High resolution replication patterns of the human Y chromosome. Intra- and interindividual variation.

Replication studies on prophasic human Y chromosomes reveal 4 early replicating segments in the euchromatic portion. The distal segment of Yp replicates first. After replication of the euchromatic part is almost finished 3 to 5 segments start replication in the heterochromatic portion of Yq. These segments exhibit considerable intraindividual variation with respect to the origin of onset of replication. While the location of these bands--once they are differentiated--is fixed within one individual, the number of these bands varies interindividually.

Homologous early replication patterns of the distal short arms of prometaphasic X and Y chromosomes.

Replication studies on prometaphasic human sex chromosomes reveal a distinct early replicating segment on both distal Xp ad Yp. These segments correspond to high resolution bands Xp22.3 and Yp11.3. The findings demonstrate synchronous replication of these parts of the sex chromosomes and correspond to the comparatively long stretches of Xp and Yp that participate in a synaptonemal complex. Furthermore these observations are compatible with Polani's view that suggests homologous segments with similar genetic information on both sex chromosomes (Polani 1982).

X-linked genes of the H-Y antigen system in the wood lemming (Myopus schisticolor).

H-Y antigen was investigated in 18 specimens representing six different sex chromosome constitutions of the wood lemming (Myopus schisticolor). The control range of H-Y antigen was defined by the sex difference between normal XX females (H-Y negative per definitionem) and normal XY males (H-Y positive, full titer). H-Y antigen titers of the X*Y and X*0 females were in the male control range, while in the X*X and X0 females the titers were intermediary. Data were obtained with two different H-Y antigen assays: the Raji cell cytotoxicity test and the peroxidase-antiperoxidase (PAP) method. Fibroblasts, gonadal cells, and spleen cells were checked. Presence of full titers of H-Y antigen in the absence of testis differentiation is readily explained by the assumption of a deficiency of the gonad-specific receptor of H-Y antigen. Since sex reversal is inherited as an X-linked trait, genes for this receptor are most likely X-linked. The implications of our findings are discussed in connection with earlier findings concerning H-Y antigen in XY gonadal dysgenesis in man and the X0 situation in man and mouse.

The effect of prostaglandins in the rat ovary: specific or unspecific stimulation of cAMP, cGMP and progesterone?

Ovarian membranes of the rat do not specifically bind prostaglandin E2 at any stage of development. Attachment of [3H]prostaglandin E2 in the presence of an excess of unlabelled prostaglandin E2 increased in a linear fashion and was not saturable. Prostaglandin E2 stimulated cAMP accumulation by ovarian cells of all developmental stages. This effect was not saturable. An effect of prostaglandin E2 on cGMP levels was not observed. At high concentrations of prostaglandin E2 an increase of ovarian progesterone synthesis was detected. In comparison hCG stimulated progesterone synthesis at much lower concentrations. PGF2 alpha increased cellular and extracellular cAMP at 1000-fold higher concentrations than PGE2 and did not affect cGMP levels.

Long-term study of virus contamination of surface water in the German Democratic Republic.

Between 1 January 1970 and 31 December 1979, a study of the concentration of viruses in surface water was carried out by 4 virological laboratories in different regions of the German Democratic Republic. All these laboratories used the same methods for virus detection.Altogether 1908 samples from 30 sampling points were evaluated. The rate of virus isolation ranged from 8% to 92% with a mean of 20%. There were considerable differences in isolation rate among the sampling points, and the rate for any particular point varied from year to year. The mean value of virus concentration, determined by the most probable number technique, was 2.7 cytopathogenic units (CU) per litre, while the maximum was 22.1 CU/litre. Viruses seen throughout the 10-year investigation included poliovirus, types 1, 2, and 3, echovirus types 6, 11, and 30, and coxsackievirus B 3 and B 5; echovirus 7 and 24, coxsackievirus B 1, and adenovirus 5 were seen occasionally. The results of the study reflected the high level of use of surface waters in the German Democratic Republic.Where water is intended for human use, e.g., as drinking-water or for recreation, reasonable safety measures, such as water treatment and disinfection, should be taken, in order to ensure that the level of viral contamination is within the permissible limits.

[Indications of valvular pulmonary stenosis on plain chest films (author's transl)].

