[Selective culture media for Haemophilus bacteria].

A selectivity factor was specified to the previously developed nutrient medium meant for the cultivation of Haemophilus bacteria--CAE, which is based on the acid hydrolysate of casein, enzyme hydrolysate of animal blood (aminopeptide) and an extract of nutrient yeast. The above nutrient medium containing additionally growth factors V and X, glucose and bacitracin was shown to fit well for the primary sowing of Haemophilus bacteria and it can be used in diagnostic examinations.

[Design of a nutrient medium for bacteria of the genus Haemophilus--producers of restriction endonucleases].

A culture medium for the cultivation of hemophilic bacteria, containing acidic casein hydrolysate, aminopeptide and fodder yeast extract, has been proposed. The growth-stimulating properties of this medium have been studied on 5 strains producing restrictases differing in their specificity. In growing these producer strains in a model ANKUM-2 fermenter with the supply of carbohydrate substrates (glucose, sucrose, glycerin) the yield of biomass, considered to be high for hemophilic bacteria (10-14 g of humid substance from 1 liter of the medium), has been achieved. As shown on H. influenzae Rc B-2297 used as an example, an increase in the yield of microbial biomass leads to a decrease in restrictase specific activity.

[Polysaccharide-protein complexes isolated from different strains of Neisseria meningitidis serogroup B].

Fractionation of the biomass of 3 serogroup B N. meningitidis strains, obtained from solid serum-free and liquid synthetic media, by increasing concentrations of cetavlone revealed that the formation of natural polysaccharide-protein complexes with the ratio of their components approaching 1:1 was possible under the conditions ensuring the intensive synthesis of capsular polysaccharide. Two strains, 125 and 1642, grown on a solid amino peptide-containing medium regularly produced two polysaccharide-protein complexes with the protein/polysaccharide ratio approaching 1:1. One of these complexes passed easily into the supernatant fluid and could be precipitated with 0.1% cetavlone. The second complex was more firmly bound to the outer membrane of the cell and could be precipitated with 1% cetavlone. In most experiments an additional fraction with high protein content in relation to sialic acid was isolated from the biomass.

[Effect of the peptide and amino acid composition of nutrient bases on the parameters of microorganism growth].

The parameters of the growth of three microbial strains cultivated in growth media prepared with the use of different nutrient bases, both locally produced and imported, have been studied. The characteristics of the processes of bacterial cultivation have been found to correlate with the peptide composition of hydrolysates used in the experiment. Nutrient bases containing a wide spectrum of peptides with different molecular weight have ensured the most favorable conditions for the growth of microorganisms belonging to the taxonomic groups under study.

[Peptide and amino acid composition of different protein bases for culture media].

The enzymatic hydrolysates under study, obtained from different raw materials, have been shown to contain a great variety of peptides with different molecular weight. The highest content of fractions with a molecular weight of 2000 D has been observed in enzymatic meat and casein hydrolysates manufactures in the GDR. Low-molecular fractions (100-200 D) prevail in amino peptide. A great variety of peptides with different molecular weight is observed in Hottinger's meat hydrolysate and in blood clot hydrolysate obtained from the blood of laboratory animals. All peptide fractions have been shown to contain a wide spectrum of free amino acids. These data on the peptide and amino acid composition of different protein bases facilitate their rational use of microbiological culture media.

[Economic coefficients during batch and multicyclic cultivation of Clostridium perfringens type A].

The analysis of the accomplished processes of cultivation of C. perfringens, type A, has shown that at all cycles of the multicyclic process and during large-scale submerged cultivation in reactors the culture of this micro-organism remains in a physiologically active state. This is confirmed by high values of economic coefficients and by the consumption of keto acids.

[Pathology and physiology of microbes. III. Criteria for assessing functional states].

The study of different characteristics of the vital activity of microorganisms, made on type B Clostridium perfringens taken as an example, has disclosed the difficulty of evaluating the functional state of microorganisms only on the basis of the traditionally used characteristics of the population activity: the specific growth rate, the toxicity of the culture fluid and the antigenic properties of the culture. The economic and metabolic coefficients have been found to be the most suitable criteria for such evaluation; the state of the maximum physiological activity of the population is characterized by the maximum values of the economic coefficient and the minimum values of the metabolic coefficient. The minimum values of the economic coefficient and the maximum values of the metabolic coefficient are characteristic of the pathological state of the population.

[Pathology and physiology of microbes. IV. Economic and metabolic coefficients in assessing the functional states of Salmonella typhi].

The study of economic and metabolic coefficients under different conditions of batch and continuous cultivation with the use of S. typhi, a facultative anaerobe, as a model has demonstrated that the functional state of the microbial population is determined by the phase of the development of the culture, the composition of the culture medium, the conditions of aeration and stirring, the concentration of the energy-providing substrate and the dilution rate in continuous cultivation. The economic coefficient with respect to the energy-providing substrate has been found to be the main characteristic of the functional state of the population: the high values of this characteristic correspond to the physiological state of the population. The decrease of the economic coefficient below a certain critical value is indicative of the pathological state of the population.

[Comparative study of different substrates as the nutrient base of a culture medium for Bacteroides and Fusobacterium].

The results of the comparative study of different substrates prepared from foodstuffs and inedible raw materials and intended for use as the nutrient base of a medium for cultivating the organisms of the genera Bacteroides and Fusobacterium have demonstrated that commonly used nutrient bases prepared from meat, fish, casein and inedible animal blood (blood substitutes) are suitable for this purpose. The dried nutrient base prepared from the mixture of outdated blood substitutes (hydrolysin and casein hydrolysate) (see text for Russian letters) has been found to ensure the optimum growth of Bacteroides and Fusobacterium.
