[An oligonucleotide microarray for detection and discrimination of orthopoxviruses based on oligonucleotide sequences of two viral genes].

An oligonucleotide microarray for detection and identification of orthopoxviruses was developed. Genus specific and orthopoxvirus species-specific regions of the genes encoding chemokine binding and alpha/beta-interferon binding proteins were used as a target. The developed microarray allows the variola, monkeypox, cowpox, vaccinia, camel-pox and ectromelia (mousepox) viruses to be distinguished with a high degree of reliability.

[Effect of mexicor on oxidative stress in acute myocardial infarction].

Mexicor (5% solution and capsules) was used in 40 of 80 conventionally treated patients with acute myocardial infarction. The drug was given intravenously for 5 days, than intramuscularly (6-9 mg/kg) for 9 days and orally (0.1 mg t.i.d.) thereafter until discharge. Severity of oxidative stress was evaluated by K coefficient. Calculation of this coefficient required data on degree of oxidation of lipids in blood serum, serum levels of diene conjugates, malonic dialdehyde, alpha-tocopherol and ceruloplasmin. These parameters as well as activity of superoxide dismutase, glutathione peroxidase and catalase in erythrocytes were measured at admission, on days 2, 3, 7, 14 and at discharge. Mexicor treated compared with untreated (n=40) patients were characterized by diminished severity of oxidative stress at the account of lower levels of lipid peroxidation products and augmented compensatory potential of the endogenous antioxidant system.

[Effect of mexidol on oxidative stress in cerebral variant of hypertensive crisis].

Parameters of oxidative stress were studied in 44 patients with grade II-III essential hypertension and cerebral variant of hypertensive crisis. All patients received antihypertensive treatment. Mexidol was administered intravenously (300 mg o.d.) for 7 days starting with the second day of a hypertensive crisis. Control group consisted of 27 patients who were not treated with mexidol. The level of lipid peroxidation products and parameters of antioxidative system were used for assessment of the state of oxidative stress. Significant increases of the coefficient of oxidative stress were registered at all study points in patients who were not treated with mexidol. Treatment with antioxidant mexidol was associated with decreased coefficient of oxidative stress throughout the whole study period (up to 14-th day).

[Current trends in the treatment and outcome of acute myocardial infarction (MI)].

Current trends in the treatment and outcome of acute myocardial infarction were studied in a trial entering 2411 patients with macrofocal myocardial infarction (MI) treated in 1993-1998. There were many cases of severe and repeated MI. The age of the patients tends to increase. Lethal outcomes' probability is very high in multiple stenosis of the coronary arteries. This condition is associated with a high risk of cardiac insufficiency progression because of inadequate blood supply to necrosis-free myocardial zones. Thrombolytic therapy (TLT), ACE inhibitors reduce lethality but their use is limited by such factors as concurrent diseases, time from MI onset, rethrombosis risk. As a result, only 1/5 of the patients received pharmacological treatment. Balloon angioplasty is indicated in contraindications to TLT, in TLT failure or large residual stenosis. ACE inhibitors slow down the progression of cardiac insufficiency giving time for surgical revascularization. Updated treatment of MI improved its outcomes, but further progress in lowering MI lethality will be insignificant unless efficient methods providing adequate blood supply to the myocardium are introduced.

[Urapidil effects on oxidative stress in hypertensive crises].

Urapidil effects on oxidant stress were studied in 36 patients with hypertension stage I-III running with crises. Acute hypertensive crisis was managed by intravenous injections of urapidil (20-50 mg) for 5 min. After that for a week they received urapidil monotherapy followed by combined treatment for another week (quinapril--25 mg/day or ramipril--5-10 mg/day in combination with hypothiaside--25 mg/day; lokren--20 mg/day or atenolol--50-100 mg/day and diltiazem--180-360 mg/day) to stabilize arterial pressure finally. Oxidant stress was estimated by lipid peroxidation (LPO) and state of antioxidant system (AOS). Catabolism and anabolism were evaluated by blood levels of hydrocortisone and insulin. LPO, AOS, the blood hormones were measured before management of hypertensive crisis, immediately after it and 1, 3, 7 and 14 days after. Before the crisis treatment LPO was found high. Urapidil administration reduced levels of hydrocortisone, dienic conjugates, lipid peroxidation, oxidant stress rate (OSR). Later, LPO and OSR slowly increased. It is concluded on validity of the antioxidants use in hypertensive patients with crises.

[Sequence specific conjugates of minor groove ligands with inosine containing oligodeoxyribonucleotides].

Conjugates of the distamycin tetrapyrrole analogue containing four pyrrolecaboxamide fragments (MGB) with inosine-containing oligodeoxyribonucleotides were synthesized. The stability of duplexes formed by these conjugates depends on the composition of inosine-containing pairs and decreases in the order: IC > IA > > IT. For the duplexes (d(TTTATATA)p(MGB))2, (d(TTCICICI)p(MGB))2, and (d(TTAIAIAI)p(MGB))2, the melting temperatures are 68, 54, and 35-45 degrees C, respectively; (d(TTTITITI)p(MGB))2 forms no duplexes at temperatures above 4 degrees C. The binding was shown to be highly specific: in the duplex d(GGCATCTA)p(MGB).d(GGTAIATI)p(MGB), the substitution of only one A.T pair by A.A decreases the binding constant by almost three orders of magnitude.

