Coupling-isomerization-N,S-ketene acetal-addition sequences--a three-component approach to highly fluorescent pyrrolo[2,3-b]pyridines, [1,8]naphthyridines, and pyrido[2,3-b]azepines.

Annelated 2-amino pyridines such as pyrrolo[2,3-b]pyridines, [1,8]naphthyridines, and pyrido[2,3-b]azepines can be synthesized in moderate to good yields in a consecutive one-pot three-component process by a coupling-isomerization-enamine-addition-cyclocondensation sequence of an electron-poor (hetero)aryl halide, a terminal propargyl N-tosylamine, and an N,S-ketene acetal. After the coupling-isomerization sequence, a Diels-Alder reaction with inverse electron demand of the intermediate enimine and the N,S-ketene acetal and subsequent aromatization furnish annelated 2-amino pyridines 4 that were unambiguously characterized by numerous X-ray structure analyses. These heterocycles are highly fluorescent and partially pH sensitive, and their electronic structure was studied with spectroscopic and computational methods.

Zn2+ dependent DNA binders based on terminally modified peptide nucleic acids.

Two ligand-intercalator-peptide nucleic acid conjugates (L-NADI-PNAs) have been synthesized. Affinity of these conjugates to their complementary DNAs was found to be affected by Zn(2+). The magnitude of this effect could be controlled by a variation of the ligand. Upon binding Zn(2+) the L-NADI-PNAs form positively charged ZnL complexes, which interact with the negatively charged DNA backbone. This electrostatic interaction stabilizes PNA/DNA duplexes. It has been found that Zn(2+) dependent stabilization takes place only if the ZnL complex has a higher total positive charge than the ligand. Linear correlation has been observed between Zn(2+) induced stabilization of PNA/DNA duplexes and difference of charges of the ZnL complex and the ligand.

Molecular scaffold for the construction of three-armed and cage-like receptors.

An efficient procedure was developed for the synthesis of the C3-symmetric molecular scaffold 2. The latter can easily be converted by a single step into either the three-armed receptors 11-16 or the cage-like receptor 17. X-ray structures were obtained for 2, 11, and 16, which are discussed in regard to their aptitude as receptor platforms. The interaction of the three-armed receptors 12-16 and the cage-like receptor 17 with phloroglucinol was investigated. In accordance with the conclusions obtained from molecular modeling and X-ray crystallographic studies on the host-guest complexes, the three-armed bipyridine receptor 16 exhibits, due to its induced fit, a larger association constant toward phloroglucinol than the cage 17. This new receptor system shows all of the positive features characteristic of 2,4,6-trialkylbenzene receptor systems, such as conformational control by steric gearing, ready availability, and versatility in derivatization. These attributes, combined with the advantageous size of the components, allows this system to be readily tailored to provide receptors for larger, biologically important molecules.

(RS)-6-ethyl 2-carboxy-1,2,3,4-tetrahydroazulene-6-carboxylate.

The title compound, C(14)H(16)O(4), was obtained during the synthesis of 2,6-disubstituted azulene derivatives. In the partially reduced azulene skeleton, the absence of a H atom at the ester substitutent position of the seven-membered ring, as well as lengthened double bonds, indicate a conjugative stabilized system with two overlaid tautomers.

A novel one-pot three-component synthesis of 3-halofurans and sequential Suzuki coupling.

A novel sequence of Sonogashira coupling and electrophilic addition to an ynone, with concomitant deprotection and cyclocondensation, opens a new one-pot synthesis of 3-halofurans; the method can be readily elaborated to a new sequential Sonogashira-addition-cyclocondensation-Suzuki reaction to furnish 2,3,5-trisubstituted furans in a one-pot fashion.

Biphenyl derived oxovanadium(IV) and copper(II) salen-type complexes--structure and redox tuning.

A series of vanadyl(IV) salen (N,N'-bis(salicylidene)ethylenediaminato)-type complexes (1-4) bearing phenyl or 2-hydroxyphenyl moieties have been prepared and characterized by means of mass spectrometry, infra-red, electron paramagnetic resonance (EPR), UV/Vis spectroscopy, cyclovoltammetry and X-ray crystallography. Their structures have been compared to their copper(II) analogs 5-8. Hydrogen intralinkages have been observed in the crystal structure of 5. The pendant hydroxy groups fine-tune the redox properties of the complexes. The catalytic activity in the oxygenation of ethyl phenyl sulfide to the corresponding sulfoxide was investigated. Results indicate that complex 1 bearing hydroxyphenyl subunits and a phenylene bridge is the most selective under these reaction conditions, with the smallest amount of the over-oxidized product, sulfone.

A C3-symmetric molecular scaffold for the construction of large receptors.

