Studies on clonogenic hemopoietic cells of vertebrate in space: problems and perspectives.

Hemopoietic tissues were studied in vertebrates launched aboard the Soviet (Russian) biosatellites ("Cosmos-1129, 1514, 1667, 1887 and 2044"; "Bion-10 and 11") between 1980 and 1996. In the bone marrow of rats exposed to spaceflight conditions, a statistically significant decrease in cell number was revealed in the progenitor cell compartment accounting for the compensatory response of granulocyte-macrophage (CFU-gm) and erythrocyte lineages (BFU-e and CFU-e) and in the compartment of multipotent hemopoietic stem cells (CFU-s), which is responsible for the permanent renewal of hemopoietic tissue. The number of stromal fibroblastic progenitors (CFC-f) in the bone marrow of these rats was also reduced. Apparently, changes in the hemopoietic stroma damage the hemopoietic microenvironment and, hence, may be responsible for changes observed in the hemopoietic tissue proper. Attempts were made to develop methods for analyzing morphologically indiscernible clonogenic hemopoietic cells of newts, and studies on the effects of spaceflight factors on these cells were performed. The results showed that the numbers of clonogenic cells in newts of the flight group newts were significantly lower than in control newts. The data obtained are used as the basis for formulating the problems to be studied, drawing up a program for further research on the effects of spaceflight factors on stem and other clonogenic hemopoietic cells, and developing new experimental models for analyzing stem cells, the state of the hemopoietic stroma, etc.

[Ultrastructural characteristics of stromal mechanocytes and their interaction with hematopoietic cells in regenerating bone marrow transplants].

Extramedullar grafts of the bone marrow have been studied electron microscopically 2-8 days after transplantation. The data on ultrastructural peculiarities of the stromal mechanocytes have been obtained at various stages of their differentiation, as well as topographic interrelationships between the mechanocytes and the hemopoietic cells. Digital junctions are described between the stromal mechanocytes (primitive) and the hemopoietic cells (in 3-day-old grafts) which are, probably, the first morphological signs demonstrating restoration of the hemopoietic microenvironment in the bone marrow grafts.

[CFUs of the normal and regenerating liver in the sexually mature mouse].

Functional properties of CFUs have been studied in intact and regenerating liver of mice. According to a number of properties (proliferative activity, character of colony growth) CFUs in the liver are similar to CFUs in the peripheral blood and, probably, make the same population. In the regenerating liver relative contents of CFUs 3-5 days after a partial resection is substantially increasing. Concentration of CFUs (endogenic) increases significantly also in a locally injured and regenerating lobe, comparing to the intact lobe that is in the same organ. The transplanted bone marrow CFUs prevail in number in the regenerating liver over the intact liver. It is concluded that increasing contents of CFUs in the regenerating liver depend mainly on its creased property to invade and/or to hold the extrahepatic CFUs.

[Capacity of intact and regenerating liver to trap CFUs].

The normal and regenerating mouse liver is capable of trapping transplanted bone marrow CFUs, with this capability being increased in the regenerating liver. Inactivation of macrophages (Kupffer cells) by silica particles sharply reduces this ability, thus pointing to direct or indirect involvement of macrophages into the trapping of exogenous CFUs.

[Hematopoietic stroma and the regulation of hematopoiesis in extramedullary bone marrow transplants in mice].

The bone marrow fragments were transplanted under the renal capsule and "stromal function" of the transplants during up to 1.5 years was estimated by the numbers of CFU-S, myelokaryocytes and the weight of newly formed bone tissue. On the basis of the published and authors' data, a conclusion was drawn that the ability of the stromal precursor cells of bone marrow to provide for the growth of hemopoietic territories is limited by the size of medullary cavity; outside its limits this ability is not only realized, but may be additionally stimulated by some distant factors (preirradiation of the recipients in the given case). The self-maintenance ability of the stromal mechanocytes from the old mice during 43 months was shown by means of retransplantation. On the basis of electron microscopy and published data, a suggestion was put forward that fibroblastic reticular cells might be ancestral elements of osteogenic differentiation.

[3H-thymidine incorporation into the macrophages in the process of eye regeneration in adult tritons].

Changes in the number of labelled macrophages in the regenerating eye cavity of the adult common newts were studied within 2 to 12 days following the injection of 3H-thymidine and removal of retina and lens from the eyes. A small number of labelled macrophages was found in the eye cavity (0,2--5.9%) at all regeneration stages under study upon the pulse 3H-thymidine incorporation. Their number rapidly increased and attained by the end of observation (12 days) 73%. These results suggest that mainly mature non-dividing forms of macrophages which completed the mitotic cycle S-phase in the places of their active reproduction migrate in the eye cavity. The sharp increase of the number of labelled macrophages in the eye cavity is determined by the migration of those macrophages the precursors of which were labelled as a result of both the pulse 3H-thymidine incorporation and reutilization of the labelled precursors of DNA synthesis. New portions of labelled macrophages migrate in the eye cavity within 2 to 4 days.

[Electron microscopic analysis of the fibroblast differentiation of amebocytes in the common snail, Lymnaea stagnalis L. (Mollusca, Gastropoda) in a lesion focus].

The amoebocytes and fibroblasts of intact connective tissues of the common pond snail foot, as well as the amoebocytes of the burn wound were studied by means of electron microscope. The found transitional forms from the amoebocytes to the fibroblasts allowed to conclude that there were cells--precursors of fibroblasts among the amoebocytes migrating in the wound. A tentative scheme of histogenetic series of amoebocytes is given.

[The relationship between hemopoiesis and osteogenesis in several species of rodents].

Differentiation of the bone marrow transplants of rats, guinea pigs and dwarf hamsters placed under renal capsule of irradiated mice was studied. Active osteogenesis occurred in the transplants of rats and guinea pigs; as to hemopoiesis-it was irregular in rats and was arrested in guinea pigs. On the contrary, in the transplants of dwarf hamsters an intensive hemopoiesis was maintained and osteogenic capacity of the stroma was poor. It is supposed that the bone tissue in some rodent species was not a prerequisite for normal maintenance of myeloid hemopoiesis, while in others-primary bone stroma formation was necessary for the subsequent functioning of hemopoietic tissue.

[Differentiation of hematopoietic and stromal elements of extramedullary bone marrow transplants in irradiated recipients].

The growth of bone marrow transplants under the renal capsule is stimulated in the mice irradiated at a dose of 700 r. The size of transplants increases, primarily, at the expense of hemopoietic tissue. The osteogenic potencies of stromal elements remain at a high enough level as well. Some features which characterize the decrease of viability of hemopoietic cells may be due to incomplete restoration of the system of sinusoidal capillaries.
