[Analysis of transgenic tobacco plants carrying the gene for the surface antigen of the hepatitis B virus].

The plasmids carrying the gene encoding the hepatitis B surface antigen (HBsAg) under the control of 35S RNA single or dual promoters of the cauliflower mosaic virus CaMV 35S were constructed. These constructions were used for obtaining transgenic tobacco plants that synthesize the HBS antigen. The presence of HBsAg in tobacco plant extracts was confirmed by the enzyme-linked immunoassay using antibodies against the native HBs antigen. The antigen amount in plants carrying the HbsAg gene under a single 35 S promoter was 0.0001-0.001 of the total soluble protein whereas the use of a dual 35S promoter increased the antigen synthesis to 0.002-0.05% of the protein. The antigen-synthesizing ability was inherited by the offspring. In the F1 plants, the antigen expression varied in different lines comprising 0.001 to 0.03% of the total soluble protein, which corresponded to the antigen amount in the F0 plants.

Use of chromatography for the preparation of homologous tetanus antitoxin.

To obtain immunoglobulin of selected action with a higher titre, techniques for isolating antibodies from a donor's normal immunoglobulin by means of affinity chromatography were used. To obtain an affinity sorbent, tetanus toxoid was used as a ligand. Using an immunosorbent based on the purified ligand, tetanus antibodies were prepared with a specific activity of 65 IU/mg. According to gel-permeation chromatography, the tetanus antibodies obtained were represented mainly by IgG monomer (greater than 90%). The anticomplementary activity of the preparations obtained, being at least 11 mg of protein per 2CH50, does not rule out the possibility of intravenous administration of tetanus antibodies.

Eremomycin--new glycopeptide antibiotic: chemical properties and structure.

By a combination of chemical and spectroscopic (1H and 13C NMR) studies the structure of a glycopeptide antibiotic eremomycin has been elucidated. It is closely related to vancomycin, but differs in sugar and chlorine content. The eremomycin aglycone contains monodechlorovancomycinic acid; the only chlorine atom is situated in the second amino acid after the N-terminal amino acid residue of the peptide. The sugar part is composed of glucose and two residues of an amino sugar shown to be 2,3,6-trideoxy-3-amino-C-3-methyl-L-arabino-hexopyranose (4-epi-vancosamine). One of the amino sugar residues is a component of the disaccharide 2-O-(alpha-L-4-epi-vancosaminyl)-beta-D-glucopyranose, attached to a triphenyl ether moiety; the position of another one is at the serine oxygen in the C-terminal region of the aglycone.

[The antineoplastic antibiotic polypeptide 308 produced by Actinomadura recticatena. Its isolation and physicochemical properties].

A antitumor antibiotic belonging to the group of polypeptide antibiotics containing chromophore was isolated from the culture of Actinomadura recticatena Terekhova, Preobrazhenskaya et Galatenko, 1984, strain 308. Biosynthesis, isolation, physicochemical and biological properties of the antibiotic are described. The results of elemental analysis, the melting point, optical properties, UV, IR and NMR spectra and the data on acid hydrolysis showed that antibiotic 308 was most closely related to antibiotic BBM-928 A.

[Composition and structure of the antibiotic polypeptide 308-I from Actinomadura recticatena].

Antibiotic 308-I isolated from Actinomadura recticatena and the products of its degradation were studied with the methods of electron impact mass spectrometry, chemical ionization with ammonia, field desorption and ion extraction from solution under atmospheric pressure. It was shown that by its composition and structure antibiotic 308-I was identical to antibiotic BBM-928 A.

[Physicochemical properties of octamycin].

Octamycin, an original polyenic antibiotic was developed. The antibiotic is of an amphoteric character. The number and nature of the functional groups were determined by potentiometric titration. It was shown that the antibiotic contained the neutral sugar galactose. The equivalent weight of the antibiotic (990) was measured by potentiometric titration and the content of the sugar in the molecule. With high performance liquid chromatography it was demonstrated that the antibiotic consisted of 2 components at a ratio of 1:2. The molecular weights of the components were evaluated with mass spectrometry.

[The antibiotic varigomycin--a new representative of the carbonyl-conjugated pentaenes].

In screening new antibiotics, varigomycin produced by a new fungal species Streptomyces variegatus Sveshnikova et Timuk was isolated. By physicochemical properties varigomycin belongs to carbonyl conjugated pentaens. It is a new representative of this antibiotic group. Varigomycin differs from the known representatives of this group by the presence of glucose, sugar in its molecule.

Purification of tick-borne encephalitis virus in tris-modified porous glasses.

