[Immunofluorescent study of the origin of connective tissue cells in xenogenic radiation chimeras under normal conditions and in aseptic inflammation].

The origin of elements of the focus of aseptic inflammation and the normal subcutaneous connective tissue in the xenogenic (mouse-rat) radiation chimaeras was investigated by means of indirect Coons method with antiserum to the rat bone marrow cells. The cells of the imflammation focus (leucocytes, macrophages, fibroblasts or fibroblast-like cells, polynuclear giant cells of foreign bodies), as well as leucocytes, macrophages and some fibroblasts of the normal subcutaneous connective tissue, were shown to take their origin from the transplanted bone marrow cells of the donor.

[Immunofluorescent study of the hemopoietic organs of xenogeneic mouse radiation chimeras].

An immunofluorescent study of hemopoietic organs in xenogenic (mouse-rat) radiation chimaeras has been carried out by means of specific antiserum against hemopoietic cells of the rat bone marrow. The presence of donor cells was tested at different times after the transplantation in the bone marrow, spleen, lymph nodes, thymus and liver of radiochimaeras. The transplanted cells were shown to populate all hemopoietic organs of the recipient, first of all tissues of the bone marrow type and, then, lymphoid organs. The donor (bone marrow) origin of the extramedullar foci of hemopoiesis in the liver was established.

[The relationship between hemopoiesis and osteogenesis in several species of rodents].

Differentiation of the bone marrow transplants of rats, guinea pigs and dwarf hamsters placed under renal capsule of irradiated mice was studied. Active osteogenesis occurred in the transplants of rats and guinea pigs; as to hemopoiesis-it was irregular in rats and was arrested in guinea pigs. On the contrary, in the transplants of dwarf hamsters an intensive hemopoiesis was maintained and osteogenic capacity of the stroma was poor. It is supposed that the bone tissue in some rodent species was not a prerequisite for normal maintenance of myeloid hemopoiesis, while in others-primary bone stroma formation was necessary for the subsequent functioning of hemopoietic tissue.

[A radioautographic study of the turnover and differentiation of mast cells in normal mice and mouse radiation chimeras].

The patterns of renewal of the previously labeled by 35S-sulphate mast cells of peritoneal liquor and loose connective tissue were investigated in normal mice, after their irradiation and transplantation of singenic and xenogenic bone marrow (radiation mouse chimaeras). The renewal of labeled mast cells in the peritoneal liquor proceeds markedly (ca. 10 times) more intensively than in the loose connective tissue. A stage of lymphocyte-like cells-precursors actively synthesizing sulphomucopolysaccharides is established in the series of differentiation of mast cells during postnatal ontogenesis. The duration of this stage amounts to 10-15 days. The patterns of renewal of the previously labeled by 35S-sulphate mast cells do not differ markedly in normal animals and radiation chimaeras.

[Participation of transplanted xenogenic bone marrow cells in an inflammatory reaction].

The course of inflammation reaction was studied both in the lethally irradiated mice just after the implantation of the great number of rat bone marrow cells and the xenogenic radiation chimaeras. The transplanted donor cells did not participate in the inflammation reaction in the lethally irradiated animals just after the implantation of xenogenic bone marrow. The cells of donor type were found in the inflammation focus only after the repopulation of radiation-devastated blood-forming organs of the recipient by the transplanted xenogenic bone marrow cells.

[Histogenetic characteristics of fibroblast-like cells in a focus of de novo connective tissue formation in the presence of aseptic inflammation].

In the focus of connective tissue de novo formation under aseptic inflammation, no stem blood-forming cells were found which are an initial form of histogenesis of fibroblast-like cells. The proliferative ability of fibroblast-like and other cells of the inflammation focus at its different stages has been studied using cell tissue culture and 3H-thymidine autoradiography. Fibroblast-like cells in the focus of aseptic inflammation appear to represent an independent, not connected with macrophages, line of differentiation. The characteristics of main stages of histogenesis of fibroblast-like cells is given on the basis of the author's and literary data.

[Experimental research on the cellular interactions during hemopoiesis].

We present a review of experimental studies performed at the Laboratory of Histogenesis of the Institute of Developmental Biology, Russian Academy of Sciences, on the problem of cell interactions during hemopoiesis. Special attention has been given to original experimental models, such as production of hemopoietic foci on underlayers of fibroblasts encapsulating a foreign body in the peritoneal cavity of rodents (after intraperitoneal transplantation of hemopoietic cells) and repopulation of ectopic hemopoietic territories under the kidney capsule of mice by syngeneic or xenogeneic hemopoietic cells. We describe the competitive interactions of genetically different hemopoietic cells after the transplantation of their mixtures to irradiated mice (multicomponent radiation chimeras). Xenogeneic and multicomponent chimeras have also been obtained in long-term bone marrow culture. We have examined characteristics of hemopoiesis on stromal cell underlayers produced by cells of various origins in vitro and then transplanted into the peritoneal cavity of irradiated mice. We discuss the results obtained and possible mechanisms of these phenomena.

[Use of the copolymer ethylenevinylacetate for studying the effect of hemopoietic cytokines].

Copolymer ethylenevinylacetate is known as a polymeric carrier, which ensures a lasting release of biologically active substances. An attempt has been made to use it for testing the effects of hemopoietic cytokinins. It was shown that erythropoietin, embedded in this polymer, induces formation of erythropoietic foci on acetate-cellulose membranes in the peritoneal cavity of mice.

