Only those genes of the KIAA1245 gene subfamily that contain HERV(K) LTRs in their introns are transcriptionally active.

Insertion of LTRs into some genome locations might seriously affect regulation of the neighboring genes expression. This hypothesis is widely accepted but, however, not confirmed directly. Earlier, we have identified a family of closely related genes highly similar to the KIAA1245 mRNA counterpart. This family included a subfamily of genes some of which contained and the others lacked an LTR in their structure. We compared transcription of several closely related genes of the subfamily differing in the presence or absence of LTRs. Only LTR-containing genes were transcribed in transformed cell lines, tumorous and embryonic human tissues, whereas LTR-lacking genes remained silent. Since the genes were in the same intracellular microenvironment, we suggested that this effect was most probably due to intrinsic cis-characteristics of integrated LTRs and confirmed this by demonstrating high enhancer activity of KIAA1245 LTRs. The expression of the LTR-containing genes in embryonic tissues might suggest their involvement in evolutionary events during primate speciation.

Changes in the lipid composition of blood plasma and liver in rats induced by severe psychic trauma.

The concentration of high-density lipoprotein cholesterol and triglycerides in rat serum sharply decreased after psychic trauma caused by life hazard. The content of these substances remained unchanged for not less than 1 week after trauma. The concentration of high-density lipoprotein cholesterol was low, while serum content of triglycerides increased 6 weeks after trauma. The concentration of high-density lipoprotein cholesterol significantly decreased after repeated psychic trauma. These changes were accompanied by a sharp increase in the concentration of triglycerides in the serum. Total cholesterol concentration in the liver decreased under these conditions.

[Immunity in children with aquired cicatricial stenoses of the larynx and cervical trachea. Communication 3. Condition of phagocyting immune system].

Macrophagal-phagocyting immunity was studied in 59 children with cicatricial acquired stenosis of the larynx and cervical trachea aged 1 year 9 months to 14 years 8 months. It was found that children with acquired cicatricial stenosis had a sharp depression of all functions of the cells of the macrophagal-phagocyting system (chemotaxis, absorptive, digestive, oxidation-reduction) as well as reduction of their number. This is, on the one hand, a defense reaction of the organism to continuous antigenic stimulation; on the other hand, this marks feasibility of progressive immunocomplex and/or autoimmune processes. The most prominent alterations were seen in patients with concurrent affection of the CNS.

[Atherogenic changes of blood lipids and their components in patients after ischemic stroke].

The aim of the study is to analyze frequency and character of blood lipids abnormalities in patients after ischemic stroke (IS). Blood lipids were studied in 54 patients who survived ischemic stroke at 38-70 years. Atherogenic dyslipidemia was found in 49 (90.7%) patients, 37% had a selective low level of HDL cholesterol (< 37 mg/dl). Patients with dyslipidemia, IV type, (27.8%) showed a high level of triglycerides (413 +/- 72mg/dl) and patients with IIa or IIb types--LDLP cholesterol (223 +/- 12.5 mg/dl). A complete analysis of blood lipids allows detecting atherogenic dislipedimia in the majority of patients who survived ischemic stroke (90.7%), which can be treated with statins or fibrates. An adequate therapy of such patients may significantly improve prognosis for those who survived ischemic stroke and, in some cases, prevent its development.

[Nucleotide sequences of long terminal repeats of the human endogenous retrovirus (LTR HERV-K) on the short arm of chromosome 7: identification, analysis and evaluation of transcriptional activity].

Six clones containing long terminal repeat (LTR) sequences of human endogenous retrovirus of the HERV-K family were found in the YAC library (1200 kb) of the short arm of human chromosome 7. The sequence sizes of the three clones corresponded to the full-length LTR (969 bp). The LTR localization was determined using FISH and verified by comparison with the GenBank database. All three DNA fragments containing solitary LTRs were transcribed in normal germline cells (testicular parenchyma tissue). The differences in the expression of these clones in the germline tumor cells (seminoma) were observed.

[Immunity in children with acquired scarred stenosis of the larynx and cervical trachea].

