Molecular cloning and expression of bottlenose dolphin (Tursiops truncatus) interleukin-8.

The bottlenose dolphin interleukin (IL)-8 cDNA was molecularly cloned. The dolphin IL-8 has an open reading frame of 303-bp encoding 101 amino acids. The homology of the amino acid sequence with that of other species was: sheep, 89.1%; cattle, 88.1%; pig, 85.1%; dog, 85.1%; horse, 79.2%; human, 74.5%; and macaque, 72.3%. The amino acid sequence suggested that dolphin IL-8 was a CXC chemokine. The recombinant dolphin IL-8 protein was recognized with anti-ovine IL-8 monoclonal antibody.

Development of a method of diagnosing ovarian disorders in sows and gilts using uterine ultrasonography.

In ultrasonographic diagnosis of ovarian disorders and the estrous cycle in sows, transverse observation of the uterus yielded more characteristic findings than observation of sagittal sections. Transverse ultrasonography revealed that the low progesterone (P) type uterus showed a round structure, while the high P type uterus showed a flattened structure. These results corresponded well with rectal palpation findings: the low P type uterus had a hard, pipe-like structure and the high P type a soft, balloon-like structure. For gilts, we employed a minimum convex type transrectal prostate probe that had an approximately 18 cm insertion handle. The images of the uterus obtained thereby were a similar to those obtained from sows. The above results suggest that it should be possible to diagnose and treat many ovarian disorders in sows and gilts based only on the ultrasonograhic findings. In short, ultrasonograhic findings of a round structure of the uterine wall might be an indication for PMSG (pregnant mare serum gonadotropin) treatment, while findings of a flattened structure might be an indication for PGF(2) (alpha) administration.

Molecular cloning of equine muscle-type phosphofructokinase cDNA.

The complete coding region sequence of equine muscle-type phosphofructokinase (ePFKM) was obtained from skeletal muscle of a thoroughbred horse. The deduced amino acid sequence of ePFKM showed 97%, 96%, 96%, 96% and 95% identity to canine, human, mouse, rabbit and rat PFKM, respectively. The amino and carboxyl terminal halves of ePFKM presented a structure of tandem repeat, as other mammalian PFKMs. As the amino acid residues constituting various ligand-binding sites were also conserved, it is thought that ePFKM has enzymatic activity similar to PFKM in other mammals.

Alteration of ventricular function on echocardiography associated with intra-pericardial disorder in dogs.

Two dogs with severe exercise intolerance (Cases 1 and 2) and another dog with cardiac dilation (Case 3) were referred to the Nihon University Animal Medical Center (ANMEC). Case 1 was diagnosed as pericardial effusion (PE), Case 2 as pericardial hemorrhage, and Case 3 as pericardiophrenic hernia. When these causative disorders were removed, the heart expanded, and clinical symptoms markedly improved in these three dogs. In particular, the cardiac chamber diameters and left ventricle fractional shortening (LVFS) was normalized in all 3 dogs postoperatively. There is only one case report that compares before effusion extractions in the pericardial sac with the after echocardiography findings. In this paper, echocardiography was conducted on the three endocardial disease cases, comparing before removing these causative disorders with the findings after echocardiography.

A cell line assay system for predicting the response of human blood to endotoxin.

Some parenteral drugs augment the in vivo action of endotoxin. It is necessary to regulate the overall toxic action of contaminating endotoxin by developing a clinically relevant test method for the safety control of such drugs. Although the responses of human peripheral blood cells (hPBC) to endotoxins to produce tumour necrosis factor alpha (TNF-alpha), interleukin 6 (IL-6), and IL-1beta showed considerable variation depending on the endotoxin and also on the individuals used as sources of blood, the responses to each of the endotoxins evaluated relative to that to the Japanese reference standard endotoxin were found to be highly reproducible irrespective of the sources of hPBC. The evaluation procedure based on the relative responsiveness to various endotoxins was shown to be highly effective to detect differences in responsiveness among the endotoxin test, the pyrogen test, and the cytokine induction in hPBC. When eight human monocytoid cell lines were examined, only THP-1 and 28SC cells showed a significant dose-dependent IL-6 production. However, THP-1 failed to show consistency with hPBC in responses to the panel of endotoxins. 28SC cells showed appropriate consistency with hPBC not only in terms of respective responses to the endotoxins but also with regard to detection of the effect of human interferons to augment endotoxin to induce IL-6.

Diagnosis of equine piroplasmosis in Xinjiang province of China by the enzyme-linked immunosorbent assays using recombinant antigens.

