[Double labeling of antibodies with fluorescein isothiocyanate (FITC) and horseradish peroxidase (HPOD)].

This paper presents a method for labeling antibodies with fluorescein isothiocyanate (FITC) and additional with horseradish peroxidase (HPOD). With this double labelled antisera the fluorescence-serologic antibody technique (FAT) as well as the enzyme-serologic antibody technique (EAT) was done bit by bit for the same object. How the presented pictures demonstrate the result of both techniques, FAT and EAT, are the same with the described method. So, it is possible after fluorescence-microscopical exploitation through a following histochemical proof of the HPOD to get permanent preparations with the same biological result.

[Use and results of the microscope photometry in the virology (author's transl)].

Labelled preparations are scanned fluorometrically and absorptionsphotometrically qualitative and quantitative evaluation. Thereby dynamic characteristics can be found which may serve as markers for different virus types. The titre, probability of detection and the required dilution for complete detection of antigens can be determined. The microscope photometry is applicable for screening of potential virostatica as well as topographic analyses of virus synthetisizing cells. The distribution of computer operated scanning photometry is ensued by digital plotter as a 3 dimensional or mosaic print out. Moreover, cadastres of the immunity can be obtained by the fluorometric technique.

[Comparison between single-radial-hemolysis-test and indirect fluorescence-antibody-test for the serological diagnosis of influenza].

Two serological methods for the diagnosis of influenza were tested with sera of guinea pigs and compared. Up to now, both methods have only rarely been used in the diagnosis of influenza. The methods proved to be well reproducible and suitable for routine investigations. Correlation between antibody titres measured by SRH and FAT give a correlation coefficient of v = 0.75. The possible IgG-specificity of the SRH, indicated in the literature, is reported.

[Computerized scanning analyses of FITC-labelled microscopic slides (author's transl)].

FITC-labelled virus antigen was analysed in infected cells by means of scanning-fluorometry. According to the predetermined classification the values were computerized and printed out by a mosaic printer or by a coordinate-recorder as a profile graph or a perspective image. The total impression of the 3 dimensional print-out of the quantificated topographic distribution of antigen accumulations within the cells can be optimated by variation of the angle of view.

Computer-operated scanning analysis of FITC-labelled measles virus in cell cultures.

FITC-labelled measles virus antigen in infected cells was analysed by computer-operated scanning fluorometry, printed out by a mosaic printer according to a predetermined classification, and by a coordinate-recorder as a perspective image. The three-dimensional print-out of the topographic distribution of antigen accumulations in the cells can be optimized by a variation of the angle of view.

Fluorometric determination of the quality of FITC conjugates.

A fluorometric method for the evaluation of FITC conjugates is described. By this method, the titer and the dilution required for complete antigen detection can be determined and an objective comparison of the various conjugate specimens is possible.
