The clinicoimmunological study revealed immunocompetence dysfunction in 28 patients with idiopathic ventricular arrhythmias resistant to ritmilen. After sodium nucleinate immunomodulation, ritmilen therapy proved to be beneficial in 46.6% of the patients. A positive result was obtained only in the patients who showed the same or close values as in healthy individuals. The comparison has led to the conclusion that idiopathic ventricular arrhythmias are directly related to immune dysfunction and to the recommendation that sodium nucleinate should be used in the multimodality therapy of the arrhythmias.
It is shown that sodium nucleinate applied in a complex with the generally accepted nonoperative therapy corrects the immune status of patients with atherosclerosis of the lower limb arteries in stage IIB circulatory disorders. The number of T-lymphocytes (45.2 +/- 1.22 before and 68.4 +/- 1.85 after treatment) and the percentage of B-lymphocytes (25.3 +/- 0.66 before and 19.3 +/- 0.66 after treatment) were normalized. The percentage of O-lymphocytes reduced 2.4 times, the number of T-lymphocytes sensitized to the arterial intima dropped to normal. The indices of humoral immunity improved: IgG reduced to the level encountered in healthy individuals, the level of circulating immune complexes reduced 2.23 times, the IgA concentration, however, remained just as low as before treatment. The indices of nonspecific immunity became normal: complement increased, the number of NST+ neutrophils reduced by half, the number and phagocytic activity of neutrophils significantly increased. In the group of patients who were given sodium nucleinate 18 had a good clinical effect.
Based on clinical, laboratory, bacterial, allergologic and immunologic studies a group of patients with infectious allergic urticaria (IAU) was distinguished. The immunologic examination of 30 patients with IAU identified a decrease in phagocytosis. Sodium nucleinate (SN) was employed as an immunomodulator as compared to placebo. SN was noted to produce an optimal immunomodulating effect, provided it was combined with antibacterial drugs. The use of SN in patients with IAU produced a good clinical effect elucidated for 6 months. It is recommended that SN be used as an immunomodulator in the multimodality therapy of IAU and introduced into the practice of the allergological rooms.
It was found out that macrophage superoxide production is stimulated by intracellular contacts. The high level of the nitro-blue tetrazolium salt reduction in individual cells non-contacting with others is reached when using fibronectin or serum-opsonized zymosan but not phorbol myristate acetate or chemotactic formyl-peptide as activators. The reaction enhancement in non-contacting macrophages stimulated with phorbol ester occurs in the presence of Arg-Gly-Asp-Ser synthetic peptide.
Study of the immunologic reactivity of 104 patients with chronic obstructive bronchitis revealed the deficiency of cellular immunity for the most part. The use of timoptin, sodium nucleinate and their combined application raised the clinical efficacy of the treatment of patients with chronic obstructive bronchitis, significantly reduced the concentration of medium-weight molecules in blood serum, and corrected immunity disorders.

In 116 patients with atherosclerosis of the arteries of the lower extremities in stages I-IV of the impairment in circulation and 35 healthy subjects, the immune status was studied. With aggravation of the disease, the number of leukocytes, B-cells, concentration of the IgM, IgG and circulating immune complexes, the number of immature lymphocytes and T-cells sensitizised to antigens of the arterial intima increase. At the same time, the total activity of the complement and phagocytic activity of neutrophilic granulocytes decrease, the relative number of neutrophilic granulocytes, which spontaneously generate a superoxide radical, increases. The likeness of atherosclerosis of the lower extremities to atherosclerosis of the heart vessels was noted.
The immunological action of RNA mononucleotides was studied in animal experiments. The most pronounced activation of macrophagal glycolysis urea cycle, oxidative phosphorylation, lysosomal hydrolases was induced by uridine 5'-monophosphate (5'-UMP) and guanosine 5'-monophosphate (5'-GMP); 5'-GMP also induced the maximum increase of the expression of FC gamma receptors. 5'-UMP ensured cell activation comparable with the total action of all mononucleotides. 5'-UMP and 5'-GMP, used in combination, produced the highest stimulating effect on macrophages, and the addition of low-active adenosine 5'-monophosphate (5'-AMP) to active 5'-UMP did not decrease the stimulating potency of the latter. The stimulating activity of sodium nucleinate exceeded that of all mononucleotides and their combinations. 5'-GMP and 5'-AMP induced the maximum activation of oxygen metabolism, evaluated by chemiluminescence, while 5'-UMP and cytidine 5'-monophosphate (5'CMP) proved to be inactive. The shift of the phosphate group to the third carbon atom or the production of the oligonucleotide 5'-UMP consisting of 5-15 nucleotides resulted in the appearance of the capacity for stimulating oxygen metabolism in macrophages. Their in vitro cultivation with 5'-GMP and 5'-AMP induced the maximum increase of cell spreading in comparison with other mononucleotides, while the maximum increase of phagocytosis was ensured only by 5'-UMP and 5'-GMP.2+ 5'-UMP and 5'-GMP enhanced nonospecific resistance to Salmonella typhi infection, and sodium nucleinate, to Pseudomonas pseudomallei and Pseudomonas mallei infections.

