Modulation of 1-[beta-D-arabinofuranosyl] cytosine-induced apoptosis in human myeloid leukemia cells by staurosporine and other pharmacological inhibitors of protein kinase C.

We have examined the effects of both nonspecific and highly selective pharmacological inhibitors of protein kinase C (PKC) on the capacity of a 6-h exposure to 1-[beta-D-arabinofuranosyl]cytosine (ara-C; 10 microM) to induce apoptotic DNA fragmentation and cell death in the human myeloid leukemia cell lines HL-60 and U937. Staurosporine, a highly potent, nonspecific inhibitor of PKC (20-50 nM), uniquely potentiated ara-C-related degradation of DNA to oligonucleosomal fragments in both cell lines (i.e., 2- to 3-fold), but was ineffective when given alone at these concentrations. In contrast, co-administration of the nonspecific PKC inhibitor H7 and two highly selective PKC inhibitors, calphostin C and chelerythrine, also increased the extent of DNA fragmentation observed in ara-C-treated cells, but these effects were evident only at inhibitor concentrations that were by themselves sufficient to induce DNA damage. Agarose gel electrophoresis demonstrated that cells co-exposed to staurosporine and ara-C exhibited considerably more pronounced internucleosomal DNA cleavage than did cells exposed to ara-C alone; moreover, this effect was suppressed by Zn2+ (1 mM) and the permeant Ca2+ chelator BAPTA-AM (50 microM). Potentiation of ara-C-related DNA fragmentation by subeffective concentrations of staurosporine was accompanied by a pronounced increase in the morphological features characteristic of apoptosis. A synergistic interaction between staurosporine and ara-C with respect to inhibition of clonogenicity in both HL-60 and U937 cells was demonstrated by median dose-effect analysis. The actions of staurosporine did not result from enhanced ara-C metabolism, as preincubation of cells with concentrations of this agent that potentiated ara-C actions (e.g., 20-50 nM) did not increase intracellular levels of the lethal metabolite ara-CTP. Lastly, preexposure of HL-60 and U937 cells to staurosporine did not block ara-C-mediated upregulation of c-jun, an oncogene whose increased expression has been temporally associated with ara-C-induced apoptosis. Together, these findings indicate that staurosporine exhibits a unique pattern of potentiation of ara-C-related apoptosis in human myeloid leukemias, and provide a rationale for exploring the antileukemic potential of this combination regimen.

Birth control practices and conservatism.

This study examined the relationship of conservatism to reports of contraceptive usage, reasons for nonusage of a particular birth control method, and knowledge of birth control information. The never married high conservative subjects who were sexually experienced revealed that they had (a) a lower frequency of usage as well as a lower variety of contraceptive methods; (b) endorsed the beliefs that it was morally wrong and that it was unnatural to use a given birth control method significantly more often as reasons for not using contraceptive techniques; and (c) less knowledge of birth control methods than did low conservative subjects who were sexually experienced. Results were interpreted to support the notion that the fear of loss of control of one's impulses and hence the basis for conservative attitudes are determinants of contraceptive usage.

Religiousness and devaluation of a rape victim.

In a 4X3 design (religious orientation vs. marital status of victim), 167 Ss were given a case account of a rape. Intrinsically religious Ss devalued the victim less than did Ss who were extrinsically oriented, indiscriminately proreligious and indiscriminately nonreligious. No significant effect was noted for the marital status of the victim, nor was there a significant interaction between these two variables.

Conservatism, openness to experience and sample bias.

Two experiments were conducted to examine the relationship between conservatism and openness to experiences. In Study I, the responses of 64 males and 60 females to the Conservatism Scale and the Coan (1972) Experience Inventory were correlated. For the total sample, conservatism was significantly related to the total score of the Experience Inventory plus six of seven of the component scales. Study II examined the relationship between conservatism and the expressed willingness to volunteer for certain psychological experiments that were assumed to require more openness. Subjects were 91 males and 114 females from introductory psychology classes. High conservative subjects were less willing to volunteer for the experiments that required more openness as compared to low conservative subjects. These results were discussed in the context of volunteer bias in psychological research.

Conservatism as a determinant of sexual experiences.

An examination of the relationship of conservatism to reports of sexual experiences and reasons for nonparticipation was conducted using 64 male and 50 female introductory psychology students. Conservatism was found to be inversely related to the frequency and variety of sexual behaviors. High conservative subjects gave the reason that it was morally wrong more often than any other reason for not engaging in coital activities and more intimate forms of petting. Results were in accord with previous research and were interpreted as supporting the notion that the fear of loss of control of one's impulses and hence the basis for conservative attitudes are determinants of sexual experiences.

A test of the Zucherman heterosexual scales.

Female and male undergraduate students enrolled in the introductory psychology course were administered the Zuckerman heterosexual scale as part of a larger battery of tests. The ordering of items for males was very similar to Zuckerman's with two minor exceptions. Female subjects, on the other hand, reported more sexual experiences of all kinds than did Zuckerman's sample, and two major differences in ordering Were noted. The coefficients of reproducibility for males and females were .928 and .954 respectively which were almost identical to those of Zuckerman. The rank-order correlation between the ranking of the items in the male and female scales was .958.
