Active site structure and mechanism of human glyoxalase I-an ab initio theoretical study.

The structure of the active site of human glyoxalase I and the reaction mechanism of the enzyme-catalyzed conversion of the thiohemiacetal, formed from methylglyoxal and glutathione, to S-D-lactoylglutathione has been investigated by ab initio quantum chemical calculations. To realistically represent the environment of the reaction center, the effective fragment potential methodology has been employed, which allows systems of several hundred atoms to be described quantum mechanically. The methodology and the active site model have been validated by optimizing the structure of a known enzyme-inhibitor complex, which yielded structures in good agreement with the experiment. The same crystal structure has been used to obtain the quantum motif for the investigation of the glyoxalase I reaction. The results of our study confirm that the metal center of the active site zinc complex plays a direct catalytic role by binding the substrate and stabilizing the proposed enediolate reaction intermediate. In addition, our calculations yielded detailed information about the interactions of the substrate, the reaction intermediates, and the product with the active site of the enzyme and about the mechanism of the glyoxalase I reaction. The proton transfers of the reaction proceed via the two highly flexible residues Glu172 and Glu99. Information about the structural and energetic effect of the protein on the first-shell complex has been attained by comparison of the structures optimized in the local protein environment and in a vacuum. The environment of the zinc complex disturbs the Cs symmetry found for the complex in a vacuum, which suggests an explanation for the stereochemical behavior of glyoxalase I.

Electronic structure of [(CpCr)[(CO)3M]]mu-Cot (M = Cr, Fe): a theoretical study.

The electronic structure of two cyclooctatetraene-bridged dinuclear first-row transition metal complexes of the type [(CpM)[(CO)3M']]mu-Cot (M = Cr; M' = Fe (1), Cr (2)) was investigated by complete active space self-consistent field (CASSCF) calculations. In this context the differences in the binding capabilities of the complex fragments CpM and (CO)3M are discussed on the basis of extended Huckel molecular orbital (MO) calculations. The geometries used for the CASSCF calculations for complex 1 were obtained from the crystal structure. For 2 a model structure was established by geometry optimization using density functional methods. The CASSCF results agree well with the experimental findings and provide insight into the binding situation of the two compounds. Complex 1 can be regarded as being composed of a chromocene-like subunit CpCr(eta5-C5H5) and the fragment (CO)3Fe(eta3-C3H3). A direct metal-metal bond is found, involving one initially singly occupied orbital of each fragment, leading to a doublet ground state for 1 with the remaining unpaired electron localized at the chromium center. For 2 no such direct metal-metal bond can be recognized. A very weak direct metal-metal interaction is induced by electron donation from the Cot2- ligand into a formally unoccupied metal-metal binding orbital combination. In the quartet ground state all three unpaired electrons are localized at the chromium center of the formally doubly positive charged CpCr unit, on which complex fragment [(CO)3Cr(eta5-Cot)]2- acts like a cyclopentadienyl ligand. The coordination sphere of the chromium center of the CpCr unit resembles that of a metallocene metal center and its metal 3d occupation scheme corresponds to that of vanadocene.

Purification, characterisation and cDNA sequencing of pyruvate decarboxylase from Zygosaccharomyces bisporus.

Cells of the wild-type yeast strain Zygosaccharomyces bisporus CBS 702 form alpha-hydroxy ketones from aromatic amino acid precursors during fermentation. Pyruvate decarboxylase (PDC, E.C. 4.1.1.1), the key enzyme of this biotransformation catalysing the non-oxidative decarboxylation of pyruvate and other 2-oxo-acids, was purified and characterised. The active enzyme is homotetrameric (alpha4) with a molecular mass of about 244 kDa. Activation of PDC by its substrate pyruvate results in a sigmoidal dependence of the reaction rate from substrate concentration (apparent Km value 1.73 mM; Hill coefficient 2.10). A cDNA library was screened using a PCR-based procedure, and a 1856 bp cDNA of PDC was identified and sequenced. The cDNA encodes a polypeptide of 563 amino acid residues (monomeric unit). Sequence alignments demonstrate high homologies (> 80%) to PDC genes from Saccharomyces cerevisiae, Kluyveromyces lactis and Kluyveromyces marxianus.

Transcatheter endomyocardial laser revascularization: a feasibility test.