Plain chest films of 87 children aged 4-14 years with valvular pulmonary stenosis proved by cardiac catheterization and of an age matched control group of 92 healthy children were evaluated in a retrospective study. Up to 4 years the evaluation was impaired by the overlying thymus. The evidence of poststenotic dilatation of the pulmonary trunk and the left pulmonary artery branch together with a normal or increased left sided perihilar pulmonary vascularity is essential for the diagnosis of valvular pulmonary stenosis. The pulmonary trunk showed marked dilatation in 47 cases, mild dilatation in 11 cases and no dilatation in 21 cases. A cranial displacement of the pulmonary trunk which partially reached or overlapped the aortic knuckle was seen in 44 children. 50 children showed a hilar enlargement due to the poststenotic dilatation of the left pulmonary artery branch. 1/5 of the children showed extension of the poststenotic enlargement into the left sided segmental artery branches. Cardiac size, calculated by heart volume, was within age specific limits. No correlation was found concerning the severity of the stenosis and the radiographic findings. Indications of valvular pulmonary stenosis increased with the age of the child.

[Resection therapy of bronchogenic carcinoma in old age].

In a 20 year period from 1959 to 1979, 218 resections were performed for primary cancer of the lung in patients aged 70 and more. The postoperative mortality was 5% (11 cases). Long-term results could be evaluated for 160 cases operated before 1975. A 5 years survival rate could be established after pneumonectomy for 10.7%, after lobectomy for 39.6% and after wedge lobe resection for 25%. Staging by TNM-system 5 years survival rate in St.I/T 1 was 45.1%, St.II/T1/T2 33.3% and St.III/T1-T3 15.5%. Before opening the chest, the histological diagnosis of bronchial carcinoma could be defined by bronchoscopic findings in 81.8%. In cases of primary cancer of the lung without clinical symptoms, resections were done successfully at an age of 70 years and more.

[Mycobacterium malmoense in the Federal Republic of Germany].

For the first time M. malmoense was demonstrated in Sweden, thereafter in Wales (GB) and Finland. Lately we have also found lung diseases in Germany. M. malmoense is very difficult to identify because same characteristics are instable and optimal cultivation conditions could not be obtained so far. Agglutination with homologous antisera was found to be specific for M. malmoense as well as lipid patterns which could be demonstrated by DC.

Sex-reversed XY females with campomelic dysplasia are H-Y negative.

Three families with infants affected with campomelic dysplasia, a genetically determined mesenchymal disease frequently associated with sex reversal were studied. Two XY females with ovarian gonadal differentiation and typical clinical features of campomelic dysplasia could be tested for H-Y antigen and were found to be H-Y negative.

[Diagnosis of loosening of carbon-made cup prostheses by radionuclide imaging (author's transl)].

Diagnosis of loosening of joint prostheses has become easier by using radionuclide imaging in addition to common radiography. Double-cup hip prostheses have been used more and more in joint surgery of younger patients in the last ten years. But early signs of loosening of the metal cup are difficult to demonstrate because of the high absorption rate of the used materials. Cup protheses of carbon have been implanted since several months in the Orthopedic Clinic Giessen. Tests in a model and in a patient have shown, that it might be possible to evaluate loosening of carbon-cup prostheses by radionuclide imaging.

[The influence of meteorological conditions on the side effects of contrast media during infusion urograms (author's transl)].

A relationship between certain weather conditions and the frequency of some diseases has been recognised for a long time. During a period of two and a quarter years, the course of infusion urograms in 6,862 patients was carefully monitored. At the same time, records were kept of the weather, including the general weather situation, changes in the weather, its dynamics, atmospheric pressure and "vorticity". 7.96% of patients showed side effects from the contrast medium. 32.6% of all patients admitted to having been affected by the weather, with an incidence of side effects of 9.7%. Older patients and over-weight patients tended to suffer fewer side effects. There was no correlation with the seasons. As regards the general weather situation, there was an increased incidence associated with changes coming from the east, fewer incidents during the in-flow of cold air and high pressure situations. There were marked differences during periods of high and low pressure ("vorticity"). High pressure situations and periods of increasing barometric pressure had complication rates of 5.3% and 4.7% respectively, low pressure, 11%. Statistical analysis showed no significant difference between the groups, but tendencies were unmistakable. A large number of individual factors are known to have an effect, such as abnormalities of the complement system, antigen-antibody reactions and others, but the importance of each factor remains uncertain. The present results indicate that the reaction of a patient at the moment of the investigation also plays a significant part.
