[Experimental approaches to treatment of prostatic adenoma with the luliberin analog surfagon].

Therapeutic efficacy of surfagon, an analog of LH-RH which was injected subcutaneously at a daily dose of 100 micrograms/kg for 30 days, was investigated in ACI male rats with induced prostatic adenoma. This drug was shown to be effective proceeding from a morphological study and measurement of the prostate. Measurement of blood testosterone levels by radioimmunoassay showed a decrease in this level in rats, treated with surfagon, i. e. a therapeutic effect of the drug in prostatic adenoma was accompanied by a decrease in the level of androgens.

[Effects of hypokinesia on cardiac activity in rats with high and low spontaneous motor activity].

Hypokinesia effects on cardiac contractivity and vegetative regulation were studied in rats with high and low spontaneous motor activity. It was shown that hypokinesia promotes depression of the stroke volume and decrease of sympathetic nervous activity in rats with low motor activity. At the same time, in rats with high motor activity these parameters were not significantly affected by hypokinesia. The conclusion is made about a relationship between resistance to hypokinesia and spontaneous motor activity.

[Amplification of the phage lambda DNA sequence by polymerase chain reaction using thermostable DNA polymerase].

The efficiency of phage DNA amplification by the method of polymerase chain reaction (PCR) with Tth DNA-polymerase was studied for optimization of PCR conditions. The effect on amplification efficiency of medium ionic strength and pH, the presence of univalent cations, detergents, gelatin, ATP, pyrophosphate, SH-reagents and ratio of concentrations of Mg and dNTPs, primers and template was studied. It has been found that a pH optimum for PCR with Tth DNA-polymerase varies from 8.5 to 9.0. An ionic strength optimum for PCR is about 0.08. The influence of univalent cations on the activity of Tth DNA-polymerase can be expressed as NH4+ greater than Na+ greater than K+. 0.01% Tween-20 significantly increases the efficiency of PCR and 0.01% gelatin inhibits it. Addition of ATP, pyrophosphate, SH-reagents to the reaction mixture did not increase the yield of PCR product. It has been also shown that for the given PCR-system an optimum Mg/dNTPs molar ratio is within the range of 1.5-2.0. An optimum concentration of each of the pair of primers for this PCR-system is about 0.3 microM. The possibility of PCR-amplification of 500-8500 b.p. DNA fragments has been demonstrated.

[Amplification of DNA sequences in Epstein-Barr virus and human immunodeficiency virus using DNA polymerase from Thermus thermophilus].

Using thermophilic DNA-polymerase from Thermus thermophilus we have amplified by polymerase chain reaction (PCR) specific DNA sequences of Epstein-Barr virus (EBV) and human immunodeficiency virus (HIV). DNA-polymerase from Thermus thermophilus (the molecular mass of 80 to 86 kDa) differs in its physico-chemical properties from DNA-polymerase from the Thermus acquaticus (the molecular mass of 62 to 68 kDa). To amplify the specific EBV DNA sequence oligonucleotide primers for the virus replicon region (oriP-region) were used. As a result of amplification, a specific 405 b.p. DNA fragment was produced. Primers for the virus Gag region were used for amplification of HIV DNA. The possibility to conduct amplification cycles under two temperature conditions was demonstrated.

[Regulation of expression of the Escherichia coli htpR gene by antisense RNA].

A vector plasmid directing the synthesis of antisense RNA of Escherichia coli gene htpR has been constructed. For this fragment of htpR gene encoding the synthesis of N-terminal sequence of HtpR protein has been cloned in this plasmid in the opposite orientation under the control of Pr promoter of bacteriophage lambda. Under conditions of Pr transcription the induction the growth of bacterial cells carrying the recombinant plasmid was delayed and the rate of degradation of puromycin polypeptides was decreased 3-fold. Vectors constructed on the basis of such a principle may be used for thermoinducible synthesis of heterologous proteins with a simultaneous correction of the intracellular proteolysis.

[Thermo-regulated expression of the htpR gene under the control of the PR-promotor of bacteriophage lambda induces supersynthesis of heat shock proteins].

The mechanisms of induction of heat shock protein synthesis in E. coli have been studied. For this purpose plasmids in which htpR gene expression is controlled by the PR-promoter of bacteriophage lambda and by the Trp-promoter have been constructed. An effective induction of heat shock proteins requires both an increased content of htpR protein and additional cofactors formed in the cell under heat shock conditions.

[Expression of aminoglycoside phosphotransferase gene is under the control of recA promoter].

A novel expression vector using the 300 bp promotor-operator fragment of the recA gene of Escherichia coli has been constructed. The strength of the recA promotor was examined by assaying aminoglycoside phosphotransferase (APT) activity expressed from APT gene placed downstream of the promotor. We have observed, that some plasmids, containing N-portion of recA gene caused a large increase in radiosensitivity of host bacteria cells.

[Accumulation of N-terminal fragment of recA protein in the htpR- mutant impairs the SOS-function of Escherichia coli cells].

It has been established that the plasmid encoding the N-terminal domain of RecA protein (50 amino acids residues) disturbs SOS functions of htpR- mutants of E. coli. This is expressed in increased UV-sensitivity of strains and in the poor transcription of the aminoglycosidephosphoransferase gene in the hybrid operon recA APT. The discovered effect is due to an increase in the stability of the peptide whose accumulation seems to impede the production of RecA protease. Mutation of gene lon does not lead to the accumulation of N-terminal fragment, therefore an increased stability of the peptide in htpR mutants is probably related to the absence of an unidentified peptidase.

[Regulation of expression of the htpR gene in Escherichia coli].

The hybrid operon htpR: ATP was obtained, in which the expression of the aminoglycoside phosphotransferase gene is controlled by the promoter of the htpR gene of Escherichia coli. It was shown that the transcription of the htpR gene is not induced during the "heat shock". The basal level of the htpR gene transcription in htpR mutants was found to be two times higher than that in the strains of the "wild type". These findings suggested that the expression of the htpR gene is autoregulated. Hybridization experiments demonstrated that the genome of Pseudomonas bacteria contains the htpR-like gene.