Humoral immunity was studied in 63 children with acquired laryngotracheal stenosis 1 year 9 months to 14 years 8 months of age. The study discovered humoral immunity dysfunction which was most pronounced in patients who developed scary stenosis after critical care for acute stenotic laryngotracheobronchitis. It was found that if a child has allergy (atopy) in conduction of critical care requiring long-term nasotracheal intubation, he/she is at risk to develop scary stenosis.

[Immunity in children with acquired scarry stenosis of the larynx and cervical trachea. Communication 2. The status of T-cell immunity].

T-cell immunity was studied in 59 children (39 boys-66.1% and 20 girls-33.9%) with acquired stenosis of the larynx and cervical trachea at the age of 1 year 9 months to 14 years 8 months. Compromised T-cell immunity was revealed in the examinees, especially in those who had acute infectious-inflammatory diseases or CNS damage before critical care responsible for development of the stenosis. These changes in T-cell immunity should be considered as a factor of risk to develop immune and autoimmune diseases.

[Localization of vessel lesions in arteriosclerosis and blood lipid composition].

AIM: To investigate correlations between composition of plasmic lipid fractions in patients with ischemic heart disease (IHD) and those with cerebrovascular insufficiency (CVI) caused by atherosclerosis. MATERIAL AND METHODS: 75 patients were divided into three groups; 26 patients with IHD free of CVI (group 1), 22 patients with CVI free of IHD (group 2), 27 patients with IHD and CVI (group 3). Blood lipids were measured by a standard mesiautomatic method using Technicon-AA-2 unit (USA). RESULTS: Hyperlipidemia (HLE) type II was most frequent in group 1 while HLE type IV or hypoalphalipoproteinemia without rise in cholesterol or triglycerides--in groups 2 and 3. CONCLUSION: In IHD without CVI dyslipidemia in most cases was associated with one of the additional risk factors (hypertension, smoking, diabetes mellitus) while in CVI it combined with two or three additional risk factors.

[Chlamydia infections in children with acquired cicatricial laryngeal and tracheal stenosis].

The examination for Chlamydial infection was carried out in 49 children aged from 1 year 10 months to 15 years (mean age 9 years 6 months +/- 3 years 8 months) with laryngotracheal cicatricial stenosis (CS) provoked by long nasotracheal intubation. Antibodies to C. pneumoniae in diagnostically significant titers were detected in 13 of 49 children (26.5%), to C. trachomatis--in 1 patient who had both infections. All the infected patients except the latter carried tracheal canules or were decanulated. Age of the patients with Chlamydial infection at the time of examination and development of CS was significantly older than of non-infected children. No significant difference was found between these patients by duration of the intubation and of the disease. Consideration of the fact that 12 of 13 children with C. pneumoniae infection carried tracheocanules suggests that the infection may be secondary.

Solitary human endogenous retroviruses-K LTRs retain transcriptional activity in vivo, the mode of which is different in different cell types.

Solitary long terminal repeats (LTRs) of human endogenous retroviruses (HERVs), tens of thousands of which are spread all over the genome, contain a variety of potential transcription regulatory elements. Information on transcriptional behavior of individual solitary LTRs, however, is limited. We studied the transcriptional activity of several individual HERV-K LTRs in a variety of tissues and cell lines. The RT-PCR technique targeted at specific amplification of the U3 or U5 regions of individual LTRs together with their unique genomic flanks was used to estimate the content of each region in the transcripts. An unequal abundance of the U3 and U5 regions of the transcripts of the same LTR in different cells and tumors was observed. Each LTR is transcribed differently in different cells or tissues, and transcriptional behavior of different LTRs was different in the same cell line or tissue. The transcriptional status of LTRs varies in response to mitogenic and stress factors and in tumor tissues compared to normal counterparts. The LTRs thus seem to be the subjects of specific transcription regulation. The data obtained indicate that an appreciable fraction of the LTRs retained regulatory potential throughout millions of years of evolution and thus may contribute to the overall transcription regulatory network.

Differences in HERV-K LTR insertions in orthologous loci of humans and great apes.