[Selective stabilization of AT-rich DNA duplexes by oligodeoxyribonucleotide conjugates with distamycin analogues].

Oligodeoxyribonucleotide conjugates with distamycin analogues containing up to five pyrrolecarboxamide moieties were synthesized. The stability of duplexes formed by these conjugates was shown to depend directly upon the number of pyrrolecarboxamide moieties in the ligand molecule. For the duplexes formed by octaadenylate and octathymidilate conjugates with the distamycin pentapyrrole analogue, stability was demonstrated to be achieved by either one or two ligand molecules; however, duplexes containing two ligand molecules are more stable.

[The efficacy of emoxipin and neoton in unstable stenocardia].

55 patients with unstable angina pectoris (group 1) received conventional treatment and neoton infusions (30 g/day for 3 days). Emoxipin was injected intramuscularly to 20 patients of group 2 (3 mg/kg for 20 days) and intravenously to 38 patients of group 3 (10 mg/kg). Control group consisted of 100 patients. Stabilization occurred in 89.2, 80 and 86.8% of group 1, 2 and 3 patients, respectively, versus 78% in controls. Holter ECG monitoring provided evidence on decline of myocardial ischemia for group 1 patients. Diene conjugates in group 1 decreased by 37%, ceruloplasmin level was higher by 30% compared to controls. Respective indices for emoxipin reached 52 and 37%, respectively. It is concluded that emoxipin and neoton produced a beneficial effect on unstable angina pectoris through correction of lipid peroxidation.

[Neoton in the treatment of myocardial infarct and unstable stenocardia].

A total of 485 patients with complicated myocardial infarction (MI) in the presence of concurrent abnormality and 104 patients with unstable angina (UA) were followed up. Intravenous infusion of neoton (phosphocreatine) in a dose of 70 g at day 1 and 36 g at days 2 and 3 of the onset was made in 96 patients with MI, 28 patients with UA took neoton in a daily dose of 30 g during 3 days. Control patients had conventional therapy, In UA, poor outcomes (death, MI, no effects, referral of patients for bypass surgery) were seen in 4 (14%) patients on the drug and in 18 (28%) in the controls (p > 0.05). In MI, more rapid disappearance of events (heart failure and tachyarrhythmias) was observed in the major group, the mortality being 23.9%, the incidence of cardiac ruptures 6.4 versus 33.6 and 12.6% respectively, in the control. It is concluded that large-dose neoton exerts positive action on the cause and outcome of UA and complicated MI.

[Effective synthesis and cloning of the human interleukin-2 gene and its analog: expression of the interleukin-2 gene in E. coli cells].

Artificial DNA fragments encoding human interleukin-2 (133 a.a.) and its analogue (deletion of 14 C-terminal a.a.) were prepared by means of the DNA polymerase I mediated extension of synthetic polynucleotides having short overlapping sequences at their 3'-ends. The fragments were cloned in specially designed pFH-type plasmids and then excised by the FokI and other restriction endonucleases to yield the subfragments with the structurally predetermined 5'-unique cohesive ends. The complete synthetic gene was constructed by one or two-step ligation. The expressed IL-2 was tested by analysing the T-cell proliferation activity of E.coli crude lisates containing the pEXIL2 expression plasmid.

[Study of anti-ischemic action of carnitine chloride and its effects on the effectiveness of antianginal agents].

The experiments with models of myocardial infarction established that carnitine chloride reduced the size of an ischemic zone, prevented oxidative metabolism, lowered ATP and creatine phosphate levels, inhibited lactate and free fatty acid accumulation in the ischemic myocardium. Carnitine chloride in combination with nitrong and isoptin increased the anti-ischemic potency of the former.

[The effect of various types of stress-limiting therapy on the outcome of myocardial infarction].

A method for computed determination of death probability was used to examine the effects of various modalities of stress-limiting therapy on mortality rates within a month after myocardial infarction. The examination was made in 591 patients with large myocardial infarction. A significant decrease in mortality rates was seen in patients on finoptin and phosphocreatine (by 9.6 and 11.2, respectively) as compared to the expected drop. Other agents given within the first hours of myocardial infarction such as beta-blockers, opioid peptides, sodium oxybutyrate, piracetam, antioxidants were demonstrated to cause no reduction in mortality rates.

[Reconstruction of the synthetic gene for human interleukin-2].

Chemical-enzymatic synthesis and cloning of the DNA fragment coding for the human interleukin-2 signal sequence was accomplished. A hybrid plasmid pSIL-2 containing the gene of the human interleukin-2 with this signal sequence was constructed for effective expression of the gene in eukaryotic systems. A variant permitting the removal of the interleukin-2 stop-codons was obtained, which is suitable for the construction of chimeric genes containing the interleukin-2 gene sequence at the 5'-end.

[Effectiveness of synthetic opioid peptide dalargin in acute myocardial infarction].

The synthetic opioid peptide dalargin was examined for its potency in 86 patients with primary transmural myocardial infarction. The agent was shown to contribute to a decrease in initially elevated plasma cortisol levels and an increase in beta-endorphine and leucine-enkephaline concentrations. Myocardial contractility became normal by reducing the hyperfunction of its intact segments. This was evidenced by antistressor effects of the drug. However, no substantial effect was found on the clinical course of the disease, evolution of myocardial infarction, plasma CPK MB. The left ventricular contractility showed severe depression.