A novel C3-symmetric scaffold has been efficiently synthesized exhibiting the property that variable receptor arms can be easily attached by simple alkylation reactions; the utility of the scaffold as a skeleton for large receptors has been examined with a corresponding tris(bipyridyl) derivative toward phloroglucinol.

Synthesis of new boron-rich building blocks for boron neutron capture therapy or energy-filtering transmission electron microscopy.

The synthesis of a new ortho-carborane derivative, tetracarboranylketone 4, is reported here. Ketone 4 was prepared from a tetraalkynylated ketone by the addition of decaborane. The keto group was then easily modified to yield the glycosides 17alpha and 18beta, which contain glucose or galactose, respectively, and the nucleotide 13b. In addition to ketone 4, which is acyclic, cyclic ketone 8 was also synthesised. X-ray diffraction analysis of compound 4 indicated the presence of two toluene guest molecules per molecule of the host compound. Furthermore, compound 4 displays a rather low cytotoxicity. These novel products can be used as building blocks to create a new class of biomolecules containing high-density carborane clusters. Such molecules may constitute powerful tools for applications like Boron Neutron Capture Therapy or Energy-Filtering Transmission Electron Microscopy.

Hexasubstituted donor-acceptor benzenes as nonlinear optically active molecules with multiple charge-transfer transitions.

The synthesis of three novel nonlinear optical (NLO) chromophores with threefold symmetry, namely 1,3,5-tris(4-N,N-diethylaminophenyl)-2,4,6-tris(4-nitrophenyl)benzene (3), 1,3,5-tris(4-N,N-dihexylaminophenylbutadiynyl)-2,4,6-tris(4-nitrophenyl)benzene (13) and 1,3,5-tris(4-N,N-dihexylaminophenylethynyl)-2,4,6-tris(4-nitrophenylethynyl)benzene (4 b), is reported. We used the [Co(2)(CO)(8)]-catalysed trimerisation of 4-N,N-diethylamino-4'-nitrotolane (5) to prepare 3. The trimerisation experiment carried out with 1-(4-N,N-diethylaminophenyl)-6-(4-N,N-nitrophenyl)hexatriyne (6) and [Rh(PPh(3))(3)Cl] afforded 13. A stepwise approach was used to prepare 4 b. 1,3,5-Trichloro-2,4,6-triiodobenzene (8 b) was coupled with 4-nitrophenyl-acetylene (14) under Pd(0) catalysis to yield 1,3,5-trichloro-2,4,6-tris(4-nitrophenylethynyl)benzene (15). The coupling reaction of 15 with 4-N,N-dihexylaminophenylethynyltributylstannane (21) led to 4 b. X-ray investigations on 3, 4 b and 13 confirmed the structural assignments and revealed that the peripheral aryl rings in 4 b are less twisted around the connecting bonds than in 3 and 13. A large second-order polarisability (beta) of 4 b relative to 3 and 13 was determined by hyper-Rayleigh scattering (HRS). Compound 4 b represents an NLO chromophore with second-order polarisabily among the highest obtained so far for two-dimensional nondipolar NLO chromophores.

Location of the H atoms in ammonium persulphate by deuteron NMR. Verification by X-ray diffraction.

It is demonstrated that H atoms can be located by the spectroscopic method of deuteron NMR. The requirement is that the 'heavy-atom' positions are known from diffraction studies. The technique allows an accuracy of the order of 0.01 A. The compound studied is ammonium persulphate (APS), (NH(4))(2)S(2)O(8). APS crystallizes in space group P2(1)/n with lattice parameters a = 6.1340 (2), b = 7.9324 (3), c = 7.7541 (3) A and beta = 94.966 (1) degrees at T = 118 K. In perdeuterated crystals of APS, only one of the deuterons of every ND(4)(+) ion becomes localized at low temperatures. Therefore, most of this work uses samples with 9% deuteration. In such crystals, most of the ammonium ions containing deuterons come in the form of NDH(3)(+) ions. At T < 25 K, the single deuteron of these ions becomes localized in one of four equilibrium sites. The deuteron site occupancies differ from each other and are measured at 17 K. The deuterons are located in three steps. (i) The deuteron quadrupole-coupling (QC) tensors are measured at 17 K. Their unique principal directions are identified, as is well justified, with the N-D bond directions. (ii) The fine structure of a deuteron NMR line is analyzed in terms of the magnetic dipole-dipole interactions between all nuclei in an NDH(3)(+) ion to obtain the N-D and D-H internuclear distances. (iii) An empirical relation between deuteron QC constants and D.O distances in N-D.O hydrogen bonds is exploited to assign the N-D bond vectors to the appropriate N atom of which there are four in the unit cell. The results are highly relevant for an understanding of the complex tunnelling and stochastic reorientation dynamics of the ammonium ions in APS. They are verified by a complementary X-ray diffraction study.
