Influence of phytogenic substances with thyreostatic effects in combination with iodine on the thyroid hormones and somatomedin level in pigs.

Growing pigs were given 8% rape seed meal (RSM), which was offered without or with different iodine-supplements and further trace elements. RSM without I-supplementation reduced food intake and weight gains by more than 50%, thyroid weight increased by the factor 6, T4 declined below the detectable limit, T3 by two thirds and somatomedin activity by 50% compared to the control group (soy bean meal). In spite of supplementation with iodine and further trace elements the T3-level reached the level of the control group in no variant due to nutrient deficiency (reduced food intake) and the conversion-impeding effect of goitrogenic substances. The same is true for Sm-activity and weight gains, whereas the T4-level increased even above the level of control animals due to I-supplementation. There are significant correlations (correlation coefficients 0.77 and 0.64 resp.; p less than 0.001) between food intake and T3-concentration resp. and Sm-activity. This study demonstrate that the reduced growth in case of the intake of phytogenic substances with thyreostatic effects is directed by the diminished food consumption and the peripheral hypothyroid situation via decreased Sm-synthesis. The high content of thyreostatic substances in tissues of rape seed fed animals is a potential danger for the human consumer.

Long term toxicological studies on the progestin STS 557.

The toxicity of 17 alpha-cyanomethyl-17 beta-hydroxy-estra-4, 9-dien-3-one (STS 557) was studied by its oral administration of 0.1, 1.0 or 10.0 mg/kg/day to Wistar rats for six months, and of 0.01, 0.1 or 1.0 mg/kg/day to beagle dogs for six months, respectively. Levonorgestrel at a dose of 1.0 mg/kg/day was used as the standard in the dog study. With respect to the progestational activity of the compound the main target organs were the hypophysis, the reproductive organs and the adrenals. Mammary hyperplasia was observed in dogs treated with STS 557 or levonorgestrel at the dose of 1.0 mg/kg/day, but in no case mammary nodules could be detected. At the dose of 1.0 mg/kg/day STS 557 and levonorgestrel were found to increase the plasma insulin response to i.v. glucose in bitches, but neither the mean blood glucose levels nor the glucose utilization were affected. Moreover, during administration of both steroids to dogs temporary changes in serum concentrations of triglycerides and total cholesterol were noted. The results obtained in rats and dogs from functional and morphological investigations did not reveal any toxic side effects of STS 557 on the liver, the kidneys, the bone marrow or on blood coagulation. The effects on the reproductive organs observed following STS 557 especially in dogs are related to both the hormonal effects of the compound and the specific response of the dog to potent progestagens.

["Hypothyroid" goitre in puberty treated with a combined T3/T4 preparation (author's transl)].

After administration of combined T3/T4 preparations (ratio of 4 : 1) to juveniles with goitre, size II-III (WHO), and hypothyroid T4 or PBI levels, the T3 value reached hyperthyroid levels 7-8 hours afterwards. The increased T3 level with normal T4 blood levels can lead to iatrogenic T3 hyperthyroidism. Serial tests demonstrated that at increasing doses a raised T3/T4 ratio, which is observed with low T4 values, can return to normal but with increasing doses this ratio, caused by T3 rise, can again become elevated. For this reason one should reconsider treatment with T3/T4 preparations and, if necessary, replace it by monotherapy with L-thyroxine.

[TRH refractory TSH values, triiodothyronine levels and T3/T4 quotients in hypothyroid children treated with T3/T4 preparations (author's transl)].

TRH-tests, triiodothyronine (T3), TBI, and thyroxine (T4) determinations were carried out in 23 children with primary hypothyroidism treated with T3/T4-preparations. T3/T4 quotients were calculated. 7 out of 10 patients with TRH refractory TSH values and normal T4 levels had T3 values above normal range. In 6 of them pathologically elevated T3/T4 quotients could be observed. A TSH refractory TSH suppression was only found in patients with superior normal T4 values. After a reduction of the replacement doses the T3 values were normal. The findings show--that not only the T4 values should be controlled under treatment with thyroid preparations but also the T3 and TSH serum levels. The replacement therapy of hypothyroidism with T3/T4 preparations in proportion 1:4 should be revised.