Macroporous glasses, chemically modified by tris-hydroxymethyl-aminomethane, were used for gel-permeation chromatography of tick-borne encephalitis (TBE) virus suspension. The highest yield of purified virus was observed in the carriers containing not less than 0.01 micrograms-equivalent/m2 surface-attached tris-hydroxymethyl-aminomethane groups at pH of the eluent higher than 7.6. The obtained modified porous carriers appeared to be suitable for the purification of both infectious and inactivated preparations of TBE virus.

[New species of Actinomadura recticatena sp. nov. and its antibiotic properties].

Actinomadura cultures were isolated from soil samples collected in Turkmenistan, the Moscow, Kursk and Volgograd regions. The cultures were described as representatives of a new species (Actinomadura recticatena, Preobrazhenskaya et Galatenko, sp. nov). The hydrolysates of intact cells of the cultures contain mesodiaminopumelic acid, galactose and madurose. The species is characterized by straight spore chains, wrinkled spore surface, cream-colored aerial mycelium and brown substrate mycelium with pinkish or slightly violet shade. The cultures produce no soluble pigments, including the melanoid ones. The red-violet fraction of the pigments responsible for the pinkish and violet shades of the substrate mycelium belongs to the antibiotics of the griseorodin-rubromycin group.

[New species of actinomycete Streptomyces variegatus sp. nov. synthesizing an antibiotic of the alpha-hydroxyketopentaene group].

Stroptomyces strains INA T-511 and INA 3946 were isolated from 2 different soil samples. Both the strains produced an antibiotic of the group of alpha-hydroxyketopentaenes. The antibiotic was active against gram-positive bacteria, yeasts and fungi belonging to Penicillium. In addition, the strains produced a pigment antibiotic of the prodigiosin type and a pigment identified as ferroverdin. The study of the cultural and morphological features and the natural variation of the strains has shown that they belong to a new species, designated as Streptomyces variegatus sp. nov. Sveshnikova et Timuk.

[Distribution of actinomycetes of the genus Actinomadura in the light chestnut soils of Volgograd Province and their antagonistic properties].

Prevalence of the actinomycetes of the Actinomadura genus in the virgin and cultivated light chestnut soils of the Volgograd Region was studied. It was shown that Actinomadura were one of the constituents of the actinomycetous flora of these soils. No significant changes in the number of the Actinomadura and the total number of the actinomycetes were observed in the cultivated and virgin soils. The cultures of Actinomadura isolated from the virgin and cultivated soils belonged to 17 and 18 known species, respectively. Some ecological indices were determined for estimation of the similarities and differences in the species structure of Actinomadura in the virgin and cultivated soils. It was shown that cultivation of the soils increased the species variety, equalization and general variety of Actinomadura. In the process of the species identification of Actinomadura the chemical nature of the pigments of the substrate mycelium of the same group of the isolates and the type culture of A. salmonea ATCC 33580 was studied. The pigment responsible for the colour of the substrate mycelium of A. salmonea belonged to the maduromycin group. The antagonistic properties of the Actinomadura isolated from the soils were studied. The isolates had a moderate antagonistic activity mainly against grampositive organisms.

[Isolation and identification of the antibiotics amicetin and nebramycin formed by a Streptomyces coeruleoaurantiacus 4009 culture].

It was found that Str. coeruleoaurantiacus, strain 4009 produced antibiotics 4009-A and 4009-B belonging to different groups. Antibiotic 4009-A was identified as amicetin belonging to the group of pyrimidine bases and antibiotic 4009-B as the nebramycin complex belonging to the group of aminoglycosides. The identity of the antibiotics was confirmed by the physico-chemical constants, spectral and chromatographic characteristics and their chemotherapeutic activity.

[Isolation and physicochemical study of the components of the new antibacterial antibiotic 2562].

An antibacterial antibiotic complex consisting of 2 components designated as 2562 A and 2562 B is produced by Streptomyces griseovarabilis. The antibiotic was isolated from the mycelium and purified chromatographically on a column with aqueous silicic acid. The study of the components showed that component 2562 A was chlorbiocin, while component 2562 B differed from the known antibiotics of this group. Physicochemical assays demonstrated that component 2562 B differed from chlorbiocin by the absence of the methyl group in pyrrol, which is probably attached to sugar at beta-position. It was found that component 2562 B is a new representative of the antibiotic cumero-glycoside group.

[Formation of the anthracycline antibiotics, beromycin and nogalamycin, by a new actinomycete species, Streptomyces glomeratus sp. nov].

Culture 3980 was isolated from a soil sample and described as a new species, Streptomyces glomeratus sp. nov. It was found that the culture produced simultaneously 2 close anthracycline antibiotics, i. e. nogalamycin and beromycin, which were synthesized mainly in the mycelium. S. glomeratus differed from the nogalamycin and beromycin-producing organisms described earlier.
