Rate acceleration in olefin metathesis through a fluorine-ruthenium interaction.

The synthesis, structure, and performance of new ruthenium-based olefin metathesis catalysts, featuring fluorinated NHC ligands are presented. The introduction of halogen atoms into the N-heterocyclic carbene ligand profoundly alters the catalytic activity and can afford a more efficient catalyst. Structural investigations suggest that a fluorine-ruthenium interaction is responsible for this increased activity.

Heterocyclic ring scaffolds as small-molecule cholesterol absorption inhibitors.

Enantio- and diastereoselective syntheses of a substituted oxazolidinone, isoxazoline and pyrazoline as beta-lactam surrogates are described. The substituted heterocycles were designed to incorporate side chains closely resembling those found in the beta-lactam cholesterol absorption inhibitor ezetimibe (1). Additionally, the in vitro inhibitory efficacy of the novel compounds as cholesterol absorption inhibitors is reported using a brush border membrane vesicle assay.

Diastereoselective phenol para-alkylation: access to a cross-conjugated cyclohexadienone en route to resiniferatoxin.

We document a route for the synthesis of a densely functionalized spiro-fused 2,5-cyclohexadienone as an intermediate for the synthesis of resineferatoxin. The strategy is based on an unprecedented diastereoselective, intramolecular phenol para-alkylation to a cross-conjugated cyclohexadienone. In the course of these synthetic studies we developed rapid access to a chiral nitrile possessing a quaternary stereocenter and disclose an unusual acetal rearrangement from a dioxane, which favors the corresponding dioxepane.
