[The experience of diagnosis, treatment and prophylaxis of gastric and duodenal ulcer in outpatient conditions].

Gastric and duodenal ulcer (peptic ulcer) takes the leading place among the pathology of digestive organs. According to the data of specialists from Consulting-and-Diagnostic Center N 52 among the servicemen with chronic diseases the digestive pathology constitutes 32%. Among these diseases the peptic ulcer occurs in 81% cases. Screening esophagogastroduodenoscopy conducted in 1100 servicemen without any complaints aged 22-50 has revealed duodenal ulcer in 5.4%, gastric ulcer--in 0.7% and gastroduodenitis--in 11.4%. With introduction of modern methods of diagnosis, treatment, rehabilitation and prophylaxis of peptic ulcer 98% patients with disease uncomplicated course began and completed treatment in outpatient conditions.

Gamma-radiation-induced cell death in the fetal rat brain possesses molecular characteristics of apoptosis and is associated with specific messenger RNA elevations.

Low-dose ionizing irradiation of 16-18-day pregnant rats rapidly kills stem cells in the fetal forebrain. We have examined gamma-irradiated 17-day fetal rat brain tissue for molecular characteristics of apoptosis and changes in levels of mRNAs relevant to apoptosis. In many forebrain cells radiation elicits within 5 h nuclear condensation and fragmentation consistent with apoptosis. An electrophoretic DNA ladder indicative of internucleosomal chromatin cleavage was prominent within 3 h after irradiation. Pretreatment of pregnant rats with cycloheximide, or pretreatment of dissociated fetal brain cells in culture with actinomycin D, abolished the radiation-induced internucleosomal DNA fragmentation, demonstrating requirements for protein and RNA synthesis. Irradiation dramatically increased the level of the p53 transcription factor and the abundances of mRNAs coding for the cell-cycle inhibitor p21/Waf-1/Cip-1 and the AP-1-associated transcription factors Fos and JunB. Irradiation moderately increased the level of mRNA for the positive apoptosis regulator Bax. In contrast, irradiation reduced by 50-70% the abundances of most other mRNAs tested, including those for housekeeping proteins, p53, Jun, Myc, interleukin-1-beta-converting enzyme, and the negative apoptosis regulators Bcl-2 and Bcl-xL. These results indicate that radiation-elicited apoptosis of fetal brain cells is associated with activation of the p53 system, probable increases in AP-1 Fos/JunB heterodimers, and an increased ratio of Bax to Bcl-2 + Bcl-xL.

[Biosynthesis and processing of heterogeneous nuclear RNA in rat thymocytes].

Using [14C]adenine as a labeled precursor, the biosynthesis and processing of heterogeneous nuclear RNA (hnRNA) in chromatin and nucleoplasm were studied. It was shown that intraperitoneally injected actinomycin D (50-500 micrograms/100 g of body weight) did not inhibit the biosynthesis of hnRNA and rRNA in rat thymocytes. Besides biosynthesis, the polyadenylation of the bulk of primary transcripts was also localized in the chromatin. rRNA precursors constituted only a small part of the newly synthesized hnRNA molecules. During processing, highly polymeric (28S-55S) poly(A+)-hnRNA fractions passed from chromatin to the nucleoplasm, as a result of which a large number of high molecular weight polyadenylated hnRNA molecular were pooled in the nucleoplasm. Thymocyte hnRNA was tightly bound to nuclear structures and its isolation tributed stringent conditions. The bulk of poly(A+)-hnRNA molecules were degraded without formation of stable intermediates. The pattern of poly(A-)-hnRNA maturation in chromatin and nucleoplasm was identical, i.e., in both compartments the decrease in the size and accumulation of processed 16S-20S RNA molecules was observed. About 6% of newly synthesized poly(A+)-hnRNA was converted into polysomal mRNA. Cytoplasmic poly(A+)-RNA consists of two fractions, i.e., metabolically stable high molecular weight (greater than or equal to 40S) RNA molecules and a less stable 10S-20S RNA.

[Separation of rat thymocyte populations in a ficoll-paque gradient. IV. The genetic complexity of the cellular RNA of thymocyte subfractions].

The sequence complexity of the total cellular RNA was measured by RNA-driven hybridization to 125I-single copy DNA (sc-DNA). In the high density subfraction of thymocytes 4.5% of sc-DNA was saturated with cellular RNA. The sc-DNA probe was recovered from the DNA--RNA hybrids and rehybridized with homologous RNA and heterologous RNA from the cells with low density and middle density. Heterologous hybridization reactions with recycling sc-DNA probes revealed that 5.5% RNA sequences were specific of the cells from the high density subfraction and absent in both low and middle density subfractions. This is equivalent to expression of approximately 1100 additional structural genes in the high density subfraction of thymocytes.

[Maturing of ribosomal RNAs in rat thymus cells].

The peculiarities of maturing of ribosomal RNAs in rat thymus cells were studied. It was found that the rRNA precursors (45s, 41s and 32s) are weakly extractable from the thymocytes at 45 degrees and are predominantly accumulated in the RNA 650 fraction. It was also shown that part of newly synthesized molecules of 18s RNA are broken down in the course of processing. The data obtained confirm the previously made assumption that degradation of some rRNAs in the course of maturing represents a mechanism of maintenance of a constant number of ribosomes in the cell.
