The effect of 30 day administration of undiluted propylene glycol (1,2-propanediol, PG) on plasma and liver lipids was studied in adult male Sprague-Dawley rats. When compared with the control group no significant difference was observed in plasma concentration of total phospholipids, cholesterol, triglycerides and free fatty acids, and in liver concentration of phospholipids, triglycerides and gangliosides. Liver total cholesterol was moderately, but significantly, increased.

The effect of long-term (7-11 weeks) administration of perhexiline maleate (Pexid) on liver, lung, plasma and bile lipids was studied in adult male Sprague-Dawley rats. Maximum non-toxic doses of the drug (80 mg/kg/day) produced a fall of total cholesterol concentration in liver, lung and plasma, a fall of total lung phospholipid concentration, and an elevation of bile flow and biliary bile acid excretion. Minimum lethal doses (160 mg/kg/day) produced an increased liver concentration of total phospholipids and gangliosides and decreased lung concentration of gangliosides. Plasma transaminase activity was not significantly affected in either of the treated groups.
1. Biliary excretion of sulphobromophthalein (BSP) was studied in male rats after oral administration of tilorone HCl, an anti-viral and anti-tumour agent. 2. The stimulation of bile flow by BSP was decreased 1 h after tilorone administration; this decrease was no longer apparent 24 h later but a fall of basal bile flow was noted at that time. No changes in bile flow were observed 7 days after this single dose. 3. Repeated dosing with tilorone produced a dose-dependent increase in hepatic concentration of unchanged and metabolized tilorone. This was associated with a dose-dependent decrease of bile formation and BSP excretion. Bile acid excretion was depressed as well. 4. BSP metabolism was not significantly affected by tilorone administration.
1. The hepatic disposition of tilorone HCl, an antiviral and antitumour agent, was studied in male Wistar rats after intraduodenal administration. 2. A concentrative transfer into the liver occurred, the liver/portal blood concentration ratio being 120 or higher. The relationship between dose and hepatic concentration was linear. 3. Biliary excretion of tilorone was low and dose-related; whole blood concentration was also dose-related. 4. Neither hepatic concentration nor biliary excretion exhibited saturation over the dosage studied (6--174 mg/kg). The hepatic 'depot' of tilorone lacked the characteristics of a true 'first-pass' effect.
Lower concentrations of total serum zinc (540 +/- 111 mug/1, mean +/- SEM), and of albumin-bound serum zinc (295 +/- 113 mug/1) and a higher concentration of alpha2-macroglobulin-bound zinc (245 +/- 69 mug/1) were found in 25 patients with decompensated hepatic cirrhosis, compared to 28 healthy subjects (835 +/- 91; 679 +/- 83; 156 +/- 27 mug/1 respectively). Levels of total and albumin-bound zinc were significantly and positively correlated with serum albumin levels. Higher levels of alpha2-macroglobulin-bound zinc were associated with higher levels of alpha2-macroglobulin in these patients (2.8 +/- 0.8 g/1) compared to normals (2.3 +/- 0.6). Hence, not only do decompensated cirrhotics exhibit a lower serum zinc level but a greater proportion of this zinc is associated with the tightly bound, and presumably metabolically more inert, serum fraction. This situation exaggerates the zinc deficiency state of the severe cirrhotic.
