[Microscopic organization of epithelial endocrine cells of the gastrointestinal tract mucosa in lower vertebrates. I. Epithelial endocrine cells of the stomach of the common frog].

The quantity, histotopography and submicroscopic organization of endocrine cells in various mucosal epithelium sections of frog were studied using light and electron microscopy. It is shown that the majority of endocrine cells per square millimetre is located in the proximal part of the stomach. Eight types of endocrine cells depending on the submicroscopic structure of secretory granules are defined.

[Microscopic organization of epithelial endocrine cells of the gastrointestinal tract mucosa in lower vertebrates. II. Epithelial endocrine cells of the duodenum of the common frog].

The endocrine epithelium cells of the frog duodenum mucosa were studied using light and electron microscopy. In the intestinal epithelium endocrine cells are distributed among enterocytes all over the surface of mucosa. The greatest quantity of them is observed in the intestinal part in the proximity of the stomach. Six types of endocrine cells are identified on the basis of their granular structure and size. Some differentiation in submicroscopic organization of endocrine cells depending on their functional condition is defined.

[Regeneration of the epithelium of the pancreas during organ culture].

By means of electron microscopy method, regeneration of the pancreas has been studied after its longitudinal resection in 3-day-old rats under conditions of organ cultivation, when antihypoxant is administered into the nutritive medium. It affects bioenergy of the cell, decreases hypoxia phenomena, that are observed at cultivation, and thus contributes to a prolonged preservation of vitality of the explants.

["Mixed" endocrine gland cells of the duodenal epithelium in various vertebrate animals and man].

Epithelium of the duodenal mucous membrane has been studied in frog, rat and man by means of electron microscopy. Together with typical endocrinocytes, incretory cells of "mixed" type are revealed in the epithelium. They are classified into two types: 1. exo-endocrinic containing in their cytoplasm simultaneously: a) mucous and EC-granules, b) mucous and S-granules, c) granules specific for Paneth's cells and S-granules, d) granules specific for Paneth's cells and L-granules; 2. endocrinocytes with incretory granules of various types: with D and I, with D and EC, with D and L granules. Various ultramicroscopic organization of the "mixed" exo-endocrinic cells is demonstrated. It can be considered as various stages of their differentiation. In cytoplasm of the "mixed" cells of the second type (endocrinic) a constant presence of D-granules is revealed, that is, evidently, characteristic, within the limits of the gastro-entero-pancreatic system, for the cells with two types of the endocrinic granules.

[Ultrastructural changes in the receptor portion of the vestibular apparatus in response to noise].

The experiments have been performed on 24 guinea pigs (48 labyrinthes), 6 of them--control and 18 have been subjected to noise (one octava) with the average geometrical in diaposone of 2,000 Hz, at intensity level 100 dB. Six animals are subjected to a single effect for 6 h and 12--to repeated effect during 6 days running, 4-6 h daily. Isolation of both the vestibular and the cochlear parts of the membranous labyrinth is performed simultaneously. This gives a possibility to study all the receptors of the internal ear as a whole. Certain ultrastructural changes in all the vestibular receptors both at a single and repeated effects are revealed. Dilatation of the granular endoplasmic reticulum is observed, situating mainly in the basal part of the cell; swelling of mitochondria is accompanied with a sharp clearance of their matrix. In some mitochondria there is a local destruction of their external and internal membranes. Moreover, in cytoplasm of the receptory cells sharply osmiophilic fibrillar structures are revealed, they resemble crists and are arranged in bundles. In some cells they are not numerous and localize mainly in mitochondria, in others--their number is greater, and they localize not only in mitochondria, but in the surrounding cytoplasm, too. Similar structures are observed in some preganglionic myelin fibers. These phenomena can be considered as development of calcification processes. The changes described, evidently, form the basis of the vestibular disorders under the noise effect.

[Accumulation of cholesterol in Acholeplasma laidlawii membranes in the steady-state phase of culture growth].

In early logarithmic phase of growth of the A. laidlawii cells the lipid composition of plasma membrane is changed: the total lipid, glycolipid and phospholipid contents are decreased, while that of cholesterol changes only insignificantly. In late logarithmic and steady state phases the cholesterol level in the membrane is increased in parallel with the decrease of the phospholipid content. Throughout the growth period a quantitative redistribution of membrane phospholipids in fatty acids and an increase of the molar content of saturated fatty acids are observed. Accumulation of cholesterol in the steady state phase is accompanied by an increase in the membrane viscosity which results in inhibition of membrane processes in the cell.

[Proteins of bacterial membranes. H+-adenosinetriphosphatase from Acholeplasma laidlawii cells].

