Characterization of cationic diarylethene by electron spin resonance and absorption spectra-ratio of open/closed-ring isomers.

Electrochemical cyclization/cycloreversion reactions of a diarylethene, 1,2-bis(3-methyl-2-thienyl)perfluorocyclopentene, are examined experimentally by electron spin resonance (ESR) and absorption spectra. To understand the ESR spectrum, the hyperfine coupling constants are calculated by the density functional theory (DFT) with the B3LYP exchange-correlation functional. The averaged values of the hyperfine coupling constants are approximated by imposing the C(2) symmetry on the structure of the diarylethene. We found that the spectral width of the ESR is significantly different between the open- and closed-ring isomers. This is due to the difference in the pi-conjugation between two isomers. The ESR spectral width analysis could, thus, be used to identify the isomerization of the radical species, which involve the change of the pi-conjugation. The experimentally observed spectrum is found to be the mixture of the open- and closed-ring isomers of the diarylethene. The excitation energies of the cationic diarylethenes are further identified by the SAC-CI calculations.

Effects of amiodarone on electrical and structural remodeling induced in a canine rapid pacing-induced persistent atrial fibrillation model.

Structural in addition to electrical remodeling may be induced by persistent atrial fibrillation per se and make atrial fibrillation refractory to antiarrhythmic drug therapy. Matrix metalloproteinases (MMPs) contribute to structural remodeling in the interstitial space. Amiodarone is effective in treating persistent atrial fibrillation compared with other antiarrhythmic drugs. In mongrel dogs, right atrial pacing at 540 beats/min (bpm) was performed along with ventricular pacing at 100 bpm for 6 weeks after atrioventricular node ablation. Right atrial pacing at 400 bpm was continued for 4 weeks with (n=5) or without (n=5) oral amiodarone (30 mg/kg/day). In sham dogs, only ventricular pacing was done with (n=4) or without (n=6) amiodarone. In atrial pacing without amiodarone group, electrical remodeling characterized by monophasic action potential duration shortening, loss of action potential duration-rate adaptation and depressed conduction velocity and structural remodeling characterized by slightly but significantly increased interstitial fibrosis and enhanced MMP-2 activity compared with sham group were observed, and sustained atrial fibrillation was easily induced. In atrial pacing with amiodarone group, both electrical and structural remodeling were reversed and sustained atrial fibrillation was not induced. In sham group with amiodarone, action potential duration prolongation and depressed conduction velocity compared with sham without amiodarone were observed, but either increased fibrosis or enhanced MMP-2 activity was not observed. Not only electrical but structural remodeling were induced in a canine persistent atrial fibrillation model. Amiodarone reversed both of them, which may be related to its high efficacy in preventing recurrence of persistent atrial fibrillation.

Acute pulmonary embolism after an earthquake in Japan.

There have been no reports on acute pulmonary embolism (APE) after earthquakes. Our aim was to clarify the actual the occurrence of APE following the 2004 Mid Niigata Prefecture earthquake in Japan, and to assess the risk factors for APE after the event. We sent questionnaires to 122 hospitals in the Niigata Prefecture after the earthquake. Cities, towns, and villages in the prefecture were classified into two areas (high evacuee rate area, and low evacuee rate area) due to the mean ratio of evacuees to the overall population during 1 week immediately after the earthquake. A rate of 5% and higher was encountered for the high evacuee rate area and a rate of < 5% was encountered for the low evacuee rate area. Ten out-of-hospital cases of APE (seven in the high evacuee rate area and three in the low evacuee rate area) were diagnosed within the first month after the earthquake. The relative risk of APE was high in the high evacuee rate area (13.09; P = 0.0002) and also higher in women (8.55; P = 0.04). All patients in the high evacuee rate area had stayed in their automobiles for long periods of time, but none had done so in the low evacuee rate area ( P = 0.008).

Cancer and pulmonary embolism: thrombotic embolism, tumor embolism, and tumor invasion into a large vein.

