[Effect of new synthetic anxiolytic selank on gastric wall blood flow and mesenteryc lymphatic vessels contractility in anesthetized rats].

The influence of a new heptapeptide Selank on microcirculation in anesthetized white rats was investigated. Selank (Thr-Lys-Pro-Arg-Pro-Gly-Pro) is a new synthetic anxiolytic which exerts obvious antiulcerogenic action and promotes healing of experimental ulcers. Action of the peptide on gastric blood flow in the stomach by using the method ofhydrogenic clearance and Selank action on mesenteric lymphatic contractility by microscopical observation in situ, were studied. Selank fail to influence basal gastric blood flow but it can normalize blood flow reduced by indomethacin. The study of dose-depended of Selank effect on lymphatic vessels contractility showed that its low concentration (10(-12)--10(-14) M) enhanced amplitude and increase frequency of lymphatic vessel contractions which indicates an enhancement of lymphatic flow. The high doses of peptide (10(-6)--10(-10) M) also augmented the contraction amplitude but decreased its frequency. The maintenance of adequate blood flow and lymphatic vessel contractility can be one of the mechanisms of the Selank antiulcerogenic properties.

Possible mechanism underlying the effect of Semax on the formation of indomethacin-induced ulcers in rats.

Intraperitoneal injection of Semax (synthetic analogue of ACTH4-7, MEHFPGP) in a dose of 50 mg/kg produced a protective effect on rats with experimental indomethacin-induced ulcers. Experiments on narcotized rats showed that Semax in the studied dose had no effect on basal blood flow in the stomach, but prevented reduction of blood flow induced by indomethacin. The antiulcer effect of Semax is probably related to improvement of blood flow in the gastric wall disturbed by indomethacin.

[Preoperative embolization of the portal vein in malignant tumors of the liver: preliminary results].

Preoperative embolization of the right portal vein and following hepatic resection were performed in seven patients with primary and secondary liver malignancies. The indication for the procedure was a small amount (30%) of the future remnant liver (FRL). The ultrasound-guided percutaneous transhepatic puncture of the portal vein and its embolization with gelatin sponge or silicon polymer was used. No complications of the procedure were noted. In 19-56 (mean 33) days computed or magnetic resonance tomography showed a significantly increased FRL volume (mean 40%). Right or extended right hepatectomy was performed in 5 and 2 patients respectively without lethal outcomes. A conclusion is made that the preoperative portal vein embolization is a safe and technically simple procedure that reduces the risk of postoperative hepatic failure after major liver resection.

[Correction of the stomach blood flow as a mechanism of anti-ulcer effects of short proline-containing peptides].

Intraperitoneal administration of the PGP did not change basal mucosal blood flow, whereas the PG and GP significantly decreased it. Ethanol and Indomethacin caused a rapid and stable decrease in the blood flow. Administration of the PGP prior to ethanol abolished this effect. Injections of the PGP and PG following Indomethacin administration prevented reduction of the mucosal blood flow. Administration of the GP did not change the blood flow decrease induced with Indomethacin. The mucosal blood flow correction seems to be one of the possible mechanisms of the PGP and PG antiulcer effect. The effect seems to be realised through a change in the CNS activity.

[Structural reconstruction of chemo-sensitive biomembranes during the action of low molecular weight compounds using spin resonance].

Using the spin probe technique, the changes in the supramolecular structure of the central nervous system synaptic membranes and of olfactory hair membranes of Rana temporaria induced by low molecular weight organic substances of different chemical nature, were investigated. It was found that the membrane structures under study differ considerably in their sensitivity to chemical stimulation and in temporal kinetics of structural transitions. A correlation between physiological parameters of olfactory perception of the compounds used and the parameters of structural transitions in olfactory hair membranes was established. The interrelationship between the experimental data and chemoreception of odorants and mediators as well as the applicability of these preparations for membrane screening of biologically active substances are discussed.

Preoperative portal vein embolization for liver malignancies.

BACKGROUND/AIMS: To study effectiveness of preoperative portal vein embolization before extensive hepatic resection in patients with primary or secondary liver malignancies. METHODOLOGY: Between December 1997 and May 2003, right portal vein embolization was performed in 24 patients. The indication to the procedure was a small amount (< 30%) of the future remnant liver. Ultrasound-guided percutaneous transhepatic puncture of the portal vein and embolization of its right lobar branch with Lipiodol, ethanol, gelatin sponge and/or steel coils were used. RESULTS: There were no complications of the procedure. In 19-56 (mean, 30) days, CT or MRI showed significant increase of the future remnant liver volume from 25% to 35% of the whole liver; the mean increase was 40%. Laparotomy was performed in 15 patients. Right, extended right hepatectomy, and repeated resection were performed in 7, 6, and 1 patient respectively, while one patient underwent only exploration because of tumor progression. Of 14 resected patients, 11 showed no postoperative liver failure. The latter was medically cured in two of three remaining patients, while one patient (future remnant liver < 30% even after the embolization) died from liver failure in 20 days after the surgery. Hepatic resection is planning in 4 of 9 non-operated patients. The 5 patients were not operated because of tumor progression. CONCLUSIONS: Right portal vein embolization is a safe and technically simple procedure that decreases the risk of postoperative hepatic failure after major liver resection.

[Various aspects of the effect of heparin-serotonin complex on blood and blood flow].

High-molecular heparin-serotonin complex (3:1 weight ratio) was obtained in vitro. The complex, its components, and heparin-serotonin mixture were introduced intraperitoneally in rats, and the resulting effects were registered 10, 30, and 60 min later. The complex had anticoagulant and lytic activities distinct from those of plasmin. According to decrease activity of factor XIII and decreased concentration of fibrinogen, the complex stimulated the anticoagulant system. In addition, the heparin-serotonin complex penetrated the hematoencephalic barrier and decreased the rate of brain bloodstream. The observed physiological properties of the heparin-serotonin complex were distinct from those of serotonin, heparin, and their mixture.
