[Skin microtransplantation in the surgery of burns (a clinico-experimental study)].

On the basis of experimental and clinical investigations the authors have shown that extensive burns in patients with true deficiency of donor resources might be treated by the method of micro-transplantation of the skin which consists in plasty of the wounds by pieces of auto-skin of small size. The reduction of size of the skin pieces gives the enlargement of the total line of marginal epithelialization and great plasty coefficient as compared with traditional methods of surgical treatment.

Animals

[Wound epithelialization in rats depending on the quantity and orientation of microautotransplants].

Patches of autoskin (0.5 x 1.0 mm) were grafted on a full-thickness rat back skin wound (5.4 cm2). As a result of "island" epithelialization from microautotransplants (MATR), the skin defect underwent repair. It is shown that the advance of the wound re-epithelialization did not depend on MATR number and their orientation in relation to the wound surface. The use of MATR allows to restore the skin defect with the plastic coefficient 1:100. This reduces the deficiency of donor skin at extensive burns.

Animals

Ability of microautotransplants to re-epithelialize deep and extensive skin injuries.

Experiments with rats of different ages have established the potential of microautotransplants (MATR) of skin (0.5-3.0 mm2 in size) to produce neoepidermis on fresh and granulating wounds. The MATR induced a 40-fold increased rate of closing of the wound surface. Contraction of the re-epithelialized wounds followed the same trend found for an open wound. The rate of contraction of re-epithelialized wounds depends on the age of the rat and the degree of maturity of the granulation tissue. The most extensive contraction was observed in young animals. Neoskin does not contain skin appendages, shows increased exfoliation of horn flakes from the neoepidermis surface and contains epitheloid cysts filled by a horny substance. The use of the MATR technique in the clinic also leads to wound re-epithelialization. Human neoskin histologically is similar to that of animals. These results suggest that the MATR technique is an alternative approach in the treatment of extensive skin burns.

Adult

[Systemic osteoporosis in women].

This paper is based on investigation of biopsies from the upper flaring portion of the ilium of 53 women, sick of systemic osteoporosis. The results of quantitative evaluation of the osseous tissue parameters by means of histomorphometric method allowed to reveal systemic osteoporosis of 4 types, note distinctions between osseous tissue loss mechanisms in case of systemic osteoporosis and age atrophy of bones. Comparison of osseous tissue quantitative parameters with the peculiarities of microanatomy of trabeculae allowed to verify the degree of disturbance of such remodelling mechanisms as resorption and osteogenesis for each of 4 types. The possibility of fractures of axial skeleton and limb bones at normal value of spongy bone in biopsies from the upper flaring portion of the ilium has been demonstrated.

Adult

[Micro-autotransplantation of the skin. Changes in size and morphology of neo-skin].

A new technique of wide wound skin epithelialization by free transplantation of autologous skin fragments 1 mm2 in size is demonstrated. The experiments were carried out on strainless rats of different age, 100 per cent epithelialization of wound 5 mm2 in grafting skin micro-autotransplantation (MATR) was observed. The primary effectiveness of wound closing square to MATR square was 1:20. However, as a result of wound contraction the effectiveness (on neo-epidermis square) was finally 1:15 in old rats. During histological analysis hyperplasia of the epidermis was marked and decreased gradually. Skin MATR method is considered as organ-typical culture in vivo.

[Effectiveness of the treatment of deep amd wide wounds by micro-autotransplantation of the skin].

It's experimentally show na 40-fold expansion of deep and wide skin wound covering by microtransplantation. For all this it is determined that quantitative and qualitative parameters of neoskin didn't depend on skin microfragments concentration 0.5 X 1.0 mm. Besides, histological structure of neoepidermis was analogous to epidermis in situ, despite hyperplasia. The contraction feature of neoskin conformed to contraction of open wound.

[Effect of 1-alpha oxyvitamin D3 on bone tissue in systemic osteoporosis].

Prolonged administration of oxydevit (I alpha OH D3), a synthetic analogue of the active metabolite of vitamin D (I alpha 25, (OH)2D3) in patients with systemic osteoporosis inhibited the progress of the process. Abatement of the pain syndrome and increase in physical activity were accompanied by stabilization of the roentgenologic picture and of such parameters of the bone tissue as volume density of the spongy bone and thickness of the trabeculae. The mechanism of action of the drug is not quite clear, but importance is attached to the influence of the drug on the absorption of calcium from the intestines and to persistent normalization of the level of calcium in the blood. Taking into consideration the course of such disease as systemic osteoporosis, the inhibitory impact should be regarded as achievement of therapeutic effect.

