[Heterogeneity of L-line cells in the early stages of clone development].

The clonal study of L cell culture has shown that the clone-forming cells are heterogeneous both in form and in the activities of enzymes (succinate dehydrogenase, lactate dehydrogenase, NAD- and NADP-diaphorase) which were determined by histochemical methods. The morphological heterogeneity is characteristic for clones with not less than 10 cells manifesting itself earlier and heterogeneity as to the activity of the studied enzymes--later, in clones with more than 15-20 cells.

[Differentiation and the neoplastic transformation of normal cells].

It is suggested that blastomogenic factors can be considered as the means isolating normal cells from each other. Carcinogenic substances caused cell functions to disturb, to inhibit or to intensify the synthesis of specific substances, to change the cell surface disrupted contacts of interactions between cells, pressing cells to become on the way of evolution, and to create heterogenic populations of cells.

[Rearrangements in cell populations in malignant growth].

It is emphasized that one of signs of malignant growth is heterogeneity of cell population which arises at the earliest pretumour stages and determines the character of tumour progression. At the same time by means of selective influences on population of malignant cells it is possible to change their quantitative correlation and to obtain rearrangement of the normal cell interrelations. Initiation of the tumour, its progression and regression are considered on the basis of the population-cell rearrangement, typical of each of them separately. The concept of the unity of the carcinogenesis and anticarcinogenesis processes is also formulated.

[Quantitative analysis of dyes for automated studies of cell preparations].

Spectral studies were made of the dyes in solutions and also of the cytology preparations stained by these dyes. 14 dyes of the blue-violet spectral region were examined. Absorbtion and transmission coefficients of the cell nucleus and cytoplasm were measured. The data obtained allowed to elaborate a quantitative estimation of dye selection. According to this method cresyl-violet was selected as the dye for Videocon LI-421.

[Cellular ultrastructure of a cloned L-line culture].

A study of the fine structure of cells obtained from the transformed cloned culture of strain L, that have developed from a single cell, demonstrated several different cell types. The clones contained both non-differentiated and poorly-differentiated cells. The presence of morphotypical signs in the poorly-differentiated cells allowed to reveal some cell clone types and the direction of their specialization process. The study confirms the idea on the presence of the stem cell elements in the transformed cell culture of strain L causing the heterogenous way of cytodifferentiation and determining the long-term stability of cell polymorphism in this cell strain.

[Effect of various serum fractions on L-cell differentiation].

It has been shown that the accumulation of lipids and maturation of L strain cells into mature lypocytes can be stimulated by high doses (30-60 mg/ml) of serum albumins, egg albumin lysozyme, and gelatin, or with low doses (200-600 mcg/ml) of serum globulins. The globulins are believed to induce differentiation of the L cell on depressing their proliferation. On the contrary, the low doses of albumins stimulate cell division without inducing differentiation.

[L line cell differentiation. Histochemical and electron microscopic studies].

In a medium supplemented with 60 per cent large cattle serum, part of the cell population of L-transformed culture was seen to be differentiated into mature lipocytes. The formation of structures similar to normal fat tissue took place in the same conditions, provided a reduced quantity of the seeded cells was used. The cell differentiation was accompanied with a decrease in acid phosphatase and non-specific esterase activities. The ultrastructural study has demonstrated a suitable preservation of cytoplasmic structures and a domination of the rough cytoplasmic reticulum compared to the control cell cultures where free ribosomes and polysomes predominated.

[Effect of chemical carcinogens on the ultrastructure of cultured differentiating myogenic cells].

The electron microscope studies have been carried out on primary monolayer tissue culture obtained from body tissues of rat and C3H mouse embryos. The cells of tissue culture were mainly myoblasts and fibroblast-like cells. The cultures were treated with two different carcinogenic substances--benz(a)-pyrene (BP) and methylnitrosonitroguanidine (MNNG). The changes were uniform and showed some alterations in formation of cell complexes, the inhibition of development and maturing of muscle elements, and distrophy of cytoplasmic organelles. The revealed distinction in morphological reaction of myogenic cells to the effect of BP and MNNG were wave-like myofibrillar structures in MNNG-treated cultures.

[Differentiation of a transformed culture of L cells].

The cells of line L originated from the connective and adipose tissue of mice C3H were cultivated in nutrient medium with a 60% bovine serum. Some cells differentiated into adipose cells with formation of structures similar to normal adipose tissue. The occurring differentiation was evaluated by the formation of characteristic signetring cells whose cytoplasm was filled with neutral fat with a positive reaction for lipids in staining with a mixture of sudan III and sudan IV. The conclusion was drawn that prolonged existence of cells outside the organism (in vitro cultivation) and intracellular changes (accompanying the transformation), including those of mutation character, did not deprive the cells of their ability to differentiate.
