Multinucleated tumor cells and malignant melanoma.

Multinucleated cells (MC) were counted in cell preparations obtained by dissociation of representative part of tumor lesions immediately after excision. MC were present in almost all specimens examined (39 advanced primary lesions, 90 lymph node metastases and 33 dermal plus subcutaneous metastases); in one third of the samples they were very rare (less than 1% of all cells). There were no significant differences in quantity of MC between primary tumors, node metastases and dermal plus subcutaneous metastases, between node metastases seen early in the course of the disease and those seen later, and between regional node metastases taken from Stage II patients with rapidly progressing disease and regional node metastases taken from patients of the same stage whose disease-free intervals were longer. No unique pattern of similarities or differences in quantity of MC was found when comparing autologous tumor samples excised simultaneously and/or successively during the course of the disease.

HLA-DR antigen expression on melanoma metastases and the course of the disease.

Melanoma cells freshly isolated from regional lymph node metastases of 59 stage II malignant melanoma patients were analyzed by indirect immunofluorescence staining with monoclonal antibody TAL 1B5, detecting the HLA-DR alpha chain. The expression of HLA-A,B,C antigens, using antibody W6/32, was also investigated in 45 of these cases. There were no substantial differences in the course of the disease with respect to the percentage of positive cells. In 13 malignant melanoma patients two to four simultaneous and/or successive metastases (both locoregional and distant-subcutaneous) were analysed for HLA-DR. With simultaneous metastases (7 cases) the percentage of HLA-DR-positive cells was mostly very similar, and in no case was there more than 25% variation. As to successive metastases (9 cases) the percentage of HLA-DR-positive cells remained practically unchanged or decreased during the course of the disease.

Epidemiological aspects of childhood cancer incidence in Czech regions.

In this descriptive epidemiological report a total of 5100 childhood malignancies registered in Czech regions during the period of 1963-1985 was analyzed. The incidence of these tumors compared in years of this time period has been relatively constant, we did not find any significant changes in trends. That is why the average annual incidence rate, i.e., 10.8 in males and 8.7 in females per 100,000 sufficiently characterized the epidemiological situation in this respect. Those parameters allow, by knowledge of demographic prognosis, to estimate expected numbers of tumors in further years. The highest incidence occurred in boys and in the lower age groups of both sexes. All differences presented in this report were tested and were found to be significant. The percentage ratio of solid tumors, preferentially those of bone and skin of both sexes, and brain tumors in females in the period of 1976-1985, as compared to the period of 1961-1970, decreased, whereas there was a significant increase of systemic malignancies and soft-tissue sarcomas. Finally, the geographic differences of incidence are described, and the possible causes of the data presented are discussed.

Skin tests with autologous cholesteryl hemisuccinate-treated tumor cells, DNCB and PPD and their relationship to prognosis in malignant melanoma patients.

Skin tests with autologous cholesteryl hemisuccinate-treated tumor cells were performed in Stage II malignant melanoma patients. In the majority of cases an evident reaction having features of delayed type hypersensitivity was noted. No significant correlation between this reactivity and subsequent course of the disease was found. The same was true for the results of DNCB and PPD skin testing. An analysis of the results of all three tests in the same patient revealed no prognostic significance of this score either.

Course of breast cancer disease and ABO blood groups.

The clinical records of 551 breast cancer patients were retrospectively analysed. Several parameters have been examined: age at presentation, distribution of stages at the time of diagnosis, incidence of any progression and "progression-free" interval after primary treatment, incidence of distant dissemination and distant metastasis-free interval, cancer mortality, time of survival from presentation and time of survival from detection of distant metastases (stage at presentation being taken into account in all evaluations). The results within various ABO blood groups were compared. There were no substantial differences in these parameters within different blood groups. The possible modifications of ABH (O) isoantigens on tumour cells and the immune response to these alien antigens is discussed.

No difference in delayed hypersensitivity between breast and cervical carcinomas.

DNCB and PPD skin testing was performed in 130 breast and 110 cervical cancer patients. BATES' instruction with a plea for uniformity was used [1]. Patients were tested while being diagnosed and prior to the treatment. No significant differences either in the frequency of a reactivity or grade of response in corresponding stages of breast and cervical cancer were found.

DNCB and PPD skin testing in breast cancer.

DNCB and PPD skin testing was performed in 152 breast cancer patients. Bates' instruction with a plea for uniformity was used (Cancer, 43, 1979, 2306). Majority of patients were tested while being diagnosed and prior to the treatment. There were no differences in the reactivity within early operable breast cancer patients (Stage I and II) with respect to nodal involvement. Patients with N1 reacted in the same manner as those with N0. The reactivity of patients with locoregionally advanced disease (Stage III) was similar to that of Stage I and II patients. Significantly lower responsiveness was found in Stage IV patients, the depressed response to DNCB being more pronounced than to PPD.

PKU locus: genetic linkage with human amylase (Amy) loci and assignment to linkage group I.

The linked alpha-amylase loci Amy 1 and Amy 2 were evaluated for their linkage relationship to the PKU locus using data collected from two (one Czech and one Polish) groups of families. The five sibships informative for Amy 1:PKU give a z score of 1.505 at theta = 0.00 and the eight sibships informative for Amy 2:PKU give a z score of 2.709 at theta = 0.00. Due to the tandem position of Amy 1 and Amy 2 loci, these data could be combined, and linkage between Amy and PKU loci established with a z score 4,214 at theta = 0.00. The practical significance of the linkage, especially for identifying PKU allele carriers, is emphasized.
