Measles IgA in the nasal washings of adult volunteers and children immunized intranasally with measles vaccine L-16.

Earlier it was determined that inspiration of aerosolized measles vaccines may be as effective as its injection in induction of measles antibodies formation. In the beginning of measles infection the measles virus penetrates through mucosa of nose, mouth or eyes of children. So it seems rational to use a nasal spray of measles vaccine to induce "mucosal immunity" in the nasopharinx. Local IgA measles antibodies formation was observed only after measles vaccine spray immunization of 6-7 year old children or adult volunteers. The same level of sera measles antibodies was observed in immunized people after intranasal or subcutaneous measles vaccination. Special investigation of measles vaccination side effects in adults revealed that the injected vaccine suppressed lymphocytes functional capacity much more than when intranasally introduced. Intranasal measles vaccine spray introduction may be a useful method of child revaccination in the process of measles eradication. This method is useful for investigation of "mucosal immunity" in children or adults.

[Value of exhaled air acetone level in assessing the impairement of secretory pancreatic function in acute destructive pancreatitis].

For the first time, a semiconductor gas sensor of original construction was employed to measure acetone level in the biologic liquids and gases in patients with acute destructive pancreatitis. A statistically significant difference in acetone level in the exhaled air of the healthy subjects, patients with edematous and destructive forms of acute pancreatitis and diabetes mellitus has been established.

[The immune status of children in foci of measles infection studied by an immunoenzyme method].

Children immunized with live measles vaccine in the foci of measles infection varying in intensity (1-9 cases per focus) have been subjected by two methods: the hemagglutination inhibition (HAI) test and the enzyme immunoassay (EIA). As shown in this study, in most cases (98% of all blood serum samples) the correlation between the results of the HAI test and EIA is not high (r = 0.5), which is linked with the detection of a wider spectrum of antibodies in EIA. The percentage of seronegative children detected by these two methods was practically the same (4.05 and 4.4, respectively). The analysis of the results obtained in this study indicates that EIA is a more informative and sensitive method, which confirms the effectiveness of its use for the determination of the level of collective immunity.

[Immunologic assessment of preparations of the RS virus at the stages of its purification and concentration].

The work demonstrates the immunogenic potency of some preparations of RS virus, differing in the degree of their purification and introduced by multiple intranasal administration. The immunogenic potency of these preparations was manifested by the synthesis of antibodies determined in the coagglutination test (the specificity of these antibodies was confirmed in the reaction of the neutralization of RS virus), as well as by sensitization determined in the leukocyte migration inhibition test. Immunization with concentrated virus was more effective than immunization with the preparation of virion fraction. The intranasal administration of live RS virus to mice induced the appearance of a morphologically determined disease which was not prevented by the development of immune response accompanying the disease. The conclusion was made that further improvement in the system of testing the preparations of RS virus was necessary.

[Mitogenic action of bacterial T-independent antigens under various conditions of lymphocyte culture].

The mitogenic response of lymphocytes in mouse spleen cell culture to the action of Salmonella typhi lipopolysaccharide (LPS) and Vi-antigen under different experimental conditions has been studied. The density of the culture has been shown to influence the activation of lymphocytes with Vi-antigen. Thus, macrophages stimulate mitogenesis at the concentration of lymphocytes equal to 1.0-1.2 X 10(6) cells/ml and suppress it when this concentration increases tenfold. The method used for the purification of cell suspension from adhering cells has been shown to influence the level of the mitogenic response of lymphocytes. The cultures, purified from macrophages by filtration through a column packed with cotton wool, have been found to respond to the mitogenic doses of LPS 7-8 times weaker than those purified by adhesion onto plastic. Background and mitogen-induced inclusion of 3H-thymidine into lymphocytic DNA varies in accordance with the presence of adhering cells. For this reason, in the evaluation of the influence of macrophages on mitogenesis it is expedient to consider not only the stimulation index, but also the absolute inclusion of thymidine into cells.

[Macrophage participation in the polyclonal and specific response to T-independent antigen in vitro].

Effect of adherent cells (macrophages on synthesis of total immunoglobulins (Ig) and specific antibody (Ab) in mice spleen cell culture stimulated by Vi-antigen Salmonella typhi has been studied. Ig and Ab in culture medium were determined by ELISA technique 96 hours after introducing the antigen into cell culture. The adsorption of analysed samples and reference antiserum on antigen coated microplates at pH 3.5 results in reducing nonspecific protein adsorption and promotes quantitative analysis Ab synthesized in vitro. The level of Ab produced in antigen stimulated spleen cell culture during 96 hours after antigen introduction is less than 2% of all synthesized Ig. Removal of most macrophages from spleen cell population results in a considerable decrease of specific and polyclonal immune responses. The basal Ig level in spleen cell culture (without antigen influence) does not decrease after removal of macrophages.

[Change in acid phosphatase and N-acetyl-beta-glucosaminidase activity in mouse lymphocytes induced by concanavalin A].

The treatment with concanavalin A (5 micrograms/ml) of mouse lymphocytes containing 70-72% of T cells entails an increase in the activity of acid phosphatase and a decrease in the activity of N-acetyl-beta-glucosaminidase. These changes were detectable 15 h after lymphocyte incubation with Con A. After 24 h of incubation acid phosphatase activity rose 2-fold whereas that of N-acetyl-beta-glucosaminidase dropped 45-50%. Possible mechanisms of these changes are discussed.

[Comparison of the antitumor, immunodepressive and toxic properties of carminomycin-albumin complexes differing in carminomycin content].