By means of a special catheter system, a total of 76 endomyocardial laser channels were percutaneously produced in a controllable manner at selected sites in 6 beating canine hearts. Acute patency of channels (length = 4-11 mm, diameter = 0.5-1.2 mm) was documented angioscopically and histologically. This minimally invasive method might be useful for revascularising certain patients with ischemic heart disease without resorting to open-chest surgery.

[Coronary risk factors in chronic hemodialysis patients].

BACKGROUND: In contrast to persons with normal renal function, coronary risk factors or indicators until yet could not clearly be defined in renal insufficiency. PATIENTS AND METHODS: 30 patients under chronic hemodialysis therapy were investigated; 15 patients with severe coronary artery disease and 15 patients with normal coronary angiogram were compared. Numerous factors of the manner of living (diet, smoking behaviour etc.) were registered and glucose and lipid metabolism, hemostatic and fibrinolytic system as well as blood pressure level were investigated. RESULTS: Besides higher HDL-cholesterol and tissue plasminogen activator (TPA) levels in patients without coronary heart disease, no significant difference could be found between both groups. The higher HDL levels were mainly due to the higher percentage of women in the coronary healthy group. There was no evidence of insulin resistance as a major pathogenic factor in the group with coronary heart disease. The blood pressure levels were not significantly different in both groups. CONCLUSION: Our quantitative examination of accepted or suspected coronary risk factors revealed no entity which turned out to be a reliable risk indicator for practical purposes.

[Hickman catheter for long-term parenteral therapy. A prospective interdisciplinary study].

160 patients with a median age of 38 years (range two months to 84 years) having silicone rubber central venous access catheters for long-term parenteral nutrition or chemotherapy were studied prospectively. Two different types of catheters were used, the Broviac-type "life-cath" and the Groshong -catheter. Intraoperative complications were not noticed. Parenteral therapy was performed in 124 patients (81%) without any complications. After a mean postoperative interval of 36 weeks there were 34 catheters removed because of end of therapy. 78 patients died with the catheter in place. 40 catheters had to be removed before end of therapy due to catheter-related complications. Occlusion of the lumen occurred in nine patients after an interval from three to 20 weeks. Explantation of catheters due to a suspected catheter-related sepsis was performed in 17 patients. Both types of catheter showed a high cumulative patency rate of 90% after a twelve months period. However, there were catheters that had to be removed before planned end of therapy. Thus, the cumulative rate of functioning catheters is lower (44% of all catheters).

[Drug-induced exudative erythema multiforme major with a chronic progressive course and bilateral blindness. Clinical and immunohistologic follow-up].

A 24-year-old male patient who had been received Fansidar developed exudative erythema multiforme major with massive involvement of the skin, mucous membranes, and both eyes. The chronic inflammation led to proliferation of the conjunctiva, with keratinization of the ocular surface and corneal vascularization. The final result was comparable to cicatricial pemphigoid, with blindness of both eyes. In histological sections, massive lympho-plasma-cellular infiltration with circumscribed perivasculitis was seen. The lymphocyte transformation test was still positive 18 months after onset of the disease. Class II antigens were evident in some of the conjunctival biopsies.

[Isolated infiltration of the vitreous body as a sign of blast crisis in chronic myeloid leukemia].

A 29-year-old man with chronic myeloid leukemia and two successfully treated blast crises exhibited ocular symptoms. Eighteen months after the diagnosis of leukemia he presented with unilateral hyperplastic iris stroma, anterior chamber inflammation and vitreous infiltration. Uveitis was diagnosed tentatively, and local therapy was begun with corticosteroids. Because the findings remained unchanged an invasive diagnostic evaluation followed. Although iris biopsy revealed only unspecific mononuclear cell infiltration (mainly T-lymphocytes), selectively aspirated vitreous material contained myeloid cells of different stages of maturation and blast cells. Peripheral blood, liquor and bone marrow showed no signs of acceleration or transition of the primary disease. Thus, an extramedullary blast crises with isolated vitreous infiltration was diagnosed.

[Indication for and interpretation of immunohistological studies of the conjunctiva and cornea].

Various conjunctival and corneal biopsies of eyes with chronic inflammation of the outer segment (e.g., following chemical burns, chronic proliferative conjunctival alterations) were examined using monoclonal antibodies. Class II antibodies were found in both epithelial and stromal conjunctival cells, indicating that there may be some etiological significance to an autoimmune process in altered tissue.