Vitamin D3 active metabolites as a countermeasure against disorders of calcium-phosphorus metabolism in hypokinetic rats.

Male Wistar rats that were experimentally hypokinetic were fed 24,25(OH)2D3 or 1,25(OH)2D3 separately or in combination to determine the effect on bone growth and on bone formation and resorption. It was shown that these parameters of bone metabolism are influenced by these metabolites of vitamin D3 by their effect on bone sensitivity to their activity and perhaps in the regulation of bone histogenesis.

[Complement and heterophil antibody levels in monkeys under conditions of antiorthostatic hypokinesia].

The experiments with sexually immature (about 3 years) primates Macaca mulatta exposed to head-down tilt and clinostatic hypokinesia showed an increase in the complementary activity of serum and a decrease of heterophil antibodies in the first case. It is inferred that an increase in the complement content reflects an adaptive reaction to head-down tilt and a decrease in heterophil antibodies indicates an inhibition of antibody-forming cells (B-lymphocytes).

Animals

[Hematopoietic organs in mice following a single administration of hydrocortisone].

Kinetics of the number and direction of CFU differentiating in mice after a single infusion of hydrocortisone in a dose of 5 mg per mouse was studied using the method of an exogenous cloning of the stem cells in the spleen and the bone marrow of the irradiated recipients. Against the background of the long-term involution of the thymicolymphoid apparatus a change of the number and direction of the CFU differentiating occurred. Along with this, CFU concentration in the spleen and bone marrow of mice remained constant. Under the hormone action CFU differentiating capacity in the spleen and bone marrow in the direction of erythro- and myelopoiesis sharply changes; CFU of the bone marrow behaves as CFU of the spleen, and CFU of the spleen behaves as CFU of the bone marrow of normal mice. It is suggested that the described effects of the CFU of the spleen and bone marrow of mice given hydrocortisone were due to the redistribution of T-lymphocytes and not to direct cytotoxic effect of hydrocortisone on the CFU population.

Animals

[Determination of "settling factor" (f) by the hematopoietic stem cell cloning test in mouse spleen and bone marrow].

The effect of distribution of injected colony-forming units (CFU) in the recipient's organism was studied. The distribution was assessed by the "sedimentation factor" (f) criterion. This factor was found by the number of colonies revealed on histological sections of the spleen and femoral bone marrow in comparison with the number of colonies visible on the surface of the spleen. Macroscopic count of the colonies on the spleen permits, to assess the f value both in the whole spleen volume and in the femoral bone marrow. In this case f value was independent of the character of CFU differentiation. f value could change depending on the physiological condition of CFU population. In irradiation of the bone marrow CFU in a dose of 200, 400 and 600 R f value in the spleen decreased in comparison with control, but remained unchanged in the bone marrow.

Discussion of the combined effect of weightlessness and ionizing radiation on the mammalian body: morphological data.

The combined effect of weightlessness and ionizing radiation, from the Cs137 source at 800 rads for 24 h, on the animal body was studied. The morphological examination of organs and tissues of rats flown aboard the biosatellite Cosmos-690, kept in the ground-based simulation experiment, and kept in the vivarium, indicated prevalence of radiation-induced changes in both experimental groups of rats. An exposure of animals to space flight factors did not produce a substantial aggravation of radiation-induced effects. This is indicated by the lack of significant differences in the weight of testes, thymus, and spleen of flight and simulation rats. However, this exposure affected adversely the development of reparative processes in the hemopoietic tissue of bone marrow. Inflight irradiation aggravated weightlessness-induced changes. A combined effect of weightlessness and irradiation did not result in the summation of the effects exerted on skeletal muscles by either factor alone.

Animals

[The radiosensitivity and direction of differentiation of splenic colony forming cells].

Radiosensitivity of spleen CFU localized in the bone marrow and in the spleen proved to be the same. Do is about 105--120r for the CFU-forming colonies in the spleen and 120--135r for the CFU-forming colonies in the bone marrow. As suggested, there are two CFU fractions in the bone marrow. One of them is radiosensitive and another--radio-resistant. Chiefly the radiosensitive CFU fraction is the one localized in the spleen.
