the bacteriological efficacy, the 2 groups showed high elimination rates, as 90.1% and 91.6% of the disease causing bacteria were eliminated in the CPDX-PR group and in the CCL group, respectively. Elimination rates in single infections with Staphylococcus aureus were determined to be 85.7% in the CPDX-PR group and 85.0% in the CCL group. 3. Although 6 patients in the CPDX-PR group and 2 patients in the CCL group developed side effects, which were mainly gastrointestinal symptoms, there was no significant difference in the incidence of side effects between the 2 groups. Abnormal laboratory values were found in 5 patients in the CPDX-PR group and 1 patient in the CCL group. 4. There was no significant difference in the usefulness between the 2 groups.(ABSTRACT TRUNCATED AT 400 WORDS)