BACKGROUND: The specific incidence of thrombotic pulmonary embolism (PE), tumor PE and tumor invasion into large veins according to tumor type and tumor site remains unclear. METHODS AND RESULTS: A total of 65,181 cancer patients were identified from 98,736 postmortem examinations. Thrombotic PE occurred in 2.32% of all cancer patients and comprised 88.6% of the total number of all PE events. The incidence of thrombotic PE was high in those with adenocarcinoma, leukemia and large cell carcinoma, and was low in those with hepatic cell carcinoma. The incidence of PE was high when tumor was present in hematogenous tissue, lungs, ovaries, pancreas and the biliary system, and was low when tumor was present in the liver. The incidence of tumor PE was high with large cell carcinoma, hepatic cell carcinoma and adenocarcinoma, and was also high when tumor was present in the lungs, ovaries, kidneys and liver. There was a significant correlation between the incidence of tumor PE and the incidence of tumor invasion into large veins. CONCLUSION: The incidence of thrombotic PE, tumor PE and tumor invasion into large veins varies significantly according to tumor histopathology and tumor site.

Incidence of pulmonary thromboembolism (PTE) and new guidelines for PTE prophylaxis in Japan.

Incidence of venous thromboembolism (VTE), which had been considered a relatively rare disease in Japan, has been on the increase in recent years as eating habits have become more similar to those of the West. We have investigated the recent incidence data of perioperative pulmonary thromboembolism (PTE) in Japan and have established guidelines for the prevention of VTE. Recommended thromboprophylaxis is early ambulation for low risk group, elastic stocking (ES) or intermittent pneumatic compression (IPC) for moderate risk group, IPC or low dose unfractionated heparin (LDUH) for high risk group, and LDUH + IPC or LDUH + ES for highest risk group. The management fee for PTE prophylaxis was established and covered by health insurance in April 2004. Surprisingly, the incidence of perioperative PTE decreased just after this guideline was issued. After accumulation of further evidence and application of pharmacological agents, such as low molecular weight heparin, we will establish the advanced guidelines in the future.

Ultraviolet Rayleigh-Mie lidar by use of a multicavity Fabry-Perot filter for accurate temperature profiling of the troposphere.

A UV Rayleigh-Mie scattering lidar system at 355 nm has been upgraded for more-accurate temperature profiling of the troposphere by use of a new multicavity Fabry-Perot etalon (MCFPE) filter. The MCFPE filter, which was designed to improve the stability and operational characteristics of the lidar system, has three filter bandpass functions and separates one Mie scattering and two Rayleigh scattering signals from the lidar return signal and simultaneously acts as a laser frequency discriminator to lock the laser frequency. Moreover, a high-resolution grating is employed to block signal interference from Raman scattering and the solar background. A practical lidar system, which features strong system stabilization and high measurement accuracy, has been built, and the performance of the lidar system has been verified by comparison of temperature profiling between the lidar and a radiosonde. Good agreement between the two instrument measurements was obtained in terms of lapse rate and inversion layer height. Statistical temperature errors of less than 1 K up to a height of 3 km are obtainable with 5 min observation time for daytime measurements.

Ultraviolet high-spectral-resolution Doppler lidar for measuring wind field and aerosol optical properties.

An ultraviolet incoherent Doppler lidar that incorporates the high-spectral-resolution (HSR) technique has been developed for measuring the wind field and aerosol optical properties in the troposphere. An injection seeded and tripled Nd:YAG laser at an ultraviolet wavelength of 355 nm was used in the lidar system. The HRS technique can resolve the aerosol Mie backscatter and the molecular Rayleigh backscatter to derive the signal components. By detecting the Mie backscatter, a great increase in the Doppler filter sensitivity was realized compared to the conventional incoherent Doppler lidars that detected the Rayleigh backscatter. The wind velocity distribution in a two-dimensional cross section was measured. By using the HSR technique, multifunction and absolute value measurements were realized for aerosol extinction, and volume backscatter coefficients; the laser beam transmittance, the lidar ratio, and the backscatter ratio are derived from these measurements.

Ultraviolet Rayleigh-Mie lidar with Mie-scattering correction by Fabry-Perot etalons for temperature profiling of the troposphere.

