[Comparative analysis of the population polymorphism of human silver-staining chromosomal nucleolus organizer regions].

The population study of nucleus organizing regions (NOR) activity in individuals of Georgian nationality was carried out using the Ag-method. The results obtained were compared with those for other populations: Russian, Viena-Ulm, Estonian etc. The mean number of active NORs for Georgians is 9.0, which significantly exceeds analogous indexes for the rest of populations, except Viena-Ulm one. The population differences in the distribution of NOR activity and the ability to be stained was found for individual NORs. The lowest activity was registered in chromosome 14, the highest being observed for chromosomes 21 and 13. It is assumed that some mechanism exists that controls the level of general NOR activity in the cell.

[Sequential staining of the polymorphic regions of human acrocentric chromosomes using different cytochemical methods].

The chromosome preparations of 12 normal individuals were consequently stained with four different techniques (RV, Q, C and Ag). Correlations between morphological and staining peculiarities of specific polymorphic segments of the short arms of the same acrocentrics were investigated. It was determined that except correlations for the length of satellite stalks and the size of Ag-deposits, the polymorphic signs revealed with Q-, C- and Ag-techniques appear to be independent and not correlating with each other. These data allow to think that in spite of morphological similarities between acrocentric arms, the individual chromosomes can differ in structure and properties of the nucleoprotein material available in the examined regions, which may cause a non-identical response to cytochemical treatment.

[Genetic determination of the activity of nucleolar-organizing regions of human chromosomes].

Activity of nucleolar organizer regions (NORs) was studied in cultured blood lymphocytes of 8 monozygotic and 8 dizygotic twin pairs using Ag-staining of acrocentric chromosomes. These chromosomes were identified by G-banding. NOR activity was estimated by the number of Ag-stained regions, their distribution among individuals acrocentrics and the size of silver deposits. No statistically significant differences were found as regards these criteria between the monozygotic twins. The dizygotic twins were distinguished, as a rule, in most pairs by one or another chromosome. The data obtained indicate that NOR, activity in each chromosome is of a heritable character, at least in blood lymphocytes. Chromosome 21 was found to have a greater NOR activity as compared with other acrocentrics.

[Quantitative assessment of human chromosomal polymorphism with regard to paracentromeric heterochromatin].

The measurement of C-segment length in chromosomes 1, 9 and 16 of 7 individuals was carried out. The regression analysis was employed to study a change of the C-segment sizes in the process of mitotic chromosome condensation. Typical values of C-segment length for chromosomes 1, 9 and 16 are about 1.4, 1.1 and 0.8mu respectively. Among 7 individuals there was no two which had identical size of C-segments for all three chromosomes studied. In six individuals heteromorphysm of C-segments was revealed. It was found that visually detected heteromorphysm may be expressed quantitatively as ratio length of C-segments in homologous chromosomes.

[Structure-function features of the long-arm iso-X-chromosome of 45, X/46X, i(Xq) patients].

Differences in the morphology of the human iso-X-chromosome differentially condensed by the treatment with 5-bromodeoxyuridine were revealed by means of quantitative morphometry. It is shown that these differences are not random and are correlated with the asynchrony of the reduplication of iso-arms. It is assumed that the differences in the structural organization and in the chronology of the reproduction of iso-arms are due to the different degree of their inactivation.

[Linear differentiation of the human Y-chromosome].

During the final 5.5th to 7th hr of the cycle in presence of BUdR, the human Y-chromosome was delayed in its mitotic condensation in the distal part of the long arm. This part was not homogeneous by the degree of the delay. The unevenly condensed Y-chromosome stained with quinacrine was brightly fluorescent in its stretched part. The fluorescence seemed to correlate with the degree of stretching. The distal part of the long arm was heavily labeled with H3-deoxycytidine thus demonstrating presence of late replicating DNA with GC-base pairs.

[2-colored fluorescence of differentially condensed chromosomes stained with acridine orange].

Metaphase chromosomes of the Chinese hamster differentially-condensed under the influence of a) 5-bromdeoxyuridine, b) colcemide, and c) cold, were stained with acridine-orange (AO) in concentrations of 1.5 X 10(-7) to 3 X 10(-5) g/ml at pH 4.1 to 8.5. It was found that stretched chromosomal segments fluoresced in the orange/red part of the spectrum, whereas normally condensed ones--were green. The colour distribution along the chromosome depended mainly on the AO concentration and the exposure in the UV-light, and was independent of pH and molarity of the buffer. Apparently this phenomenon cannot be attributed to the uneven denaturation of the chromosomal DNA, but rather depends on structural and/or chemical differences between euchromatin and heterochromatin.
