Specific immune responses in workers exposed to benzene.

UNLABELLED: The aim of this study was to elaborate a method for detection of specific IgG antibodies (Abs) to the haptenes p-benzoquinone (p-BQ) and hydroquinone (HQ) for assessment of specific humoral immune responses. Plasma and urine, collected from petrochemical plant workers have been analyzed. The workers were divided into three professional groups in ascending order of benzene exposure. The concentration of benzene in the air was determined by gas chromatography with mass-spectrometry and trans,trans-muconic acid (biomarker of benzene exposure) in urine-by liquid chromatography with UV-detection. Specific IgG Abs to haptenes p-BQ and HQ in plasma were determined with newly developed ELISA. The relationships "exposure-effect," revealed increased levels of specific IgG to haptens correlating with the benzene exposure. The "exposure-response" relationships demonstrated that workers with value of OD over X+2SD were 62% low exposure group, 68% in group with level of exposure on Threshold Limit Value (TLV) and 91% in the highest exposure group. The data obtained show that there is a good correlation between antibody production and the biomarker of exposure t,t-muconic acid. CONCLUSION: The newly developed method is applicable for assessment of specific humoral immune responses in workers exposed to benzene. There was a good correlation between benzene exposure and formation of antibodies against benzene metabolites.

Molecular epidemiology studies of carcinogenic environmental pollutants. Effects of polycyclic aromatic hydrocarbons (PAHs) in environmental pollution on exogenous and oxidative DNA damage.

Exposure to high levels of environmental air pollution is known to be associated with an increased carcinogenic risk. The individual contribution to this risk derived from specific carcinogenic chemicals within the complex mixture of air pollution is less certain, but may be explored by the use of molecular epidemiological techniques. Measurements of biomarkers of exposure, of effect and of susceptibility provide information of potential benefit for epidemiological and cancer risk assessment. The application of such techniques has been mostly concerned in the past with the carcinogenic polycyclic aromatic hydrocarbons (c-PAHs) that are associated with particulate matter in air pollution, and has showed clear evidence of genotoxic effects, such as DNA adducts, chromosome aberrations (CA) and ras oncogene overexpression, in environmentally exposed Czech and Polish populations. We are currently extending these studies by an investigation of populations exposed to environmental pollution in three European countries, Czech Republic, Slovak Republic and Bulgaria. This pays particular attention to PAHs, but also investigates the extent of radically induced (oxidative) DNA damage in the exposed populations. Policemen, bus drivers and controls, who carried personal monitors to determine their exposures to PAHs have been studied, and blood and urine were collected. Antioxidant and dietary status were assessed in these populations. Stationary monitors were also used for ambient air monitoring. Amongst the parameters studied in the biological samples were: (a) exposure biomarkers, such as PAH adducts with DNA, p53 and p21(WAF1) protein levels, (b) oxidative DNA damage, (c) the biological effect of the exposure by measurement of chromosome damage by fluorescence in situ hybridisation (FISH) or conventional methods, and (d) polymorphisms in carcinogen metabolising and DNA repair enzymes. Repair ability was also measured by the Comet assay. In vitro systems are being evaluated to characterise the genotoxicity of the organic compounds adsorbed to air particles.

Comparison of the clinical efficacy of standard and mucoadhesive-based nasal decongestants.

AIMS: To compare two xylometazoline 0.1% preparations: reference commercial solution (RS) and test mucoadhesive solution (TS). METHODS: Twenty subjects with perennial allergic rhinitis (age range 18-69 years, 5 atopic, 7 men) applied randomly in turn TS and RS for 5 days in a double-blind crossover clinical trial. Nasal airflow resistance (NAR), nasal symptoms (6 grade scoring), frequency of application (times/day), and side-effects were recorded. RESULTS: Mean ratio TS/RS of areas under the resistance/time curves for NAR +/- 90% CI: 3.56 +/- 0.92; mean TS-RS differences +/- 95% CI: for congestion: -1.12 +/- 0.59, for frequency of application: -1.10 +/- 0.20. Subjects experienced fewer side-effects with TS. CONCLUSIONS: A mucoadhesive solution with a decongestant had a greater and longer lasting effect on nasal congestion in subjects with perennial allergic rhinitis than the commercially available decongestant solution. It also caused fewer side effects.