A Rayleigh-Mie-scattering lidar system at an eye-safe 355-nm ultraviolet wavelength that is based on a high-spectral-resolution lidar technique is demonstrated for measuring the vertical temperature profile of the troposphere. Two Rayleigh signals, which determine the atmospheric temperature, are filtered with two Fabry-Perot etalon filters. The filters are located on the same side of the wings of the Rayleigh-scattering spectrum and are optically constructed with a dual-pass optical layout. This configuration achieves a high rejection rate for Mie scattering and reasonable transmission for Rayleigh scattering. The Mie signal is detected with a third Fabry-Perot etalon filter, which is centered at the laser frequency. The filter parameters were optimized by numerical calculation; the results showed a Mie rejection of approximately -45 dB, and Rayleigh transmittance greater than 1% could be achieved for the two Rayleigh channels. A Mie correction method is demonstrated that uses an independent measure of the aerosol scattering to correct the temperature measurements that have been influenced by the aerosols and clouds. Simulations and preliminary experiments have demonstrated that the performance of the dual-pass etalon and Mie correction method is highly effective in practical applications. Simulation results have shown that the temperature errors that are due to noise are less than 1 K up to a height of 4 km for daytime measurement for 300 W m(-2) sr(-1) microm(-1) sky brightness with a lidar system that uses 200 mJ of laser energy, a 3.5-min integration time, and a 25-cm telescope.

Ultraviolet Rayleigh-Mie lidar for daytime-temperature profiling of the troposphere.

A UV Rayleigh-Mie scattering lidar has been developed for daytime measurement of temperature and aerosol optical properties in the troposphere. The transmitter is a narrowband, injection-seeded, pulsed, third-harmonic Nd:YAG laser at an eye-safe wavelength of 355 nm. Two Fabry-Perot etalons (FPEs) with a dual-pass optical layout filter the molecular Rayleigh scattering components spectrally for retrieval of the temperature and provide a high rejection rate for aerosol Mie scattering in excess of 43 dB. The Mie signal is filtered with a third FPE filter for direct profiling of aerosol optical properties. The Mie scattering component in the Rayleigh signals, which will have influence on temperature measurements, is corrected by using a measure of aerosol scattering because of the relative insufficiency of Mie rejection of Rayleigh filters in the presence of dense aerosols or clouds, and the Mie rejection capability of system is thus improved. A narrowband interference filter is incorporated with the FPEs to block solar radiation. Also, the small field of view (0.1 mrad) of the receiver and the UV wavelength used enhance the ability of the lidar to suppress the solar background signal in daytime measurement. The system is relatively compact, with a power-aperture product of 0.18 W m(-2), and has a high sensitivity to temperature change (0.62%/K). Lidar measurements taken under different weather conditions (winter and summer) are demonstrated. Good agreement between the lidar and the radiosonde measurements was obtained in terms of lapse rates and inversions. Statistical temperature errors of less than 1 K up to a height of 2 km are obtainable, with an averaging time of approximately 12 min for daytime measurements.

Three bits eight states photochromic recording and nondestructive readout by using IR light.

A photochromic polymer film containing three different diarylethene derivatives, that is, 1,2-bis(3,5-dimethyl-2-thienyl)perfluorocyclopentene (1), 1,2-bis(2,5-dimethyl-3-thienyl)perfluorocyclopentene (2), and 1,2-bis(2-methyl-5-phenyl-3-thienyl)perfluorocyclopentene (3) was prepared. Upon UV irradiation, the three derivatives changed to their closed-ring isomers having different colors, yellow, red, and blue. They showed different spectra not only in UV/Vis region but also in the IR spectral region. Upon irradiation with visible light of appropriate wavelengths, each closed-ring isomer was selectively bleached, and three bits eight states recording was performed. The eight states could be read out nondestructively by using IR light of appropriate wavenumbers.

Dendritic cells activated by cross-linking B7-DC (PD-L2) block inflammatory airway disease.

BACKGROUND: Allergic asthma is an increasing clinical problem that might be addressed with immunologically based interventions. A novel human antibody that activates human and mouse dendritic cells (DCs) by cross-linking B7-DC has strong immunomodulatory properties and blocks antigen-induced inflammatory lung disease in mice. OBJECTIVE: We sought to evaluate the ability of activated DCs to mediate an antibody-induced protective response against inflammatory lung disease. METHODS: An adoptive transfer strategy was used to test the ability of antibody treatments to activate DCs, inducing a protective response against inflammatory lung disease in mice presensitized to ovalbumin (OVA). After transfer of activated DCs, recipient mice were exposed repeatedly to airway antigen and evaluated for changes in immune reactivity and airway inflammatory disease. RESULTS: Animals presensitized to OVA receiving either systemic treatments with B7-DC cross-linking antibody (XAb) or adoptively transferred antibody-activated DCs were completely protected from airway inflammatory responses normally induced by repeated exposure to OVA. Lymphocytes isolated from spleens or lung-draining lymph nodes of treated animals were highly responsive to OVA and secreted TNF-alpha, IFN-gamma, and IL-10. In contrast, IL-4 was not produced by cells isolated from animals receiving B7-DC XAb. CONCLUSION: Activation of DCs with B7-DC XAb ex vivo before adoptive transfer into presensitized mice is sufficient to protect animals completely from inflammatory lung disease induced by subsequent repeated airway exposure to the offending antigen. This finding is consistent with our hypothesis that in vivo administration of B7-DC XAb modulates the immune response by activating endogenous DCs.