[A study of hemopoiesis on a sublayer of peritoneal cells in the presence of hemopoietic cytokines].

We studied the effects of erythropoietin and thrombopoietin on the clonogenic capacity and direction of differentiation of the hemopoietic cells that form colonies on acetate cellulose membrane in the peritoneal cavity of mice. An increased level of erythropoietin in the blood of recipient mice after blood letting led to the appearance of erythroid colonies upon transplantation of syngeneic hemopoietic cells but did not affect the differentiation of transplanted xenogeneic (guinea pig) hemopoietic cells. Erythropoietin transported top the stromal sublayer by a polymeric carrier also induced erythroid differentiation, while thrombopoietin transported in a similar way somewhat enhanced megakaryocytopoiesis.

[Hemopoiesis on stromal sublayers formed by constant lines of fibroblasts].

We studied the formation of hemopoietic colonies on an artificial sublayer in the peritoneal cavity of mice under the influence of stromal sublayers consisting of fibroblasts of six constant cell limes. All studied lines of stromal cells supported the formation of granulocytic foci and some of them supported the formation of erythroid foci as well. It was shown that hemopoiesis was preserved or, in some cases, enhanced on sublayers of fixed (metabolically inactive) cells. The treatment of fibroblasts by E. coli lipopolysaccharide did not lead, as a rule, to significant stimulation of hemopoiesis.

[Hematopoiesis on cellulose acetate membranes in the presence of lipopolysaccharides].

The formation of hemopoietic colonies on acetate cellulose membranes in the peritoneal cavity of mice was markedly enhanced after the injection of bacterial lipopolysaccharide. In addition to granulocytic-macrophagal differentiation, the foci of erythropoiesis appeared. The stimulating effect of lipopolysaccharide was not expressed in nonirradiated mice and during the formation of hemopoietic foci on acetate cellulose membranes in the subcutaneous connective tissue.

[The bone marrow origin of the cells encapsulating a foreign body in the peritoneal cavity of xenogeneic radiation chimeras].

The origin and ultrastructure of the cells, encapsulating foreign body in peritoneal cavity of xenogeneic (rat in mouse) radiation chimeras was studied. The donor nature of the cells was identified by their karyotype and by DNA hybridization with rat ID-element. Cells with ultrastructural characteristics of fibroblasts encapsulating foreign body in the peritoneal cavity of the xenogeneic radiation chimeras were shown to originate from the transplanted (donor) bone marrow.

[The characteristics of the hematopoietic cells forming colonies on cellulose acetate membranes and their distinctions from other clonogenic cells].

In order to characterize hemopoietic cells forming colonies on membranes of cellulose acetate (CFU-acm) implanted into the peritoneal cavity of mice, we studied the effect of factors stimulating and inhibiting granulocytopoiesis on these cells. Proliferation of CFU-acm can be controlled by humoral factors and this allows us to conclude that they are not identical to CFUs and probably belong to the compartment of early hemopoietic cells of the granulocyte series. We also present evidence for fractional composition, ultrastructure and bone marrow origin of cells belonging to the layer providing for hemopoietic microenvironment for the resulting foci of hemopoiesis; we also present evidence for the role of fibroblasts (fibroblast-like cells) in the maintenance of hemopoiesis. Experiments on transplantation of bone marrow from several rodent species to syn-, allo- and xenogenic recipients allowed us to study interactions of hemopoietic elements of colonies with cells of the underlying layer.

[Precursor cells of newly formed connective tissue in inflammatory foci in various organs in mice].

The origin of fibroblast-like cells of the capsule around a foreign body in the spleen, liver, peritoneal cavity, subcutaneous connective tissue of mice, the localization of cells-precursors, their proliferative potencies and the ability to migrate through blood were studied using 3H-thymidine autoradiography. Precursors of the inflammation focus cells (irrespective of localization) reproduce intensively outside the limits of intraorganic connective tissue, supposedly, in hemopoietic organs of the bone marrow type and migrate, through the blood channel, into tissues (inflammation foci), where they terminate their differentiation.

[Immunofluorescent research on the species specificity of the antigens of hematopoietic cells in rodents].

The bone marrow cells of mice, rats, guinea pigs, Syrian and dwarf hamsters exhibit a positive immunofluorescence reaction with antisera against insoluble antigens of the bone marrow cells of mice, Syrian and dwarf hamsters and, hence, contain common "cross-reacting" antigens. The use of different methods of antiserum absorption made it possible to reveal, in addition, antigens of "narrow" specificity in (1) mice, Syrian and dwarf hamsters, (2) Syrian and dwarf hamsters, as well as species specific antigens of the bone marrow cells of mice, Syrian and dwarf hamsters.

[NO synthetase in the cells of different tissues and organs of the mouse].

Cellular NO synthase was assayed in bone marrow, spleen, thymus, lymph nodes, epidermis, hypodermic connective tissue, small gut, peritoneal exudate, liver, kidney, and heart by cytochemical and immunofluorescent methods. As a rule the enzymatic activity was found in differentiated cells that finished proliferation (mature hemopoietic cells, epithelium of gut villi, etc.). The enzyme activity was undetectable in actively reproducing low-differentiated cells (basal layer of the epidermis, crypt cells of the small gut, or blast hemopoietic forms). NO is proposed to participate in regulation of tissue-specific terminal differentiation (maturation) of the cells.
