Periodic paralysis with cardiac arrhythmia.

In 1963, Klein et al. first described two girls with normokalemic periodic paralysis and cardiac arrhythmia. We have observed a 15-year-old girl with cardiac arrhythmia and normokalemia but with some features of hyperkalemic, periodic paralysis. The patient showed a waddling gait and Gower's sign. She had atrophy of the proximal muscles and deep tendon reflexes were reduced. Her ECG showed bigeminy with multifocal premature ventricular contractions. At the start of an episode of muscle weakness, the serum potassium concentration rose from 3.1 to 4.4 mEq/l. Muscle weakness was not provoked by intravenous administration of 45 g glucose or of 72 g glucose followed by 10 units of regular insulin. Muscle weakness was evoked by the ingestion of 4 g potassium chloride. During the provoked muscle weakness, the ECG showed normal sinus rhythm temporarily.

Establishment of cell strains from human urothelial carcinoma and their morphological characterization.

We have examined the conditions for cultivation of enzymatically dispersed cells from 34 human urothelial transitional cell carcinomas (TCC) of various types. By employing two culture methods, stationary and tapping suspension, and by using the synthetic medium DM 160 supplement with human umbilical cord serum and fetal bovine serum, six cell strains were established. In two strains the tapping suspension culture method was suitable for growth of highly malignant cancer cells that detach easily from the glass surface in stationary cultures. Each of the six cell strains has been maintained in culture for over 30 months with repeated subcultures of 32 to 128 times. The histopathological features of the original TCC were three differentiated papillary types and three anaplastic nonpapillary types. In two cell strains from TCC with low malignancy, however, the cancer masses that formed in nude mice differed from the original TCC in which they became more malignant, and one cell strain resembled the original TCC closely. In three stationary culture cell strains the epithelial nature was demonstrated by the presence of desmosomes and tonofilaments. In one cell strain only tonofilaments were present. In two tapping suspension culture cell strains the presence of desmosomes was not shown clearly, but fine tonofilaments were observed in one cell strain.

Quantitative analysis of changes in cell shape of Amoeba proteus during locomotion and upon responses to salt stimuli.

A new parameter expressing the complexity of cell shape defined as (periphery)2/(area) in 2D projection was found useful for a quantitative analysis of changes in the cell shape of Amoeba proteus and potentially of any amoeboid cells. During locomotion the complexity and the motive force of the protoplasmic streaming in amoeba varied periodically, and the Fourier analysis of the two showed a similar pattern in the power spectrum, giving a rather broad peak at about 2.5 X 10(-3) Hz. The complexity increased mainly due to elongation of the cell as external Ca2+ increased. This effect was blocked by La3+, half the inhibition being attained at 1/200 amount of the coexisting Ca2+. On the other hand, the complexity decreased due to rounding up of the cell as the concentration of other cations, such as Sr2+, Mg2+, Co2+, Ni2+, Na+, K+ etc., increased. Irrespective of the opposite effects of Ca2+ and other cations on the cell shape, the ATP concentration in amoeba decreased in both cases with increase of all these cations. The irregularity in amoeboid motility is discussed in terms of a dynamic system theory.

Xanthogranulomatous transformation of metastatic testicular tumor.

A case of testicular tumor with pulmonary metastasis which was transformed into xanthogranuloma is reported. The metastatic lesion was treated first by combination chemotherapy regimen of platinum, vinblastine, and peplomycin, but the pulmonary metastasis did not completely disappear and a segmental resection of the left upper lung including a metastatic nodule was performed. Interestingly, histologic examination of the pulmonary metastasis revealed so-called xanthogranuloma.

Echocardiographic evaluation of aortic cusp prolapse in children with ventricular septal defect.

Echocardiograms were obtained from 48 Japanese children with ventricular septal defect (16 having aortic cusp prolapse, Group I, and 32 without it, Group II). In the case of right coronary cusp prolapse, the right coronary sinus protrudes anteriorly into the right ventricular outflow tract, and thus, the anteroposterior diameter of the aortic root increases. In the case of non-coronary cusp prolapse, the non-coronary sinus bulges posteriorly into the right ventricle, and thus, the aortic root increases in size. For evaluating the degree of these prolapses quantitatively, we measured the aortic root diameter echocardiographically and expressed them as a percent of a normal one. In Group I the aortic root diameter was 131 +/- 9% (mean +/- SD) and in Group II it was 105 +/- 7%, and the difference between the 2 groups was statistically significant (p less than 0.001). In Group I 14 of the 16 patients had a value greater than 120%, while all 32 patients of Group II had a value smaller than 120%. Thus, in children with ventricular septal defect, an aortic root diameter greater than 120% of normal suggests the presence of aortic cusp prolapse. Systolic semiclosure of the aortic valve was found in 8 patients of Group I (50.0%) and in 2 of Group II (8.7%). Coarse systolic fluttering of the pulmonary valve with an amplitude of greater than 3 mm was detected in 6 of Group I (40.0%) and in 3 of Group II (10.0%). Therefore, semiclosure of the aortic valve and fluttering of the pulmonary valve are considered to be also useful for evaluating aortic cusp prolapse qualitatively.

Inhibition of carp liver mitochondrial monoamine oxidase by some commonly-used detergents.