Severe gastrointestinal bleeding during pregnancy in a case of blue rubber bleb nevus syndrome.

Blue rubber bleb nevus syndrome (BRBNS) is a rare disorder characterized by distinctive cutaneous and gastrointestinal venous malformations that usually cause massive or occult gastrointestinal hemorrhage and iron deficiency anemia secondary to the bleeding episodes. Only two reports of BRBNS in pregnant women can be found, according to the MEDLINE search. Both cases did not describe severe complications during pregnancy. This is the first case report of BRBNS with severe intestinal hemorrhage in a pregnant woman. The woman had been symptom-free for 5 years before the pregnancy and remained symptom-free for 2 years after delivery. This suggests that pregnancy induced the hemangiomas of the gastrointestinal tract to bleed. Celiotomy was needed to deal with massive gastrointestinal bleeding. We propose that women with BRBNS should be checked for systemic hemangiomas before pregnancy. However, it is difficult to check the small intestine thoroughly for hemangioma. Therefore, adequate attention needs to be paid to possible gastrointestinal bleeding when caring for patients with this condition. Our experience suggests that when dealing with pregnant women with BRBNS, it is essential to bear in mind the possibility that pregnancy may trigger the symptoms of hemangioma to become manifest, possibly causing life-threatening massive bleeding from the digestive tract.

Exogenous vascular endothelial growth factor can induce preeclampsia-like symptoms in pregnant mice.

It is reported that expression of vascular endothelial growth factor (VEGF) in trophoblasts increases in cases with preeclampsia. Recently, we demonstrated that the lack of cyclin-dependent kinase inhibitor, p57kip2, expression in the fetus and the placenta plays a role in the development of preeclampsia-like symptoms in pregnant mice. Furthermore, we observed that VEGF mRNA and protein levels, especially VEGF (164), were higher and its expression was stronger in placentas of p57kip2-null embryos than in placentas of wild-type embryos. In this study we investigated whether exogenous murine VEGF (164) induced preeclampsia-like symptoms in pregnant mice, and anti-VEGF neutralized antibody could suppress these symptoms. Administration of VEGF induced hypercoagulation in the placental circulation and a significant elevation of systolic blood pressure in pregnant mice. Furthermore, we demonstrated that treatment with anti-VEGF antibody could suppress the hypercoagulability in placenta and the elevation of systolic blood pressure. These data suggest that VEGF is related to the pathophysiology of preeclampsia.

Phosphatidylserine/phosphatidylcholine microvesicles can induce preeclampsia-like changes in pregnant mice.

We established a phosphatidylserine (PS)/phosphatidylcholine (PC) microvesicles-induced preeclampsia-like model in mice. PS/PC were prepared by mixing 80% PC and 20% PS, and suspended in 0.05 M Tris-HCl at a concentration of 10 mg/mL. One hundred microliters of PS/PC (n = 6) and saline as a control (n = 10) were injected in tail veins of Institute of Cancer Research (ICR) mice every day from days 5.5 to 16.5 of pregnancy. Systolic blood pressure (SBP) was measured by means of the tail-cuff method. On day 17.5, the mice were anesthetized by diethyl ether and euthanized with the collapse of the circulation by drawing blood from the heart. The animals were dissected and the fetuses and placentas removed. Fetal weight and placental weight were evaluated. Plasma antithrombin activity (AT), thrombin-antithrombin complex (TAT), platelet counts, and proteinuria were measured on day 17.5. Placentas were fixed in 4% paraformaldehyde for histologic studies. Statistical analysis was evaluated by analysis of variance and Welch's t-test. Mice injected with PS/PC showed a significant elevation in SBP (124 versus 101 mm Hg; p < 0.001), a significant increase in TAT levels (23 versus 6.6 mug/L; p < 0.05), a significant decrease in platelet counts (88 versus 102 x 10 (10)/L; p < 0.05), a decrease in AT, an increase in proteinuria, and a significant reduction in fetal weight (1.2 versus 1.3 g; p < 0.0001) and placental weight (0.13 versus 0.15 g; p < 0.001), compared with controls. Placentas of mice injected with PS/PC showed diffuse fibrin depositions in the labyrinth layer. We have demonstrated that the artificial PS/PC vesicles induce intrauterine growth restriction with elevations of SBP. The elevation of plasma TAT and the diffuse fibrin depositions in the placentas indicate enhanced thrombin formation, and the significant elevations of SBP indicate preeclampsia-like changes that can be induced by hypercoagulation in the placenta.