Bovine serum albumin (BSA) and carminomycin, an anthracycline antibiotic, were subjected to conjugation with glutaraldehyde and their complexes with various contents of the antibiotic were prepared. The molar ratios of carminomycin and BSA were 8:1, 4:1, and 2:1. The antitumor effect of the preparations was studied on the models of mouse transplantable lymphosarcoma LIO equal 1 and ascitic forms of mouse lymphadenosis NK/Ly in vivo and in vitro. Their immunodepressant effect was evaluated from the decrease in the hemagglutinin titers in the mice immunized with sheep red blood cells. It was shown that when the toxicity of the complexes was the same, their antitumor and immunodepressant activities were different. The therapeutic activity of carminomycin in the four- and eight-substituted complexes was much higher than that of carminomycin alone. It is suggested that the differences in the activity of the complexes were connected with differences in their pharmacokinetics. It was found that the chemotherapeutic properties of the complexes may have changed by variation of the number of the cytostatic residues in the albumin molecule. The findings indicate that the whole complex molecule interacts with the malignant cell and not carminomycin preliminarily detached from it.

[Immunodepressive action of anthracycline antibiotics studied on 2 different transplantation immunity models].

The effect of carminomycin and rubomycin in equitoxic doses on the reaction of rejection of the allogenic skin graft and the reaction of the transplant to the host in mice (RTAH) was studied. Carminomycin significantly hampered the rejection of the skin graft and under certain conditions prevented development of the RTAH which indicated suppression of the formation of the clone of the immune T-killers. Rubomycin almost did not hamper the rejection of the skin graft and had no effect on the RTAH which indicated the absence of the selective effect on the system of K-killers. Carminomycin eliminated the RTAH only when it was administered to the recipients of the allogenic lymphoid cells 24 hours after implantation of the cells to the mice treated with cyclophosphamide. When carminomycin was administered to the donors of lymphoid cells 24 hours before the beginning of the experiment, it did not suppress the ability of the cells from these donors to induce the RTAH. Therefore, its effect was directed to the precursors of T-killers activated in the host of the allogenic recipient.

[Development of antitumor immunity in animals treated with carminomycin].

Sensitivity of the mice inoculated with ascitic NK/Ly cells and cured with carminomycin to repeated inoculation with the same tumour was studied. It was found that curing with carminomycin was accompanied by development of resistance to the repeated inoculation. Such resistance might be transferred to intact singeneic mice with a suspension of the cells of the spleen or lymph nodes from unsusceptible donors. The transfer of the thymus cells from the above donors protected the recipients only partially. The spleen cells from intact donors, as well as their lymph node cells did not inhibit the tumour development. The thymus cells of the intact mice induced some suppression, thus probably favouring the active immune response in the recipient. Curing of the NK/Ly-inoculated mice with carminomycin was a more effective method for induction of immunity to the tumour than immunization of mice with NK/Ly cells inactivated by UV irradiation in vitro

[Antitumor activity of bleomycetin against ascitic tumors in mice].

When used intraperitoneally in doses of 18.0-0.006 mg/kg daily bleomycetin inhibited th growth of lymphadenosis NK/Li by 90-100 per cent. When used subcutaneously or intravenously in maximum tolerated doses the antibiotic inhibited the development of lymphadenosis NK/Li by 90 and 70 per cent respectively. The antitumor effect of bleomycetin on Ehrlich's carcinoma was somewhat less pronounced. Inhibition of Ehrlich's carcinoma growth by 50-90 per cent was observed with intraperitoneal and subcutaneous use of the antibiotic in doses of 0.2-1.0 and 5.0-13.0 mg/kg respectively. When used intraperitoneally bleomycetin markedly prolonged the life span of mice with lymphocytic leucosis P-388 and was superior by its efficiency to methotrexate used as a reference. On subcutaneous administration the efficiency of the antibiotic was almost the same as that of methotrexate and on intravenous administration it amounted to 70 per cent of the methotrexate efficiency. No effect of bleomycetin on its intraperitoneal use was observed with respect to lymphoid leucosis L-1210.

[Combined action of rubomycin and diuretics on the induction of antibody synthesis in mice].

The effect of 2 diuretics, diacarb and lasix on the intensity of immunodepressant action of low doses of rubomycin was studied. It was shown that diacarb increased the inhibitory effect of rubomycin on accumulation of the antibody-forming cells in the spleen. This effect was evident by the 3rd, 4th and 7th day after the antigen injection to the mice (the 4th, 5th and 8th day after the last injection of rubomycin). Combined use of rubomycin and diacarb also resulted in a low number of nuclear cells in the spleen as compared to control. Lasix did not increase the immunodepressant effect of rubomycin. Still, when used alone it had a capacity for changing the time course of the immune response development.

[Immunodepressive action of carminomycin and rubomycin derivatives].

The immunodepressive effects of carminomycin and its 3 semi-synthetic derivatives, as well as rubomycin and its derivative R-103 were compared. It was found that 14-hydroxycarminomycin was much superior to the other substances in the experiments with synthesis induction suppression of antibodies against sheep red cells in mice. Suppression of the rejection of the skin allogenic grafts in the mice by carminomycin was higher as compared to that by the other substances. Probably different populations of the immune competent cells have selective sensitivity to separate anthracyclines.

[Comparison of the action of carminomycin and rubomycin on the dynamics of the primary and secondary immune responses].

The effect of rubomycin and carminomycin on the dynamics of the primary and secondary immune response and formation of the immunologic memory to sheep red cells in mice was studied. Differences in the character of the antibiotics effect indicative of the higher selective action of carminomycin on multiplying cells, precursors of the antibody-forming plasmids, were found. Theoretically interesting discrepancies in the effect of the antibiotics on the content of the antibodies in the serum and the antibody-producing cells in the spleen were shown. It was demonstrated that carminomycin had no effect on formation of the immunologic memory inspite of a noticeable decrease in the total number of the spleen nuclear cells and the number of the antibody-forming cells at the moment of immunization under the effect of the antibiotic.