The prevalence of equine piroplasmosis in Xinjiang province, China, was examined by enzyme-linked immunosorbent assays (ELISAs). A total of 70 serum samples were taken from horses pastured on three farms in western Xinjiang, and examined for diagnosis of equine Babesia equi (B. equi) infection and B. caballi infection by ELISAs using recombinant equi merozoite antigen 1 (EMA-1) and recombinant P48 antigen, respectively. Of the 70 samples, 28 (40.0%) and 17 (24.3%) samples were positive for B. equi infection and B. caballi infection, respectively. In addition, 11 (15.7%) samples were positive for both B. equi and B. caballi infections. These results indicate that equine piroplasmosis is widespread and therefore a cause for serious concern in western Xinjiang. To our knowledge, this is the first report describing a survey on equine piroplasmosis in Xinjiang province, China.

Priming of dolphin neutrophil respiratory burst by recombinant tumor necrosis factor alpha.

We studied the effects of recombinant dolphin tumor necrosis factor alpha (rdoTNFalpha) on the respiratory burst activity of dolphin neutrophils. rdoTNFalpha enhanced the luminol-dependent chemiluminescence response of dolphin neutrophils induced by concanavalin-A, opsonized zymosan, and heated plasma, but not that induced by phorbol myristate acetate. The TNF-associated priming activity was concentration- and preincubation time-dependent, and heat-instable. These data suggest that, as in human neutrophils, TNFalpha enhances the respiratory burst in dolphin neutrophils that follows short-term incubation with various receptor-mediated agonists.

Serological investigation of Bartonella henselae infections in clinically cat-scratch disease-suspected patients, patients with cardiovascular diseases, and healthy veterinary students in Japan.

Seroprevalence of Bartonella henselae was investigated in Japan in 48 individuals clinically suspected of having cat-scratch disease (CSD), 159 patients with cardiovascular diseases, and 129 healthy veterinary students. Of 48 CSD-suspected patients examined, 19 (39.6%) were positive for B. henselae-IgG and 4 (8.3%) for B. henselae-IgM. Of 159 patients with cardiovascular diseases, 5 (3.1%) were positive for B. henselae-IgG. In healthy veterinary students, 14 of 129 (10.9%) were positive for B. henselae-IgG and 1 (0.8%) for B. henselae-IgM. The positive rates of B. henselae-IgG and -IgM in CSD-suspected patients were significantly higher than in other sources. Most CSD-suspected and healthy individuals who were positive for B. henselae antibody had had some contacts with cats. In CSD-suspected patients, the B. henselae positive rate in females was significantly higher than in males, and high seropositive rates to B. henselae were found in younger age groups.

Bovine milk enhances the oxidative burst activity of polymorphonuclear leukocytes in low concentrations.

Bovine milk contains various immunoreactive components, and the activation of polymorphonuclear leukocytes (PMNLs) function in breast-fed infants has been reported. In this study, the effect of milk on the oxidative burst of bovine PMNLs was investigated in vitro. When PMNLs were incubated with 0.1% colostrum or normal milk, the oxidative burst induced by serum-opsonized Staphylococcus aureus was enhanced, and the enhancement declined dose-dependently. The enhancement of the oxidative burst by milk was not due to opsonins but the priming activities. Also, the phorbol 12-myristate 13-acetate (PMA)-induced oxidative burst increased after incubation with 0.1% colostrum, but the colostral enhancement of the oxidative burst was unaffected by the incubation time. These results suggest that bovine milk contains oxidative burst promoting factor(s).

A canine case of gliosis with cyst formation in the posterior fossa.

A 5-month-old male Great Pyrenees with symptoms of convulsions, circling, and a head tilt was referred to the Animal Medical Center of Nihon University. On a magnetic resonance image (MRI), a cyst in the posterior fossa was noted and a part of the cyst enhanced by gadoteridol. Based on MRI and clinical findings, the patient was tentatively diagnosed with a cyst formation tumor, and an operation to open the cyst and remove the part enhanced by contrast was performed. Postoperatively, the clinical course was good. Pathologically, the removed tissue was diagnosed as a gliosis with cyst formation.

The respiratory burst activity of bottlenose dolphin neutrophils elicited by several stimulants.

To investigate the early host defense function in aquatic animals, the respiratory burst activity of bottlenose dolphin neutrophils against soluble and particulate stimulants was measured by luminol-dependent chemiluminescence assays and compared with those of bovine and human. Dolphin neutrophils generated the respiratory burst in response to phorbol 12-myristate 13-acetate (PMA), concanavalinA (ConA), heated-plasma (HP), and homologous-plasma opsonized zymosan except N-formyl-Met-Leu-Phe (fMLP). However, the respiratory burst of dolphin neutrophils stimulated by lipopolysaccharide and Staphylococcus aureus was inferior to those of bovine and human. Furthermore, DP-OZ also induced the respiratory burst of bovine and human neutrophils. In conclusion, dolphin neutrophils responded to several soluble and particulate stimulants as well as human neutrophils, but were refractory or slightly responded to bacterial agents.