The cytoplasmic membrane of micoplasmic cells, in particular of A. laidlawii cells, contains a proton-carrier Mg2+ -activated ATPase. A whole H+ -ATPase complex (F0-F1) was isolated from these cells and characterized. The isolation procedure included solubilization of the enzyme with Triton X-100 followed by ion-exchange chromatography on DEAE-cellulose and gel filtration on Sepharose 6B. The enzyme was inhibited by dicyclohexylcarbodiimide (10(-4) M). The Km value for ATP hydrolysis and Ki for ADP hydrolysis were determined. The order of the constants did not differ from those measured earlier for factor F1 of the complex. The purified enzyme, similar to its hydrophylic moiety is sensitive to the action of bivalent cations. The subunit composition of the whole complex and of its water-soluble part was investigated. The complex was found to contain 11 polypeptides, five of which belong to factor F1. The molecular weights of these polypeptides were determined.

[Proteins of bacterial membranes. Purification of soluble ATPase from Acholeplasma laidlawii].

A purified preparation of ATPase (factor F1) from the Acholeplasma laidlawii was obtained. The purification procedure included extraction of the enzyme complex from the isolated membranes by ultrasonication, chromatography on DEAE-cellulose and gel filtration on Sepharose 6B. The specific activity of the ATPase was increased 30-fold as compared to the original activity. The Km value for ATP hydrolysis was 7,4 . 10(-4) M. ADP competitively inhibited the enzyme (Ki = 2,0 . 10(-4) M). Ouabain (2,5 . 10(-4) M) and dicyclohexylcarbodiimide (1,0 . 10(-4) M) did not inhibit the ATPase activity. The enzyme was activated by Mg2+, but was inhibited by a combination of Na+ and K+. The enzyme is cold-labile, but can be stabilized by storage in buffer solutions, containing methanol, glycerol or lecithin.

[Liver function disorders and hepatic insufficiency in diseases of the abdominal cavity organs].

The tissue respiration and oxidative phosphorylation in mitochondrial preparations of the liver were studied by means of the polarography in 14 cases of peptic ulcer localised in the stomach and the duodenum and 46 cases of cancer of the stomach. The deterioration of the indices of oxidative phosphorylation in the liver (decrease of the respiratory control, desintegration of the respiration and phosphorylation processes) has been found to occur with the development of the tumor process.

[Differentiation of mesothelial cells during their reparative regeneration].

The investigation on regenerative processes of mesothelium of the parietal peritoneum was performed in 120 white mice under the effect of certain irritants producing lesions various in depth and intensity. Nuclear-cytoplasmic relations and ultramicroscopic cellular rearrangement were studied during the process of differentiation of the mesothelial regenerate. Two periods of the regenerative process are stated and it is demonstrated that rearrangement of the mesothelial cells and the mode of their division depend on intensity of the lesions. When the peritoneal lesion is severe, at the first stages of regeneration (the 1st period) rearrangement of cells towards their hypertrophy and increased functional activity is predominant in the mesothelium. Further (the 2d period), the number of mitotically dividing cells is increasing in the mesothelial regenerate and in rearrangement of the mesothelial cells the processes connected with a partial loss of their signs of specialization predominate. The transition from one period into another is gradual and duration of each depends on intensity of the lesion.

[Ultrastructure and secretory cycle of the islands of Langerhans cells in pancreatic cancer].

By electron microscopy, the islets of Langerhans of 5 patients with cancer of the pancreas were studied. The most obvious ultrastructural changes were found in the insulin-producing B-cells. Their cytoplasm contained a reduced number of secretory granules. Many B-cells were seen at different stages of the secretory cycle, part of them being destructed. On studying secretory process, the formation and maturation of B-granules was demonstrated in the cistern of ergastoplasm without the Golgi complex being involved. The data obtained suggest the functional tensity of the insular tissue at the neoplastic process in the exocrine part of the pancreas.

[Autoradiographic study of mesothelial proliferation in white mice under experimental conditions].

The proliferative activity (mitotic cycle, proliferative pool) of the mesothelium of the parietal peritoneum of white mice acted upon by distilled water was studied autoradiographically with H3-thymidine. The mesothelium regenerated due to high mitotic activity and increased amount of polynuclear cells. The latter are formed as a result of amitotic and mitotic division of the nucleus without plasmotomy. The distribution of cells at the stage of DNA synthesis and mitosis in all the parietal peritoneum speaks of the absence of special proliferation zones.

[Role of the mesothelium of the parietal peritoneum in the process of absorption of true solutions and india ink suspension from the abdominal cavity].

The method of electron microscopy was applied to the study of absorption from the abdominal cavity of hypotoanic, physiological, hypertonic solutions and of the Indi-ink suspension on physiological saline. Quantitative assessment of pynocytosis was conducted on electron microphotographs. Passage through the mesothelium of true solutions was realized by way of diffusion through the cytoplasm of its cells and by the intercellular spaces; as to the India ink suspension, it passed only by way of the open intercellular spaces. Pynocytosis in the mesothelial cells coursed from their basal portion to the apical one and was directed to the balancing of the concentration of the protein and salt composition in the serous fluid.
