[Isolation and properties of an extracellular proteinase of Staphylococcus aureus].

Serine proteinase splitting the peptide bonds which are formed by carboxyl groups of dicarboxylic amino acids was isolated from the supernatant of the Staphylococcus aureus culture liquid. It is similar to the enzyme isolated by Drapeau from Staphylococcus aureus strain V8 in its specifidity, molecular weight, amino acid composition, existence of two pH optima (pH 4.6 and 8.2). But there are some differences between the two proteinase in the content of dicarbonic amino acid residues. It was found that the enzyme can exist in two molecular forms.

[Proteolysis of horse blood serum proteins at various stages of antitoxic sera production by the Diaferm-3 method].

When preparing antitoxic sera by the method "Diaferm-3", it was found that proteolysis of the horse blood serum occurs not only at the fermentation step (pepsin treatment at the enzyme-substrate ratio of 1/10, t 20-23 degrees for an hour at pH 3.3 and for the next hour at pH 4.2), but also at the heat treatment step (45 min at pH 4.3, t 56-58 degrees in the presence of ammonium sulfate at a concentration of 140-145 g/l). At the fermentation step immunoglobulins do not split completely into F(ab')2 fragments, 40% of the antitoxic activity being lost at this step. Some ballast proteins--albumin, fibrinogen, etc.--quickly split up into peptides, while other proteins undergo only limited proteolysis. Structural destabilization of serum proteins during thermodenaturing is favorable for the pepsin action and provides for a complete conversion of immunoglobulins to F(ab')2 and Fab' fragments. At this step about 15% of the antitoxic activity is additionally lost. Thus, proteolysis at the thermodenaturation step is an essential part of the "Diaferm-3" process.

[The reaction of pepsin and pepsinogen with dicyclohexylcarbodiimide].

The reaction of pig pepsin and pepsinogen with 3H-dicyclohexylcarbodiimide (DCC-T) is studied. It is found that about three residues of DCC-H incorporate into pepsin molecule and about four residues--into pepsinogen molecule under the excess of carbodiimide. The incorporation of DCC-T into protein results in a deep enzyme and zymogen inactivation: pepsine retains 20% of the proteolytic activity with respect to hemoglobin, and pepsinogen completely loses its potential proteolytic activity. It is found that a part of DCC-T residues attaches to N-terminal part of pepsinogen, which splits off under the activation of proenzyme in acid medium. It is demonstrated that the interaction of DCC-T with carboxyl groups of dicarbonic amino acids results in the change of isoelectric point of pepsin and pepsinogen.

[The interaction of carriers with proteolytic enzymes used for enzymatic synthesis of peptides in organic solvents].

The adsorption of some proteolytic enzymes on inorganic solvents (Silochromes C-80 and C-120, macroporous glass CPG-10, and celite 535) and the effect of sorption-desorption processes on the activity and stability of the enzymes have been studied. The ability of the enzymes to be adsorbed on the carrier depended on the specific surface of the carrier and properties of the enzyme. Adsorption-desorption processes did not affect the activity of the enzymes. Acetonitrile had no noticeable effect on the activity of the enzyme adsorbed on the inorganic carrier. The enzymes adsorbed on such carriers catalysed reactions in media with a low (4-5%) content of water, since the carrier seems to protect the enzyme from inactivation with organic solvents. The loading of carrier with enzyme influenced the rate of the enzymatic reaction, the optimal loading corresponding the value of the maximum adsorption of the enzyme on the carrier.

[A gas chromatographic analysis of the fatty acid composition of Yersinia pestis].

The fatty acid composition of Y. pestis strains, the causative agent of plaque, has been studied. Y. pestis cells have been found to contain great amounts of palmitoleic acid, methylenehexadecanoic acid, oleic acid with elaidic acid, palmitic acid and pentadecanoic acid. Lauric acid, myristic acid, 3-oxymyristic acid and methyleneoctadecanoic acid have been detected in moderate amounts. Vaccine strains, virulent museum and newly isolated strains, while differing in their antigenic structure, have proved to be uniform in their fatty acid composition. In Y. pestis cels, grown on a solid medium for a longer period and at higher temperature, an increased proportion of saturated acids is observed.

[Multiple genes of delta-endotoxins from Bacillus thuringiensis subspecies galleriae].

Cloning and expression in E. coli delta-endotoxin genes cryIAb7, cryIG and, cryIX from Bacillus thuringiensis ssp galleriae has been performed. Restriction mapping and partial sequencing have shown the identity of the 3'-terminal parts of cryIG and cryIX, although their 5'-terminal halves corresponding to the toxin are unique. Sequencing 5'-flanking region of cryIX revealed no translation initiation site, which indicates a nonfunctional state of the gene. The clusterized locating of the cryIG and cryIG genes has been shown. The absence of a promoter-like structure in the 5'-flanking region of cryIG suggests transcription of the gene as a bicistronic mRNA with cryIX. The extremely high homology of the cryIG and cryIX 3'-terminal parts (2 kB long) suggests a recombination act in the gene origin.

[The fatty acid composition of the cellular structures of Yersinia pestis].

Fatty acid composition of lipopolysaccharides (LPS) and cytoplasmic membranes (CPM) of Yersinia pestis [correction of the plaque microbe] has been studied. Concentration of certain acids in the LPS content proved sharply different and thus the strains were separated into groups. High content of 3-oxymiristinic acid was a distinction of vaccine strains. Virulent strains of one of the groups regularly differed from the rest of virulent strains by high content of laurinic acid. Laurinic and two nonidentified acids, which were not found in the virulent cultures, were present in CMP lipids of vaccine strains.

[Zoonotic infections in plague foci of the Republic of Kazakhstan].

Examining zoonotic infections in the plague foci in Kazakhstan over a decade has revealed their wide spread of vectors of pseudotuberculosis, intestinal Yersinia infection, pasteurellosis, listeriosis, salmonellosis, erysipeloid. Some causative agents under study can regulate the number of the main vector in the foci of plague, hamper the bacteriological and serological diagnosis of plague and be of predictive value in the assessment of the epizootic situation.

[Activity of endogenous proteinases in glomerulonephritis in children].

Protease activities in serum and urine of 116 children with glomerulonephritis and 16 healthy children were tested using chromogenic peptide substrates Z-Dala_Leu-Arg-pNA and Glp-Ala-ALa-Leu-pNa. We found the dependence between activity of serine proteinases and clinical, morphological forms of primary glomerulonephritis and hypertension.