[Clinical types of immunologic transplant reactions following perforating keratoplasty].

The incidence of allograft rejection was determined for 740 penetrating keratoplasties performed between 1980 and 1987. All 740 cases were followed up for at least 1 year. The reaction forms of allograft rejection were grouped according to biomicroscopic appearance. The incidence and progression of symptoms are described. Rejection types are subdivided into patients with favorable versus poor prognosis. Of the patients 37.9% demonstrated an immune response (including discrete forms). Clear reaction patterns within the two groups of patients became apparent when reaction variations were carefully differentiated. Epithelial immune reaction was found in 5.2%/10.5% and subepithelial infiltration in 1.7%/4.8% of the patients with favorable/poor prognosis. The largest disparity in frequency occurred in progressive endothelial reaction; 3.8% in patients with favorable versus 36.7% in patients with poor prognosis. Focal endothelial reactions occurred in both groups with comparable frequencies (14.1%/13.3%). The large percentage of immunological reactions, including late manifestations (approximately 12% after 1 year) and some with irreversible progression, warrants continuing efforts to treat and prevent this complication.

Free-field sensitivity level of audiometric earphones to be used for speech audiometer calibration.

Speech audiometric test results strongly depend on the frequency response of the equipment used for speech transmission. For the comparison of data measured with different kinds of acoustic transducers, e.g. earphones and loudspeakers, a common reference basis for the specification of the frequency response of speech audiometers is therefore needed. The most obvious basis is the free sound field. In order to provide data for earphone specification in terms of the equivalent free-field response, which are to be included in an IEC standard on speech audiometers under preparation, comprehensive loudness comparison measurements with free progressive sound waves were performed on various audiometric earphones. When these data are used, calibration and testing of speech audiometers are as simple as in the case of puretone audiometers.

A study of Campylobacter jejuni and Campylobacter coli in a river system.

Campylobacter isolates were detected in 82.1% of river water samples moderately polluted with organic wastes. In the majority of the samples less than 10 Campylobacter cfu/100 ml were detectable. The presence of waterfowl and the faecal contamination from a poultry-farm resulted in considerable higher Campylobacter contents (greater than 240/100 ml). Correlation studies showed a high correlation coefficient (P less than 0.05) between the number of campylobacters and that of total coliforms. Campylobacter isolates were never found in river water samples of 100 ml of volume with total coliform contents less than 10/ml. Campylobacter contents and the spectrum of serotypes isolated indicate hygienic precautions for drinking water supply from oligotrophic surface waters if the water is distributed without chlorination.

[Antibiotic sensitivity of Campylobacter jejuni and Campylobacter coli strains with special reference to sensitivity to erythromycin and neomycin].

166 strains of C. jejuni and 46 strains of C. coli were typed against 14 antibiotics using the agar diffusion test. All strains were resistant against Penicillin G, Oxacillin and Rifampicin. Susceptibility were found against Chloramphenicol, Erythromycin, Neomycin, Oxytetracyclin, Streptomycin and Gentamycin. The minimal inhibition concentration was performed for Erythromycin and Neomycin. All strains tested were inhibited by less than or equal to 2 micrograms/ml Neomycin, one strain was resistant against 4 micrograms/ml Erythromycin.

Characterization of Campylobacter jejuni and Campylobacter coli isolated from waste water.

The raw sewage samples of the investigated municipal sewage plant (sewage flow of about 30,000 m3/d) contained about 10(3) campylobacters/100 ml. The sewage plant reduced 80.8% of organic wastes (COD), 85.0% of colony counts (20 degrees C), 92.5% of total coliforms and 88.0% of campylobacters. The sewage plant effluent contained an average of 1.3 X 10(2) campylobacters/100 ml. About 10(10) campylobacters are discharged with the effluent of the high-output sewage plant into the receiving water every day. No reduction of the indicator bacteria was found in the activated sludge tank, there was, however, a significant reduction (94.5%) of campylobacters. In total, 94 strains were differentiated by biotyping and serotyping. 59.6% were found to be Campylobacter jejuni and 40.4% were Campylobacter coli. 8 Serotypes were found among the C. jejuni strains and 12 among C. coli. The raw sewage of the investigated oxidation pond treatment plant contained an average of 51 campylobacters/100 ml. The sewage plant effluent did not show campylobacters in any case.