The classification of the long terminal repeats (LTRs) of the human endogenous retrovirus HERV-K (HML-2) family was refined according to diagnostic differences between the LTR sequences. The mutation rate was estimated to be approximately equal for LTRs belonging to different families and branches of human endogenous retroviruses (HERVs). An average mutation rate value was calculated based on differences between LTRs of the same HERV and was found to be 0.13% per million years (Myr). Using this value, the ages of different LTR groups belonging to the LTR HML-2 subfamily were found to vary from 3 to 50Myr. Orthologous potential LTR-containing loci from different primate species were PCR amplified using primers corresponding to the genomic sequences flanking LTR integration sites. This allowed us to calculate the phylogenetic times of LTR integrations in primate lineages in the course of the evolution and to demonstrate that they are in good agreement with the LTR ages calculated from the mutation rates. Human-specific integrations for some very young LTRs were demonstrated. The possibility of LTRs and HERVs involvement in the evolution of primates is discussed.

Efficient human IFN-gamma expression in the mammary gland of transgenic mice.

Two hybrid genes (BLG-HuIFN-gamma2 and BLG-HuIFN-gamma3) were constructed on the basis of sheep beta-lactoglobulin (BLG) and human interferon-gamma (HuIFN-gamma) gene sequences. They were used to direct HuIFN-gamma synthesis in the mammary gland of transgenic mice. HuIFN-gamma was efficiently produced in the mammary gland of transgenic mice. BLG-HuIFN-gamma2 transgenic females expressed HuIFN-gamma in the milk at concentrations up to 570 mg/ml, and BLG-HuIFN-gamma3 transgenic females expressed up to 350 mg/ml. All females carrying the BLG-HuIFN-gamma3 gene expressed HuIFN-gamma in their milk. No significant changes were observed in the HuIFN-gamma expression level during the lactation period. Using RT-PCR analysis, ectopic expression for both hybrid genes was found in transgenic mice. Despite ectopic expression of HuIFN-gamma in transgenic mice, their development and pregnancy were normal. The heritability of the HuIFN-gamma expression level in milk was demonstrated up to the F2 generation. This work demonstrates that hybrid genes have the potential to develop in transgenic domestic animals producing HuIFN-gamma in milk.

[The effect of different concentrations of dimexide and ortofen on the T-cell and neutrophil components of immunity].

Clinical and immunological efficacy of 2.5% orthophene and 50% dimexide used to arrest an acute inflammatory process in the periodontium is assessed. Three types of salivary neutrophils are distinguished, similar to previously detected types of response of blood immunocompetent cells. Both drugs had a favorable impact on the functional activity of blood T-lymphocytes, blood and salivary neutrophils and are recommended for patients with types I and II reactions of immunocompetent cells of the blood.

[Comparative study of peripheral circulation, lipid metabolism and hemocoagulation in families with atherosclerosis and diabetes mellitus].

AIM: The study of peripheral circulation (PC) in patients with atherosclerosis (AS), diabetes mellitus (DM) type I and II as well as their close relatives to clarify correlations between circulatory disorders, disturbances of lipid metabolism (LM) and hemocoagulation. MATERIALS AND METHODS: The patients and their relatives were divided into 3 groups: with AS, DM type I and II. Individuals without relevant hereditary predisposition served control. A total of 564 subjects were examined. Thermography (TG), TV capillaroscopy of the nail bed (TVCNB), measurements of total blood cholesterol (TBC), high density lipoproteins (HDLP) cholesterol, triglycerides, coagulation time, plasma recalcification, free heparin, fibrinogen concentration, calculation of the atherogenic and prothrombin indices were conducted. RESULTS: Microcirculation suffered most of all, especially in patients and their relatives with AS and DM type I. TVCNB detected morphological changes of the capillaries confirmed by TG. DM patients and their relatives, especially those with DM type I had most distinct morphological shifts. CONCLUSION: The above PC disorders result from lipid metabolism and hemocoagulation abnormalities affecting blood rheology. In DM patients and their relatives especially in those with DM type I specific alterations of the capillary structure in DM and hereditary predisposition to DM are a contributing factor.

Human gamma-interferon expression in the mammary gland of transgenic mice.

Transgenic mice carrying a hybrid gene consisting of ovine beta-lactoglobulin gene sequences and human gamma-interferon (hIFN-g) cDNA were produced. hIFN-g expression in the mammary gland of two lactating transgenic founder females was found. The concentration of active hIFN-g in the milk was estimated as being ca. 1800 IU/ml. The hIFN-g ability to express in the mammary gland was found in the progeny of transgenic founder male.