The inhibitory effects of some detergents commonly used in biochemical research on carp liver mitochondrial monoamine oxidase were examined. Sodium dodecylsulfate, octyl-beta-D-glucopyranoside, sodium cholate and Triton X-100 at relatively low concentrations caused strong dose-dependent inhibition of the activity towards tyramine, but digitonin caused only weak inhibition. Sodium dodecylsulfate, octyl-beta-D-glucopyranoside and sodium cholate caused almost complete inhibition of activity in the concentration ranges tested. The extent of inhibition by Triton X-100 was greater after preincubation at 37 degrees C for 30 min than that without preincubation, but with or without preincubation, the inhibition was not substrate-selective and was not complete at a relatively high concentration (2%) of Triton X-100. Without preincubation, the mode of inhibition by Triton X-100 was competitive and reversible with respect to the oxidations of 5-HT, tyramine and PEA, but after preincubation (37 degrees C for 30 min), it became noncompetitive and irreversible, depending on the concentration of detergent used. These findings suggest that it had different actions on the enzyme depending on preincubation. Triton X-100 also slightly changed enzyme sensitivity towards clorgyline and deprenyl, regardless of the preincubation time or the substrate used. Some possible mechanisms of the inhibitory effect of Triton X-100 are discussed.

Effects of room temperature on reproduction, body and organ weights, food and water intakes, and hematology in mice.

Two consecutive generations of mice were raised at graded room temperatures ranging from 12 to 32 degrees C at intervals of 2 degrees C. The delivery rate decreased at 30 and 32 degrees C, and the litter size and weaning rate decreased above 28 degrees C. No significant difference was demonstrated within the 12-26 degrees C range for any reproduction parameters observed. The body weights of the first-generation mice born at 22 degrees C and transferred to rooms at assigned temperature did not significantly differ within the 14-28 degrees C range after 8 and 16 weeks of exposure. The second-generation mice born and reared at various temperature levels did not show significant inter-group difference in weight within the 20-26 degrees C range at any age in the growth period. No significant inter-group difference was observed within the 20-26 degrees C range in respect of food and water intakes in the second-generation mice. Hematological values and organ weights in the first and second generation mice of both sexes did not significantly differ in any parameter within the 20-26 degrees C range. The results suggest the temperature range of 20-26 degrees C to be optimal for laboratory mouse rooms.

[Effect of 1-[3-[4-(diphenylmethyl)-1-priperazinyl]-propyl]-1,3-dihydro-2H-benzimidazol-2- one (oxatomide) on contraction of canine bronchial muscle].

Effect of oxatomide on contraction of canine bronchial smooth muscle caused by administration of histamine and by challenge of ascaris-antigen were investigated and compared against the effects of histamine H1-receptor antagonist (CPM) and inhibitor of histamine release from mast cells (DSCG). Oxatomide in concentrations of 0.05 microgram/ml to 2.0 microgram/ml inhibited, dose-responsively, contraction of bronchial smooth muscle strip by administration of histamine (10 micrograms/ml). When this concentration was less than 0.3 microgram/ml, CPM inhibited the contraction significantly stronger than oxatomide. However, in concentration over 0.5 microgram/ml, no difference in inhibitory effect could be demonstrated. On the other hand, DSCG could not inhibit the contraction. Oxatomide in concentrations of 0.05 microgram/ml to 1.0 micrograms/ml inhibited, dose-responsively, contraction of the muscle strip sensitized spontaneously by ascaris antigen challenges. There was no difference in inhibitory effect between oxatomide and DSCG at the same level of concentration. Oxatomide (0.5 microgram/ml) and CPM (0.5 microgram/ml) both inhibited completely the constriction response of bronchus caused by histamine stimulation (10 micrograms/ml). Although constriction response by ascaris antigen challenge was inhibited completely by oxatomide (0.5 microgram/ml), it could not be inhibited completely by CPM (0.5 microgram/ml). Oxatomide inhibited even the residual constriction response which could not accomplished by CPM. These results suggest that oxatomide might have not only an effect as a histamine H1-receptor antagonist and anti-histamine releasing agent from mast cell, but also anti-SRS-A.

[Pre-operative endoscopic infusions of an adsorbent containing mitomycin C (MMC-CH44) in stomach cancer].

Small charcoal particles adsorbing Mitomycin C (MMC-CH) were used in the treatment of lymphnode metastasis of gastric cancer. Immediately after injection of MMC-CH into the gastric wall, lymph capillaries and regional lymphnodes were stained in black and the high concentration of MMC was delivered with charcoal particles. MMC-CH was injected preoperatively using endoscope into the gastric wall of 20 patients with gastric cancer. MMC-CH was observed in 70% of metastatic regional lymphnodes. Systemic side effect was not noted at all. Erosion and shallow ulcers were observed at the site of injection but no perforation occurred.

Structural analysis of bovine mitochondrial tRNASer (AGY).

Mitochondrial serine tRNA lacking D stem (anticodon AGY) was purified from bovine cardiac muscle in the mg order. The nucleotide sequence was identical to that reported previously, with t6A adjacent to the 3' end of anticodon (Arcari, P. and Brownlee, G.G. (1980) Nucleic Acids Res. 8 5207-5212). The procedures for large scale preparation and some properties of this tRNASer are reported.
