[Controlled release antigen from the injection site--new model for a single-shot vaccination?].

Continuously increasing number of available vaccines and a wide use of mass vaccination campaigns lead to the development of simpler methods of vaccine delivery. There is the WHO-sponsored project, including several scientific groups, which aims to reduce the number of repeated administrations of vaccines required for long-term protection and to obviate the need for booster immunization. Such a task may be achieved by the use of controlled-release parenteral systems. One such delivery system comprises microspheres constructed from biodegradable, biocompatible polymers based on poly lactic/glycolic acid. Recent developments of a single-shot tetanus toxoid, encapsulated in the biodegradable polymeric microspheres or microcapsules are reviewed. There are promising advances in the WHO-sponsored project. However, further efforts are needed to elaborate an affective late release of antigen and to obtain a strong booster effect in vaccinated animals.

[Examination of protein MPB 70 properties in BCG substrains used for the production of Polish antituberculosis vaccines].

The aim of the study was examination of MPB70 protein production by different BCG substrains and testing this protein as reagent in allergenic skin test on BCG vaccinated guinea pigs. Three BCG substrains: Danish 1331 (D), Japanese 172 (J) and Polish BCG Moreau (P) were used for the study. The protein MPB70 received from dr Nagai from Osaka University was used as reference preparation. It has been shown that MPB70 protein of molecular weight about 21,500 Da was present only in BCG Moreau (P) and Japanese (J) substrains. In immunoblotting test monoclonal antibodies reacted additionally with 43,000 Da protein in all tested substrains. We suspect that the detected additional molecule was a dimer of MPB70 protein. The development of skin reactions to MPB70 protein was seen in guinea pigs vaccinated with J and P substrains. The peak of alergy to MPB70 protein was observed 9 weeks after vaccination. In sera of vaccinated animals antibodies against MPB70 protein were detected by ELISA method. The possibility of using MPB70 protein for diagnostic purpose is discussed.

[Evaluation of antibacterial and antitoxic activity of normal human immunoglobulin for intravenous use (IVIG)].

In the present study, human immunoglobulins for intravenous use (IVIG), were tested for contents of diphtheria antibodies, antistreptolysin O, antistaphylolysin and antibody level of endotoxin of B. pertussis, enterobacterial common antigen and group B Streptococci. It was shown that all examined immunoglobulin preparation contained antibodies against all tested antigens. Our investigation revealed differences between IVIG preparations and IVIG lots. Basing on these results, we know that the biological activity of Bioglobulin is the same as biological activity of other IVIG preparations produced by foreign manufactures.

[Usefulness of the cytotoxic test for testing levels of diphtheria antibodies in sera of immunized animals].

Vero cells method was used for determination of antibodies level in humans vaccinated with Td vaccine and also in animals immunized with Di, DiTe, Td and DTP vaccines. We have found quite high levels of antibodies in both groups of animals. There was also a significant increase of antibody level in the human sera after Td booster dose in the CCV and HA tests, but the correlation with antitoxin titre was low. CCV test could be useful for determination of immunogenicity of diphtheria toxid.

[Polysaccharide vaccines. II. Meningococcal vaccines].

A short history of vaccination and troubles connected with preparation of vaccine effective in preventing serogroup B meningococcal diseases was described. Different kinds of meningococcal vaccines used all over the world and epidemiological situation in Poland was also discussed.

[Evaluation of the usefulness of the antibody binding test for the estimation of the potency of the rabies vaccine].

Two methods of the estimation of the potency rabies vaccine were used: in vivo NIH Test and in vitro Antibody Binding Test (AB-Test). Six series of vaccines were studied, they passed NIH Test. The results of AB-test were comparable with the results of NIH Test. The AB-Test could be used instead NIH Test when the vaccine must be released in short time.

Evaluation Studies as Topic↗

[Evaluation of safety and immunogenicity of the Td vaccine in laboratory studies].

The new vaccine recommended as the booster dose contains in 0.5 ml: 10 BU Te-toxoid, 2 Lf Di-toxoid, 4.5-0.5 mg Al and 0.01% merthiolate. In 7 consecutive series it was state, the vaccine was safety and stable. The potency was 40-50 JO for Te and 30-40 for Di. The last series was recommended for vaccination under clinical control.

[Investigation of immunogenic properties of diphtheria antitoxin by intradermal active test of potency].

Usefulness of four variants of intradermal test for routine testing of protective potency and diphtheria anatoxin was studied. Comparing with classical active test it was found that full intradermal test possesses data system and results comparable to the classic test and may replace it. This in effect results in four-fold reduction of guinea pig number necessary for performance of the investigation. As orientating test, estimate determining AnaDi potency a single dose performed on six guinea pigs was selected. In the intradermal test the time of investigation, as compared with the classic test, may be shortened by one week.

[A rapid method of measuring the level of aluminum in biological preparations using aluminon].

The aim of this study was to adapt aluminon for determination of Al3+ content in biopreparations adsorbed on Al(OH)3. Aluminum + aluminon complex was identified by spectrophotometry at A535. It was found that the method applied allows to obtain reproducible results. Its sensitivity for Al3+ contains between 85 to 680 micrograms. This method is less laborious in comparison with so far used versenian method.

[Migration of Al(OH)3 in the bodies of experimental animals].

The aim of this study was to observe aluminium hydroxide distribution in mice given this substance subcutaneously and intraperitoneally in various concentrations (0.03 mg-0.8 mg of Al3+ per dose). Each experimental group consisted of 5-6 mice of which brain, liver and kidney were isolated after 7, 14, 28, and 35 days after injection. Control group was composed of unvaccinated mice. Sera of animals vaccinated subcutaneously with the following doses: 0.2 mg, 0.4 mg and 0.8 mg were also tested. A distinct accumulation of Al3+ in liver after intraperitoneal injection with a tendency of rising after 28 days of observation was seen. The same was observed in kidneys after subcutaneous injection, especially doses of 0.4 mg Al3+ and 0.8 mg Al3+. The aluminium content in sera was high in an early period of observation only, and subsequently its elimination was fast.