The heterogeneity of resident peritoneal macrophages and peritoneal macrophages was studied in different periods following oral administration of sodium nucleinate according to their ability to bind and phagocytize sheep erythrocytes opsonized by means of specific rabbit IgG. Using a mathematical method developed earlier, it has been possible to demonstrate that resident peritoneal macrophages can be divided into two subpopulations--actively and poorly binding macrophages but, after activation by sodium nucleinate--into three subpopulations. Fractionation of the macrophages according to their ability to adhesion within a temperature gradient has shown that the same peaks are traced in the fractions as in the overall pool of cells, but in different quantitative ratios. It has also been demonstrated that phagocytic activity is reduced in macrophages capable of adhesion to plastic at lower temperatures.
A method of the Poisson fitting of flow cytometry data is suggested. The estimation of distribution parameters was carried out by the steepest descent method. The phagocytosis of fluorescent latex particles by murine macrophages was used as an experimental model. The results obtained by the Poisson fitting of cytofluorograms were in good agreement with the visual estimation of macrophage distribution with regard to the amount of uptaken particles. A high value of correlation index (more than 0.99) showed the validity of the suggested fitting method. The applicability of the method to cytofluorimetric analysis of human lymphocytes was successfully demonstrated.

Sodium nucleinate and its mononucleotides have been found capable of stimulating the multiplication and virulence of shigellae, as well as their sensitivity to antibiotics, in cultures grown in nutrient media.
Using the phenomenon of the formation of the active forms of oxygen by phagocytes in the process of adhesion, changes occurring in the cell membrane under the influence of sodium caseinate, macrophage migration inhibition factor, aggregated IgG and hydrolytic enzymes could be registered. Sodium caseinate inhibited chemiluminescence during the first 10 minutes in the adhesion of mouse peritoneal macrophages, human lung macrophages and adhesive marrow cells, while large doses of this preparation activated chemiluminescence at the latter stages of adhesion. Macrophage migration inhibition factor induced the rapid and prolonged formation of the active forms of oxygen in the process of macrophage adhesion, aggregated IgG decreased this cell function, and the subsequent phagocytosis of zymosan was accompanied by increased chemiluminescence. The experimental and practical possibilities of this phenomenon are discussed.

Heterogeneity of resident peritoneal macrophages was studied as revealed by their phagocytosis and binding of sheep erythrocytes opsonized by specific rabbit IgG. It has been shown that the macrophage heterogeneity is due to the presence of active and inactive cellular subpopulations. A mathematical model, based on the Poisson fitting of experimental histograms, was elaborated for the analysis of subpopulation composition of macrophage pool in the test of rosette formation and phagocytosis. The validity of the model was supported by physicochemical isolation of the macrophage subpopulation which was inactive in erythrocyte binding. The macrophage pool was separated into fractions by absorption--elution at different temperatures. Active and inactive macrophage subpopulations were found in all the fractions but in different ratios.