Des-gamma-carboxyprothrombin (PIVKA-II) levels in maternal serum throughout gestation.

The status of vitamin K in pregnant women was investigated using the highly sensitive method for des-gamma-carboxyprothrombin (protein induced by vitamin K absence [PIVKA-II]), electrochemiluminescence immunoassay. A gradual elevation of PIVKA-II related to gestational weeks was observed in healthy pregnant women, suggesting that a modest vitamin K deficiency takes place in gestation. Furthermore, throughout gestation the majority of pregnant women exceeded the healthy adult levels in PIVKA-II. Among complicated gestations of preeclampsia, a remarkable elevation of PIVKA-II was observed in severe preeclampsia, in which a high correlation between PIVKA-II level and coagulation parameters, including thrombin-antithrombin (TAT) complexes, was revealed. These data were suggestive that the vitamin K status readily decreased into a deficient status in hypercoagulative conditions. Among other complicated gestations, a moderate elevation of PIVKA-II was demonstrated in hyperemetic conditions, and, if at all, only a slight elevation of PIVKA-II was observed in other maternal diseases. The present study is the first report regarding the changes of PIVKA-II in pregnant women.

"V-H-A Pattern" as a criterion for the differential diagnosis of atypical AV nodal reentrant tachycardia from AV reciprocating tachycardia.

BACKGROUND: During ventricular extrastimulation, His bundle potential (H) following ventricular (V) and followed by atrial potentials (A), i.e., V-H-A, is observed in the His bundle electrogram when ventriculo-atrial (VA) conduction occurs via the normal conduction system. We examined the diagnostic value of V-H-A for atypical form of atrioventricular nodal reentrant tachycardia (AVNRT), which showed the earliest atrial activation site at the posterior paraseptal region during the tachycardia. METHODS: We prospectively examined the response of VA conduction to ventricular extrastimulation during basic drive pacing performed during sinus rhythm in 16 patients with atypical AVNRT masquerading atrioventricular reciprocating tachycardia (AVRT) utilizing a posterior paraseptal accessory pathway and 21 with AVRT utilizing a posterior paraseptal accessory pathway. Long RP' tachycardia with RP'/RR > 0.5 was excluded. The incidences of V-H-A and dual AV nodal physiology (DP) were compared between atypical AVNRT and AVRT. RESULTS: V-H-A was demonstrated in all the 16 patients (100%) in atypical AVNRT and in only 1 of the 21 (5%) in AVRT (P < 0.001). DP was demonstrated in 10 patients (63%) in atypical AVNRT and in 4 (19%) in AVRT (P < 0.05). The sensitivity of V-H-A for atypical AVNRT was higher than that of DP (P < 0.05). Positive and negative predictive values were 94% and 100%, respectively, for V-H-A and 71% and 74%, respectively, for DP. CONCLUSIONS: The appearance of V-H-A during ventricular extrastimulation is a simple criterion for differentiating atypical AVNRT masquerading AVRT from AVRT utilizing a posterior paraseptal accessory pathway.

Atomic hydrogen emission induced by TEA CO(2) laser bombardment on solid samples at low pressure and its analytical application.

Hydrogen emission has been studied in laser plasmas by focusing a TEA CO(2) laser (10.6 microm, 500 mJ, 200 ns) on various types of samples, such as glass, quartz, black plastic sheet, and oil on copper plate sub-target. It was found that H(alpha) emission with a narrow spectral width occurs with high efficiency when the laser plasma is produced in the low-pressure region. On the contrary, the conventional well-known laser-induced breakdown spectroscopy (LIBS), which is usually carried out at atmospheric air pressure, cannot be applied to the analysis of hydrogen as an impurity. By combining low-pressure laser-induced plasma spectroscopy with laser surface cleaning, a preliminary quantitative analysis was made on zircaloy pipe samples intentionally doped with hydrogen. As a result, a good linear relationship was obtained between H(alpha) emission intensity and its concentration.
